Hydrog&eacute;nosomes et mitosomes repr&eacute;sentent des adaptations remarquables mitochondriales dans les protistes parasites ana&eacute;robies tels que Trichomonas vaginalis et Giardia intestinalis, respectivement. Afin de fournir un outil pour &eacute;tudier ces organites dans les cellules vivantes, le HaloTag a &eacute;t&eacute; fusionn&eacute;e &agrave; g. intestinalis IscU et T. vaginalis frataxine et exprim&eacute;e dans les mitosomes et les hydrog&eacute;nosomes, respectivement. L&#39;incubation des parasites avec le ligand-Halo fluorescent a entra&icirc;n&eacute; dans l&#39;&eacute;tiquetage des organites hautement sp&eacute;cifique, permettant l&#39;imagerie direct des organites. Hydrogenosomes e mitosomes rappresentano notevoli adattamenti mitocondriali nei protisti anaerobici parassitarie come Trichomonas vaginalis e Giardia intestinalis, rispettivamente. Al fine di fornire uno strumento per lo studio di questi organelli nelle cellule dal vivo, il HaloTag fu fuso a g. intestinalis IscU e T. vaginalis fratassina ed espressa in mitosomes e hydrogenosomes, rispettivamente. L&#39;incubazione dei parassiti con l&#39;Halo-ligando fluorescente provocato etichettatura organellare altamente specifico, permettendo di imaging dal vivo gli organelli. Con la matrice di ligandi disponibile la tecnologia HaloTag offre un nuovo strumento per studiare la dinamica dei compartimenti correlati a mitocondri, cos&igrave; come altri componenti cellulari in questi intriganti eucarioti unicellulari.

Hydrogenosomes and mitosomes represent remarkable mitochondrial adaptations in the anaerobic parasitic protists such as Trichomonas vaginalis and Giardia intestinalis, respectively. In order to provide a tool to study these organelles in the live cells, the HaloTag was fused to G. intestinalis IscU and T. vaginalis frataxin and expressed in the mitosomes and hydrogenosomes, respectively. The incubation of the parasites with the fluorescent Halo-ligand resulted in highly specific organellar labeling, allowing live imaging of the organelles. With the array of available ligands the HaloTag technology offers a new tool to study the dynamics of mitochondria-related compartments as well as other cellular components in these intriguing unicellular eukaryotes.
