&#x94DC;&#x94FE;&#x9709;&#x83CC; lividans &#x4E2D;&#x7684;&#x5E94;&#x6FC0;&#x53CD;&#x5E94;&#x8D85;&#x51FA;&#x4E86;&#x94DC;&#x654F;&#x611F;&#x64CD;&#x7EB5;&#x5B50;&#x963B;&#x904F;&#x7269;&#x86CB;&#x767D; (CsoR) &#x7684;&#x76F4;&#x63A5;&#x63A7;&#x5236;&#x4E0B;&#x7684;&#x57FA;&#x56E0;&#x3002;

A copper-sensitive operon repressor protein (CsoR) has been identified in Streptomyces lividans (CsoR(Sl)) and found to regulate copper homeostasis with attomolar affinity for Cu(I). Solution studies reveal apo- and Cu(I)-CsoR(Sl) to be a tetramer assembly, and a 1.7-â„« resolution crystal structure of apo-CsoR(Sl) reveals that a significant conformational change is necessary to enable Cu(I) binding. In silico prediction of the CsoR regulon was confirmed in vitro (EMSA) and in vivo (RNA-seq), which highlighted that next to the csoR gene itself, the regulon consists of two Cu(I) efflux systems involving a CopZ-like copper metallochaperone protein and a CopA P(1)-type ATPase. Although deletion of csoR has only minor effects on S. lividans development when grown under high copper concentrations, mutations of the Cu(I) ligands decrease tolerance to copper as a result of the Cu(I)-CsoR mutants failing to disengage from the DNA targets, thus inhibiting the derepression of the regulon. RNA-seq experiments carried out on samples incubated with exogenous copper and a Î”csoR strain showed that the set of genes responding to copper stress is much wider than anticipated and largely extends beyond genes targeted by CsoR. This suggests more control levels are operating and directing other regulons in copper homeostasis beside the CsoR regulon.

R&eacute;ponse au stress chez Streptomyces lividans de cuivre s&#39;&eacute;tend au-del&agrave; des g&egrave;nes sous le contr&ocirc;le direct d&#39;une prot&eacute;ine de r&eacute;presseur de l&#39;op&eacute;ron sensibles au cuivre (ROSC).

Ein Kupfer-Sensitive Operon Hemmer Protein (CsoR) in Streptomyces Lividans festgestellt wurde (CsoR(Sl)) und gefunden, Kupfer Hom&ouml;ostase mit Attomolar Affinit&auml;t zu Cu(I) zu regulieren. L&ouml;sung Studien zu offenbaren, Apo- und Cu(I)-CsoR(Sl) zu einem Tetramer-Versammlung und eine 1.7-&Aring; Aufl&ouml;sung Kristallstruktur von apo-CsoR(Sl) zeigt, dass eine wesentliche &Auml;nderung der Konformations&auml;nderungen Cu(I) Bindung aktivieren muss. In Silico Vorhersage der CsoR Regulon in Vitro (EMSA) best&auml;tigt wurde und in-vivo (RNA-Seq), hervorgehoben, die neben den CsoR das Gen selbst das Regulon besteht aus zwei Cu(I)-Efflux-Systeme mit einem CopZ-wie Kupfer Metallochaperone-Protein und eine CopA P-1-Typ-ATPase. Obwohl Streichung von CsoR nur geringf&uuml;gige Auswirkungen auf S. Lividans Entwicklung beim unter hohen Kupfer Konzentrationen angebaut hat, verringern Mutationen der Cu(I) Liganden Toleranz infolge der Cu (I) Kupfer-CsoR Mutanten vers&auml;umt, von der DNA-Ziele, l&ouml;sen somit Hemmung der Derepression das Regulon. RNA-Seq Experimente an Proben inkubiert mit exogenen Kupfer und ein &Delta;csoR-Stamm zeigte, dass die Gene, die reagieren auf Kupfer Stress viel gr&ouml;&szlig;er ist als erwartet und weitgehend Gene gezielt von CsoR hinausgeht. Dies deutet darauf hin, mehr Kontrolle-Ebenen sind Betriebs- und andere Regulons in Kupfer Hom&ouml;ostase neben der CsoR Regulon Regie.

Als Reaktion auf Stress bei Streptomyces Lividans Kupfer hinausgeht Gene unter die direkte Kontrolle &uuml;ber ein Kupfer-Sensitive Operon Hemmer Protein (CsoR).

Una proteina repressore operone rame-sensibili (CsoR) &egrave; stata identificata in Streptomyces lividans (CsoR(Sl)) e trovato per regolare l&#39;omeostasi del rame con affinit&agrave; attomolar per Cu(I). Soluzione studi rivelano apo - e Cu(I)-CsoR(Sl) per essere un tetramero assembly, e una struttura di cristallo di 1,7-&Aring; risoluzione di apo-CsoR(Sl) rivela che un significativo cambiamento conformazionale &egrave; necessario per attivare l&#39;associazione Cu(I). Anche se l&#39;eliminazione del csoR ha solo lievi effetti sullo sviluppo di lividans S. Quando coltivate in alte concentrazioni di rame, mutazioni di ligandi della Cu(I) diminuiscono tolleranza di rame a seguito del Cu (I)-CsoR mutanti non riuscendo a sganciarsi dagli obiettivi del DNA, inibendo quindi la derepression del Regulone. RNA-seq esperimenti condotti su campioni incubati con rame esogeno e un ceppo di &Delta;csoR ha dimostrato che l&#39;insieme dei geni che risponde alle sollecitazioni di rame &egrave; molto pi&ugrave; ampia del previsto e in gran parte si estende di l&agrave; di geni bersagliati di CsoR. Questo suggerisce ulteriori livelli di controllo sono operativi e dirigere altri regulons nell&#39;omeostasi del rame al lato del Regulone CsoR.

Risposta a stress in Streptomyces lividans di rame si estende oltre geni sotto il controllo diretto di una proteina repressore operone rame-sensibili (CsoR).

Una prote&iacute;na del represor del oper&oacute;n de cobre-sensible (CsoR) ha sido identificada en Streptomyces lividans (CsoR(Sl)) y regular la homeostasis del cobre con afinidad de attomolar para Cu(I). Soluci&oacute;n estudios revelan apo - y Cu(I)-CsoR(Sl) a ser una Asamblea de tetr&aacute;mero, y una estructura de cristal de 1.7-&Aring; de resoluci&oacute;n de apo-CsoR(Sl) revela que un cambio conformacional significativo es necesario activa a Cu(I) enlace. En la predicci&oacute;n de silico de la CsoR regul&oacute;n fue confirmada en vitro (EMSA) y in vivo (RNA-seq), que destac&oacute; que al lado del propio gen csoR, el regul&oacute;n consta de dos sistemas de emanaci&oacute;n de Cu(I) que involucra una prote&iacute;na similar a CopZ cobre metallochaperone y una CopA P 1-tipo ATPasa. Aunque la eliminaci&oacute;n de csoR tiene s&oacute;lo efectos menores en desarrollo lividans cuando se cultiva bajo altas concentraciones de cobre, las mutaciones de los ligandos Cu(I) disminuyen la tolerancia al cobre como resultado de la Cu (I)-CsoR mutantes incapaces de desenganche de los objetivos de la DNA, lo que inhibe la derepression del regul&oacute;n. RNA-seq experimentos realizados en las muestras que se incubaron con cobre ex&oacute;gena y una cepa de &Delta;csoR demostraron que el conjunto de genes de respuesta al estr&eacute;s de cobre es mucho mayor de lo esperado y en gran parte se extiende m&aacute;s all&aacute; de los genes de CsoR. Esto sugiere m&aacute;s niveles de control est&aacute;n funcionando y dirigir otros regulons en la homeostasis del cobre al lado del regul&oacute;n CsoR.

Respuesta al estr&eacute;s en Streptomyces lividans de cobre se extiende m&aacute;s all&aacute; de genes bajo el control directo de una prote&iacute;na del represor del oper&oacute;n de cobre-sensible (CsoR).
