Associative plasticity is a basic essential attribute of nervous systems. As shown by numerous reports, Drosophila is able to establish simple forms of appetitive and aversive olfactory associations at both larval and adult stages. Whereas most adult studies on aversive learning employed electric shock as a negative reinforcer, larval paradigms essentially utilized gustatory stimuli to create negative associations, a discrepancy that limits the comparison of data. To overcome this drawback, we critically revisited larval odor-electric shock conditioning. First, we show that lithium chloride (LiCl), which was used in all previous larval electric shock paradigms, is not required per se in larval odor-electric shock learning. This is of considerable practical advantage because beside its peculiar effects LiCl is attractive to larvae at low concentration that renders comparative learning studies on genetically manipulated larvae complicated. Second, we confirm that in both a 2-odor reciprocal and a 1-odor nonreciprocal conditioning regimen, larvae are able to associate an odor with electric shock. In the latter experiments, initial learning scores reach an asymptote after 5 training trials, and aversive memory is still detectable after 60 min. Our experiments provide a comprehensive basis for future comparisons of larval olfactory conditioning reinforced by different modalities, for studies aimed at analyzing odor-electric shock learning in the larva and the adult, and for investigations of the cellular and molecular substrate of aversive olfactory learning in the simple Drosophila model.

Comme l&#39;attestent de nombreux rapports, Drosophila est en mesure d&#39;&eacute;tablir des formes simples d&#39;app&eacute;tence et d&#39;associations olfactives aversifs aux stades larvaires et adultes. Consid&eacute;rant que plus adultes &eacute;tudes sur l&#39;apprentissage aversif employ&eacute; &eacute;lectrocution comme un agent de renfor&ccedil;ateur n&eacute;gatif, larves paradigmes utilis&eacute;s essentiellement des stimuli gustatifs pour cr&eacute;er des associations n&eacute;gatives, un &eacute;cart qui limite la comparaison des donn&eacute;es. Pour pallier cet inconv&eacute;nient, nous avons revisit&eacute; critique conditionn&eacute; odeur-&eacute;lectrocution larvaire. Tout d&#39;abord, nous montrons que le chlorure de lithium (LiCl), qui a &eacute;t&eacute; utilis&eacute; dans tous les paradigmes de larves &eacute;lectrocution pr&eacute;c&eacute;dente, n&#39;est pas n&eacute;cessaire de soi dans apprentissage larvaire odeur-&eacute;lectrocution. C&#39;est d&#39;avantage pratique consid&eacute;rable, car &agrave; c&ocirc;t&eacute; de ses effets particuliers que LiCl est attrayant pour les larves &agrave; faible concentration qui rend comparative studies d&#39;apprentissage sur g&eacute;n&eacute;tiquement manipul&eacute;es larves compliqu&eacute;s. Deuxi&egrave;mement, nous confirmons que dans 2-odeur r&eacute;ciproque et un conditionnement 1-odeur non r&eacute;ciproque, les larves sont capables d&#39;associer une odeur &agrave; un choc &eacute;lectrique. Dans les exp&eacute;riences de ce dernier, apprentissage initial scores atteignent une asymptote apr&egrave;s 5 essais de formation, et m&eacute;moire aversif est encore d&eacute;tectable apr&egrave;s 60 min. Nos exp&eacute;riences constituent une base compl&egrave;te pour de futures comparaisons de conditionnement olfactif larvaire, renforc&eacute;e par des modalit&eacute;s diff&eacute;rentes, pour des &eacute;tudes visant &agrave; analyser les odeurs-&eacute;lectrocution d&#39;apprentissage dans la larve et l&#39;adulte et enqu&ecirc;te du substrat cellulaire et mol&eacute;culaire d&#39;aversif apprentissage olfactif dans le mod&egrave;le simple de la drosophile.

Assoziative Plastizit&auml;t ist eine grundlegende wesentliche Eigenschaft von Nervensystemen. Wie zahlreiche Berichte zeigen, ist Drosophila einfache Formen Vorabend und unangenehmen olfaktorischen Assoziationen in sowohl Erwachsene als auch larvalen Stadien zu etablieren. W&auml;hrend die meisten Erwachsene Studien auf aversive lernen Stromschlag als eine negative Verst&auml;rker eingesetzt, genutzte larvalen Paradigmen im wesentlichen gustatorisches Reize negative Assoziationen, eine Diskrepanz zu erstellen, die den Vergleich von Daten beschr&auml;nkt. Um diesen Nachteil zu &uuml;berwinden, revisited wir kritisch larvalen Geruch-Elektroschock-Klimaanlage. Zuerst zeigen wir, dass Lithiumchlorid (LiCl), die in alle vorherigen larvalen Elektroschock-Paradigmen verwendet wurde, nicht per se in larvalen Geruch-Stromschlag Lernen erforderlich ist. In den letzteren Experimenten anf&auml;ngliche lernen Resultate erreichen eine Asymptote nach 5 Training versuchen und aversive Speicher ist nach 60 min noch nachweisbar. Unsere Experimente bieten eine umfassende Grundlage f&uuml;r zuk&uuml;nftige Vergleiche larvalen olfaktorischen Konditionierung durch verschiedene Modalit&auml;ten f&uuml;r Studien zur Analyse von Geruch-Stromschlag in der Larve und der Erwachsene lernen und f&uuml;r Untersuchungen der zellul&auml;ren und molekularen Substrat aversive olfaktorischen lernen im einfachen Drosophila Modell verst&auml;rkt.

Come dimostrato da numerosi rapporti, drosofila &egrave; in grado di stabilire forme semplici di appetitiva e associazioni olfattive aversive a stadi larvali e di adulti. Considerando che pi&ugrave; adulti studi sull&#39;apprendimento aversive impiegato scosse elettriche come un reinforcer negativo, larvale paradigmi utilizzati essenzialmente stimoli gustativi per creare associazioni negative, una discrepanza che limita il confronto dei dati. Per ovviare a questo inconveniente, abbiamo rivisitato criticamente condizionata larvale odore-folgorazioni. In primo luogo, vi mostriamo che cloruro di litio (LiCl), che &egrave; stato utilizzato in tutti i precedenti paradigmi larvale scossa elettrica, non &egrave; necessaria per s&eacute; nell&#39;apprendimento larvale odore-folgorazioni. Questo &egrave; di notevole vantaggio pratico perch&eacute; accanto a suoi effetti peculiari che LICL &egrave; attraente per le larve a bassa concentrazione che esegue il rendering comparativo apprendimento studi su geneticamente manipolato le larve complicate. In secondo luogo, si conferma che, in un reciproco 2-odore sia un regime di condizionamento nonreciprocal 1-odore, le larve sono in grado di associare un odore a scosse elettriche. Plasticidad asociativa es un atributo esencial b&aacute;sico del sistema nervioso. Como se muestra por numerosos informes, Drosophila es capaces de establecer formas simples de asociaciones olfativas aversivas y apetitivo en etapas tanto larvas como adultos. Mientras que los adultos m&aacute;s estudios sobre aprendizaje aversivo emplean descarga el&eacute;ctrica como un reforzador negativo, larvas paradigmas utilizan esencialmente gustativos est&iacute;mulos para crear asociaciones negativas, una discrepancia que limita la comparaci&oacute;n de datos. Para superar este inconveniente, nos revisar cr&iacute;ticamente acondicionado larval olor-electrocuci&oacute;n. En primer lugar, mostramos que cloruro de litio (LiCl), que fue utilizado en todos anteriores paradigmas de larvas de descarga el&eacute;ctrica, no es necesario per se en el aprendizaje de larvas olor-electrocuci&oacute;n. Esto es de considerable ventaja pr&aacute;ctica porque al lado de sus efectos peculiares que LiCl es atractivo a las larvas en baja concentraci&oacute;n que hace comparativo aprendizaje estudios en gen&eacute;ticamente manipulados larvas complicadas. En segundo lugar, confirmamos que tanto un rec&iacute;proco 2-olor en un r&eacute;gimen de condicionamiento t&eacute;rminos 1-olor, las larvas son capaces de asociar un olor con descargas el&eacute;ctricas. En los experimentos de este &uacute;ltimos, resultados de aprendizaje inicial a una as&iacute;ntota despu&eacute;s 5 ensayos de entrenamiento, y memoria aversivo es todav&iacute;a perceptible despu&eacute;s de 60 minutos. Nuestros experimentos proporcionan una base integral para futuras comparaciones de larva condicionamiento olfativo reforzado por diferentes modalidades, para estudios destinados a analizar olor-electrocuci&oacute;n aprendiendo en la larva y el adulto y para las investigaciones del sustrato celular y molecular de aprendizaje aversivo olfativo en el modelo simple de Drosophila.

Gustatory stimuli allow an organism not only to orient in its environment toward energy-rich food sources to maintain nutrition but also to avoid unpleasant or even poisonous substrates. For both mammals and insects, sugars-perceived as "sweet"-potentially predict nutritional benefit. Interestingly, even Drosophila adult flies are attracted to most high-potency sweeteners preferred by humans. However, the gustatory information of a sugar may be misleading as some sugars, although perceived as "sweet," cannot be metabolized. Accordingly, in adult Drosophila, a postingestive system that additionally evaluates the nutritional benefit of an ingested sugar has been shown to exist. By using a set of seven different sugars, which either offer (fructose, sucrose, glucose, maltodextrin, and sorbitol) or lack (xylose and arabinose) nutritional benefit, we show that Drosophila, at the larval stage, can perceive and evaluate sugars based on both nutrition-dependent and -independent qualities. In detail, we find that larval survival and feeding mainly depend on the nutritional value of a particular sugar. In contrast, larval choice behavior and learning are regulated in a more complex way by nutrition value-dependent and nutrition value-independent information. The simplicity of the larval neuronal circuits and their accessibility to genetic manipulation may ultimately allow one to identify the neuronal and molecular basis of the larval sugar perception systems described here behaviorally.

The Drosophila larva has turned into a particularly simple model system for studying the neuronal basis of innate behaviors and higher brain functions. Neuronal networks involved in olfaction, gustation, vision and learning and memory have been described during the last decade, often up to the single-cell level. Thus, most of these sensory networks are substantially defined, from the sensory level up to third-order neurons. This is especially true for the olfactory system of the larva. Given the wealth of genetic tools in Drosophila it is now possible to address the question how modulatory systems interfere with sensory systems and affect learning and memory. Here we focus on the serotonergic system that was shown to be involved in mammalian and insect sensory perception as well as learning and memory. Larval studies suggested that the serotonergic system is involved in the modulation of olfaction, feeding, vision and heart rate regulation. In a dual anatomical and behavioral approach we describe the basic anatomy of the larval serotonergic system, down to the single-cell level. In parallel, by expressing apoptosis-inducing genes during embryonic and larval development, we ablate most of the serotonergic neurons within the larval central nervous system. When testing these animals for naÃ¯ve odor, sugar, salt and light perception, no profound phenotype was detectable; even appetitive and aversive learning was normal. Our results provide the first comprehensive description of the neuronal network of the larval serotonergic system. Moreover, they suggest that serotonin per se is not necessary for any of the behaviors tested. However, our data do not exclude that this system may modulate or fine-tune a wide set of behaviors, similar to its reported function in other insect species or in mammals. Based on our observations and the availability of a wide variety of genetic tools, this issue can now be addressed.

Die Drosophila Larve ist ein besonders einfaches Modellsystem zur Erforschung der neuronalen Grundlagen der angeborenen Verhaltensweisen und h&ouml;heren Hirnfunktionen geworden. La larva de Drosophila se ha convertido en un sistema modelo simple para estudiar la base neuronal de comportamientos naturales y funciones superiores del cerebro. Redes neuronales implicados en el olfato, percepci&oacute;n, visi&oacute;n y aprendizaje y la memoria se han descrito durante la &uacute;ltima d&eacute;cada, a menudo hasta el nivel unicelular. As&iacute;, la mayor&iacute;a de estas redes sensoriales se define sustancialmente, desde el nivel sensorial hasta las neuronas de tercer orden. Esto es especialmente cierto para el sistema olfativo de la larva. Dada la riqueza de herramientas gen&eacute;ticas en Drosophila ahora es posible para que aborden el asunto sistemas moduladores interfieran con sistemas sensoriales y afectan el aprendizaje y la memoria. Aqu&iacute; nos centramos en el sistema serotonin&eacute;rgico que mostr&oacute; ser involucrados en la memoria, as&iacute; como el aprendizaje y la percepci&oacute;n sensorial de mam&iacute;fera e insecto. Larvarios estudios sugirieron que el sistema serotonin&eacute;rgico est&aacute; implicado en la modulaci&oacute;n de olfato, alimentaci&oacute;n, regulaci&oacute;n de la visi&oacute;n y la frecuencia card&iacute;aca. En un doble enfoque anat&oacute;mico y de comportamiento se describe la anatom&iacute;a b&aacute;sica del sistema serotonin&eacute;rgico larval, hasta el nivel unicelular. En paralelo, expresando los genes que inducen apoptosis durante el desarrollo embrionario y larval, nos ablar de la mayor&iacute;a de las neuronas serotonin&eacute;rgicas dentro de la larva del sistema nervioso central. Al probar estos animales para olor de ingenuos, az&uacute;car, sal y luz de la percepci&oacute;n, no profunda fenotipo era perceptible; incluso apetitivo y aprendizaje aversivo era normal. Nuestros resultados proporcionan la primera descripci&oacute;n completa de la red neuronal del sistema serotonin&eacute;rgico larval. Adem&aacute;s, sugieren que la serotonina per no se es necesaria para cualquiera de las conductas probadas. Sin embargo, nuestros datos no excluyen que este sistema puede modular o ajustar un amplio conjunto de comportamientos, similares a su funci&oacute;n reportado en otras especies de insectos o mam&iacute;feros. Basado en nuestras observaciones y la disponibilidad de una amplia variedad de herramientas gen&eacute;ticas, este problema puede ahora ser abordado.
