Traditional biochemistry of contact activation of blood coagulation suggesting that anionic hydrophilic surfaces are specific activators of the cascade is inconsistent with known trends in protein adsorption. To investigate contact activation reactions, a chromogenic assay was used to measure prekallkrein (PK) hydrolysis to kallikrein (Kal) by activated factor XII (FXIIa) at test hydrophilic (clean glass) and hydrophobic (silanized glass) surfaces in the presence of bovine serum albumin (BSA). Hydrolysis of PK by FXIIa is detected after contact of the zymogen FXII with a test hydrophobic surface only if putatively-adsorbed FXIIa is competitively displaced by BSA. By contrast, FXIIa activity is detected spontaneously following FXII activation by a hydrophilic surface and requires no adsorption displacement. These results (i) show that an anionic hydrophilic surface is not a necessary cofactor for FXIIa-mediated hydrolysis of PK, (ii) indicate that PK hydrolysis does not need to occur by an activation complex assembled directly on an anionic, activating surface, (iii) confirms that contact activation of FXII (autoactivation) is not specific to anionic hydrophilic surfaces, and (iv) demonstrates that protein-adsorption competition is an essential feature that must be included in any comprehensive mechanism of surface-induced blood coagulation.

Sounds are filtered in a spatial- and frequency-dependent manner by the head and pinna giving rise to the acoustical cues to sound source location. These spectral and temporal transformations are dependent on the physical dimensions of the head and pinna. Therefore, the magnitudes of binaural sound location cues-the interaural time (ITD) and level (ILD) differences-are hypothesized to systematically increase while the lower frequency limit of substantial ILD production is expected to decrease due to the increase in head and pinna size during development. The frequency ranges of the monaural spectral notch cues to source elevation are also expected to decrease. This hypothesis was tested here by measuring directional transfer functions (DTFs), the directional components of head-related transfer functions, and the linear dimensions of the head and pinnae for chinchillas from birth through adulthood. Dimensions of the head and pinna increased by factors of 1.8 and 2.42, respectively, reaching adult values by ~6Â weeks. From the DTFs, the ITDs, ILDs, and spectral shape cues were computed. Maximum ITDs increased by a factor of 1.75, from ~160Â Î¼s at birth (P0-1, first postnatal day) to 280Â Î¼s in adults. ILDs depended on source location and frequency exhibiting a shift in the frequency range of substantial ILD (>10Â dB) from higher to lower frequencies with increasing head and pinnae size. Similar trends were observed for the spectral notch frequencies which ranged from 14.7-33.4Â kHz at P0-1 to 5.3-19.1Â kHz in adults. The development of the spectral notch cues, the spatial- and frequency-dependent distributions of DTF amplitude gain, acoustic directionality, maximum gain, and the acoustic axis were systematically related to the dimensions of the head and pinnae. The dimension of the head and pinnae in the chinchilla as well as the acoustical properties associated with them are mature by ~6Â weeks.

There are three main cues to sound location: the interaural differences in time (ITD) and level (ILD) as well as the monaural spectral shape cues. These cues are generated by the spatial- and frequency-dependent filtering of propagating sound waves by the head and external ears. Although the chinchilla has been used for decades to study the anatomy, physiology, and psychophysics of audition, including binaural and spatial hearing, little is actually known about the sound pressure transformations by the head and pinnae and the resulting sound localization cues available to them. Here, we measured the directional transfer functions (DTFs), the directional components of the head-related transfer functions, for 9 adult chinchillas. The resulting localization cues were computed from the DTFs. In the frontal hemisphere, spectral notch cues were present for frequencies from âˆ¼6-18Â kHz. In general, the frequency corresponding to the notch increased with increases in source elevation as well as in azimuth towards the ipsilateral ear. The ILDs demonstrated a strong correlation with source azimuth and frequency. The maximum ILDs were <10Â dB for frequencies <5Â kHz, and ranged from 10-30Â dB for the frequencies >5Â kHz. The maximum ITDs were dependent on frequency, yielding 236Â Î¼s at 4Â kHz and 336Â Î¼s at 250Â Hz. Removal of the pinnae eliminated the spectral notch cues, reduced the acoustic gain and the ILDs, altered the acoustic axis, and reduced the ITDs.

Conductive hearing loss (CHL) is known to produce hearing deficits, including deficits in sound localization ability. The differences in sound intensities and timing experienced between the two tympanic membranes are important cues to sound localization (ILD and ITD, respectively). Although much is known about the effect of CHL on hearing levels, little investigation has been conducted into the actual impact of CHL on sound location cues. This study investigated effects of CHL induced by earplugs on cochlear microphonic (CM) amplitude and timing and their corresponding effect on the ILD and ITD location cues. Acoustic and CM measurements were made in 5 chinchillas before and after earplug insertion, and again after earplug removal using pure tones (500Â Hz to 24Â kHz). ILDs in the unoccluded condition demonstrated position and frequency dependence where peak far-lateral ILDs approached 30Â dB for high frequencies. Unoccluded ear ITD cues demonstrated positional and frequency dependence with increased ITD cue for both decreasing frequency (Â±420Â Î¼s at 500Â Hz, Â±310Â Î¼s for 1-4Â kHz) and increasingly lateral sound source locations. Occlusion of the ear canal with foam plugs resulted in a mild, frequency-dependent conductive hearing loss of 10-38Â dB (mean 31Â Â±Â 3.9Â dB) leading to a concomitant frequency dependent increase in ILDs at all source locations. The effective ITDs increased in a frequency dependent manner with ear occlusion as a direct result of the acoustic properties of the plugging material, the latter confirmed via acoustical measurements using a model ear canal with varying volumes of acoustic foam. Upon ear plugging with acoustic foam, a mild CHL is induced. Furthermore, the CHL induced by acoustic foam results in substantial changes in the magnitudes of both the ITD and ILD cues to sound location.

En outre, la LCH induite par des r&eacute;sultats de mousse acoustique dans des changements substantiels dans les amplitudes des rep&egrave;res de l&#39;ITD et ILD &agrave; son emplacement.

Nach Ohr mit Akustikschaum einstecken wird eine milde CHL induziert. Le differenze di intensit&agrave; di suono e temporizzazione vissuto tra le due membrane tympanic sono importanti spunti per localizzazione audio (ILD e ITD, rispettivamente). Anche se molto &egrave; conosciuto circa l&#39;effetto di CHL su livelli di ascolto, piccola indagine &egrave; stata condotta l&#39;effettivo impatto della CHL su stecche di posizione il suono. Questo studio investigato gli effetti di CHL indotta da tappi per le orecchie su coclea microfonico ampiezza (CM) e la tempistica ed il loro effetto corrispondente sul ILD e ITD spunti di posizione. Acustica e misure CM sono state fatte in 5 cincill&agrave; prima e dopo l&#39;inserimento del tappo e nuovamente dopo la rimozione del tappo auricolare utilizzando toni puri (500 Hz a 24 kHz). Spunti ITD unoccluded orecchio dimostrato posizionale e dipendenza di frequenza con aumento ITD cue per entrambi frequenza decrescente (&plusmn;420 &mu;s a 500 Hz, &plusmn;310 &mu;s per 1-4 kHz) e sempre pi&ugrave; laterale localit&agrave; sorgente sonora. Occlusione del condotto uditivo con schiuma spine ha provocate un lieve, frequenza dipendente conduttivo di 10-38 dB (Media 31 dB &plusmn; 3,9) portando ad un aumento dipendente dalla frequenza concomitante ILDs la perdita dell&#39;udito a tutti i luoghi di origine. L&#39;efficace ITDs aumentato in maniera dipendente dalla frequenza con occlusione dell&#39;orecchio come un risultato diretto delle propriet&agrave; acustiche del collegamento materiale, quest&#39;ultimo ha confermato tramite misurazioni acustiche, utilizzando un modello condotto uditivo con diversi volumi di schiuma acustica. All&#39;orecchio collegando con schiuma acustica, un lieve CHL &egrave; indotta. Inoltre, la CHL indotta da risultati schiuma acustica in cambiamenti sostanziali nelle magnitudini di ITD sia ILD spunti di posizione il suono.

Gli effetti della sordit&agrave; indotta sperimentalmente sugli aspetti temporali e spettrali di trasmissione del suono attraverso l&#39;orecchio.

Hipoacusia conductiva (CHL) es conocido por producir d&eacute;ficits de audici&oacute;n, incluyendo d&eacute;ficits en la capacidad de localizaci&oacute;n del sonido. Las diferencias en la intensidad de sonido y tiempo vivido entre las dos membranas timp&aacute;nicas son se&ntilde;ales importantes para la localizaci&oacute;n del sonido (ILD y ITD, respectivamente). Aunque se sabe mucho sobre el efecto de CHL en los niveles de audici&oacute;n, poca investigaci&oacute;n se ha realizado sobre el impacto real de CHL en se&ntilde;ales de localizaci&oacute;n del sonido. Este estudio investig&oacute; efectos de CHL hab&iacute;an inducida por tapones para los o&iacute;dos sobre amplitud de (CM) microf&oacute;nico coclear y el calendario y sus correspondientes efectos en el ILD y ITD se&ntilde;ales de ubicaci&oacute;n. Ac&uacute;stica y CM medidas fueron hechas en 5 chinchillas antes y despu&eacute;s de la inserci&oacute;n de tap&oacute;n para los o&iacute;dos y despu&eacute;s de la eliminaci&oacute;n del tap&oacute;n usando tonos puros (a 500 Hz a 24 kHz). ILDs en la condici&oacute;n unoccluded posici&oacute;n demostrada y la dependencia de la frecuencia donde pico ILDs lejos-lateral se acerc&oacute; a 30 dB para las frecuencias altas. Se&ntilde;ales ITD unoccluded o&iacute;do demostraron posicional y dependencia de la frecuencia con mayor ITD cue para ambos frecuencia decreciente (&mu;s de &plusmn;420 a 500 Hz, &plusmn;310 &mu;s para 1-4 kHz) y lateral cada vez m&aacute;s lugares de la fuente de sonido. Obstrucci&oacute;n del conducto auditivo con tapones de espuma result&oacute; en una p&eacute;rdida auditiva conductiva leve, dependiente de la frecuencia de 10-38 dB (media &plusmn; 31 dB 3.9) conduce a un aumento concomitante de frecuencia dependiente ILDs en todos los lugares de origen. La efectiva ITDs aumentaron de una manera dependiente de frecuencia con la obstrucci&oacute;n del o&iacute;do como consecuencia directa de las propiedades ac&uacute;sticas del material taponamiento, este &uacute;ltimo confirm&oacute; mediante mediciones ac&uacute;sticas utilizando un modelo del conducto auditivo con diferentes vol&uacute;menes de espuma aislante. Al o&iacute;do tapando con espuma ac&uacute;stica, se induce una leve CHL. Adem&aacute;s, la CHL inducida por resultados de espuma ac&uacute;stica en cambios sustanciales en las magnitudes de ITD tanto ILD se&ntilde;ales para la localizaci&oacute;n del sonido.

Otitis media with effusion (OME) is a pathologic condition of the middle ear that leads to a mild to moderate conductive hearing loss as a result of fluid in the middle ear. Recurring OME in children during the first few years of life has been shown to be associated with poor detection and recognition of sounds in noisy environments, hypothesized to result due to altered sound localization cues. To explore this hypothesis, we simulated a middle ear effusion by filling the middle ear space of chinchillas with different viscosities and volumes of silicone oil to simulate varying degrees of OME. While the effects of middle ear effusions on the interaural level difference (ILD) cue to location are known, little is known about whether and how middle ear effusions affect interaural time differences (ITDs). Cochlear microphonic amplitudes and phases were measured in response to sounds delivered from several locations in azimuth before and after filling the middle ear with fluid. Significant attenuations (20-40Â dB) of sound were observed when the middle ear was filled with at least 1.0Â ml of fluid with a viscosity of 3.5 Poise (P) or greater. As expected, ILDs were altered by ~30Â dB. Additionally, ITDs were shifted by ~600Â Î¼s for low frequency stimuli (<4Â kHz) due to a delay in the transmission of sound to the inner ear. The data show that in an experimental model of OME, ILDs and ITDs are shifted in the spatial direction of the ear without the experimental effusion.

Comme pr&eacute;vu, ILDs ont &eacute;t&eacute; alt&eacute;r&eacute;s par ~ 30 dB. Wiederkehrende OME bei Kindern in den ersten Jahren des Lebens ist gezeigt worden, um schlechte Erkennung zugeordnet werden und Anerkennung der T&ouml;ne in einer lauten Umgebung, um wegen f&uuml;hren die Hypothese ge&auml;ndert Ton Lokalisierung Hinweise. Um diese Hypothese zu erkunden, simuliert wir eine Mittelohr-Erguss durch Ausf&uuml;llen des Raumes des mittleren Ohrs von Chinchillas mit verschiedenen Viskosit&auml;ten und Mengen von Silikon&ouml;l unterschiedlichem OME simuliert. Per esplorare questa ipotesi, abbiamo simulato un effusione orecchio medio riempiendo lo spazio dell&#39;orecchio medio di cincill&agrave; con diverse viscosit&agrave; e volumi di olio di silicone per simulare vari gradi di OME. Mentre sono noti gli effetti di effusioni dell&#39;orecchio medio su cue interaural differenza di livello (ILD) a posizione, poco si sa se e come effusioni dell&#39;orecchio medio possono influenzare differenze di tempo interaural (ITDs). Fasi e ampiezze microfonica cocleare sono stati misurati in risposta a suoni forniti da diverse posizioni in azimuth prima e dopo il riempimento dell&#39;orecchio medio con fluido. Significative attenuazioni (20-40 dB) del suono sono stati osservati quando l&#39;orecchio medio &egrave; stato riempito di almeno 1,0 ml di fluido con una viscosit&agrave; di 3,5 Poise (P) o maggiore. Otitis media con efusi&oacute;n (OME) es una condici&oacute;n patol&oacute;gica del o&iacute;do medio que conduce a una leve a moderada hipoacusia conductiva como resultado de l&iacute;quido en el o&iacute;do medio. OME recurrente en ni&ntilde;os durante los primeros a&ntilde;os de vida ha demostrado estar asociados con pobre detecci&oacute;n y reconocimiento de sonidos en ambientes ruidosos, la hip&oacute;tesis de resultar debido a alterarse se&ntilde;ales de localizaci&oacute;n del sonido. Para explorar esta hip&oacute;tesis, simul&oacute; una efusi&oacute;n del o&iacute;do medio llenando el espacio del o&iacute;do medio de chinchillas con diferentes viscosidades y vol&uacute;menes de aceite de silicona para simular diversos grados de OME. Aunque se conocen los efectos de efusiones del o&iacute;do medio en la se&ntilde;al de la diferencia interaural de nivel (ILD) a ubicaci&oacute;n, poco se conoce acerca de si y c&oacute;mo las efusiones del o&iacute;do medio afectan las diferencias de tiempo interaural (ITDs). Fases y amplitudes microf&oacute;nicos cocleares se midieron en respuesta a sonidos entregado desde varias ubicaciones en Azimut antes y despu&eacute;s de llenar el o&iacute;do medio con el l&iacute;quido. Se observaron importantes atenuaciones (20-40 dB) de sonido cuando el o&iacute;do medio se llen&oacute; de menos de 1,0 ml de l&iacute;quido con una viscosidad de 3,5 Poise (P) o mayor. Como era de esperar, ILDs fueron alterados por ~ 30 dB. Adem&aacute;s, cambiaron de ITDs por ~ 600 &mu;s para est&iacute;mulos de baja frecuencia (&lt; 4KHz) debido a un retraso en la transmisi&oacute;n del sonido al o&iacute;do interno. Los datos muestran que en un modelo experimental de la OMD, ILDs y ITDs se cambian de puesto en la direcci&oacute;n espacial del o&iacute;do sin la efusi&oacute;n experimental.

El conductor p&eacute;rdida debido a un derrame del o&iacute;do de medio inducida experimentalmente altera el nivel interaural de la audiencia y tiempo se&ntilde;ales de diferencia a la localizaci&oacute;n del sonido.
