HIV infection may be modified by CD8(+) T cells by the production of nonlytic antiviral factors. To determine subpopulations that mediate nonlytic, antiviral activity, we examined the production of beta chemokines and of CD8 antiviral factor (CAF) by different subsets, using CD8(+) cells derived from 24 HIV-1-infected and 25 uninfected individuals. Subjects with CD8(+) cell counts greater than 200/microl produced increased levels of MIP-1alpha by CD8(+)CD28(+), CD8(+)CD38(-), and CD8(+)HLA-DR(+) subsets as compared with uninfected controls. CD8(+)CD38(-) cells produced higher levels of MIP-1beta and RANTES. CAF production was increased by CD8(+)CD38(+) and CD8(+)HLA-DR(+) cells of HIV-infected individuals as compared with uninfected controls. Chemokine production was increased by cells that do not express activation markers, whereas CAF activity was increased by cells expressing CD38 or HLA-DR. These findings shed light on CD8(+) T cell noncytotoxic antiviral factor production during HIV infection.

Sujetos con c&eacute;lulas CD8 cuenta mayor que 200/microl produce niveles elevados de MIP-1alfa por subconjuntos CD8(+)CD28(+), CD8(+)CD38(-) y CD8(+)HLA-DR(+) en comparaci&oacute;n con los controles no infectados. Human immunodeficiency virus (HIV) is a mucosally transmitted infection that rapidly targets and depletes CD4+ T cells in mucosal tissues and establishes a major reservoir for viral persistence in gut-associated lymphoid tissues. Therefore, vaccines designed to prevent HIV infections must induce potent and durable mucosal immune responses, especially in the genital tract. Here we investigated whether intranasal (i.n.) immunization with inactivated gp120-depleted HIV-1 antigen (Ag) plus CpG oligodeoxynucleotide (ODN) as an adjuvant induced local immune responses in the genital tract and cross-clade protection against intravaginal (IVAG) challenge. Lymphocytes isolated from the iliac lymph nodes (ILNs) and genital tracts of female mice i.n. immunized with HIV-1 Ag plus CpG showed significant HIV-specific proliferation and produced significantly higher levels of gamma interferon (IFN-gamma) and beta-chemokines than mice immunized with HIV-1 Ag alone or mixed with non-CpG ODN. CD8+ lymphocytes were dramatically increased in the genital tracts of mice immunized with HIV-1 Ag plus CpG, and protection following IVAG challenge with recombinant vaccinia viruses (rVVs) expressing HIV-1 gag was shown to be CD8 dependent. Finally, cross-clade protection was observed between clades A, C, and G but not B following IVAG challenge with rVVs expressing HIV-1 gag from different clades. These studies provide evidence that mucosal (i.n.) immunization induced strong local T-cell-mediated immune responses in the genital tract and cross-clade protection against IVAG challenge.

Virus de inmunodeficiencia humana (VIH) es una infecci&oacute;n de transmisi&oacute;n mucosally que r&aacute;pidamente apunta y agota las c&eacute;lulas T CD4 + en los tejidos de la mucosas y establece un reservorio importante para persistencia viral en tejidos linfoides gut-asociados. Por lo tanto, las vacunas dise&ntilde;adas para prevenir infecciones por el VIH deben inducir respuestas inmunitarias mucosas potentes y duraderas, especialmente en el tracto genital. Aqu&iacute; Investigamos si la vacuna intranasal (i.n.) gp120-agotado HIV-1 ant&iacute;geno inactivado (Ag) y oligodeoxynucleotide CpG (ODN) como coadyuvante inducida inmunorespuestas locales en la zona genital y la protecci&oacute;n de la Cruz-clado contra intravaginal challenge (IVAG). Linfocitos aislados de los ganglios il&iacute;acos (ILNs) y tractos genitales de ratones hembra i.n. inmunizados con HIV-1 Ag m&aacute;s CpG mostraron proliferaci&oacute;n significativa relacionadas con el VIH y producidos significativamente mayores niveles de interfer&oacute;n gamma (IFN-gamma) y beta-quimiocinas que ratones inmunizados con HIV-1 Ag solo o mezclado con CpG ODN de-no. Los linfocitos CD8 + se incrementaron dram&aacute;ticamente en el tracto genital de ratones inmunizados con HIV-1 Ag m&aacute;s CpG y protecci&oacute;n tras desaf&iacute;o IVAG con virus vaccinia recombinante (rVVs), expresando la mordaza de HIV-1 mostr&oacute; ser CD8 dependiente. Finalmente, protecci&oacute;n de la Cruz-clado se observ&oacute; entre clados A, C, y G pero no B tras desaf&iacute;o IVAG con rVVs expresando HIV-1 gag de clados diferentes. Estos estudios proporcionan evidencia que mucosa (i.n.) inmunizaci&oacute;n inducidas fuerte local mediada por c&eacute;lulas-T inmune en el aparato genital y la protecci&oacute;n de la Cruz-clado contra desaf&iacute;o IVAG.

CD8 T-cell response provides an important defense against rotavirus, which infects a variety of systemic locations in addition to the gut. Here we investigated the distribution, phenotype, and function of rotavirus-specific CD8 T cells in multiple organs after rotavirus infection initiated via the intranasal, oral, or intramuscular route. The highest level of virus-specific CD8 T cells was observed in the Peyer's patches of orally infected mice and in the lungs of intranasally infected animals. Very low levels of virus-specific CD8 T cells were detected in peripheral blood or spleen irrespective of the route of infection. Rotavirus-specific CD8 T cells from Peyer's patches of orally infected mice expressed high levels of CCR9, while CXCR6 and LFA-1 expression was associated with virus-specific CD8 T cells in lungs of intranasally infected mice. Oral infection induced the highest proportion of gamma interferon(-) CD107a/b(+) CD8 T cells in Peyer's patches. When equal numbers of rotavirus-specific CD8 T cells were transferred into Rag-1 knockout mice chronically infected with rotavirus, the donor cells derived from Peyer's patches of orally infected mice were more efficient than those derived from lungs of intranasally infected animals in clearing intestinal infection. These results suggest that different routes of infection induce virus-specific CD8 T cells with distinct homing phenotypes and effector functions as well as variable abilities to clear infection.

Qualitative and quantitative characteristics of rotavirus-specific CD8 T cells vary depending on the route of infection.

Rotavirus espec&iacute;ficos de c&eacute;lulas T CD8 de placas de Peyer de ratones infectados por v&iacute;a oral expresaron altos niveles de CCR9, mientras que CXCR6 y LFA-1 expresi&oacute;n se asoci&oacute; con virus espec&iacute;ficos de c&eacute;lulas de T CD8 en pulmones de ratones infectados intranasal. Infecci&oacute;n oral inducida por la mayor proporci&oacute;n de c&eacute;lulas de T de CD8 de CD107a/b(+) de interferon(-) de gamma en placas de Peyer. Major impediments to developing a Chlamydia vaccine lie in identifying immunologically relevant T-cell antigens and delivery in a manner to stimulate protective immunity. Using an immunoproteomic approach, we previously identified three immunodominant Chlamydia T-cell antigens (PmpG-1, PmpE/F-2, and RplF). Because RplF has high homology to a human ortholog, it may not be suitable for human vaccine development. Therefore, in this study, we evaluated protection against Chlamydia infection in the genital tract in C57BL/6 mice immunized with Chlamydia-specific membrane proteins PmpG-1, PmpE/F-2, and major outer membrane protein (MOMP; as a reference) or a combination of them formulated with one of three adjuvants, CpG oligodeoxynucleotide (CpG-ODN), AbISCO-100 (AbISCO), or DDA/TDB (dimethyldioctadecylammonium bromide/D-(+)-trehalose 6,6'-dibehenate). The results show that immunization with the CpG-ODN formulation failed to provide protection against Chlamydia infection; the AbISCO formulation conferred moderate protection, and the DDA/TDB formulation showed the highest degree of protective efficacy. The combination of PmpG-1, PmpE/F-2, and MOMP proteins formulated with DDA/TDB exhibited the greatest degree of protection among all vaccine groups studied. Moreover, this vaccine combination also engendered significant protection in BALB/c mice, which have a different major histocompatibility complex (MHC) background. We measured cell-mediated immune cytokine responses in mice immunized with PmpG-1 mixed with each of the three adjuvants. The results demonstrate that mice immunized with the DDA/TDB formulation induced the strongest gamma interferon (IFN-gamma) and interleukin-17 (IL-17) responses, characterized by the highest frequency of IFN-gamma/tumor necrosis factor alpha (TNF-alpha) and IFN-gamma/IL-17 double-positive CD4(+) T cells. In conclusion, a Chlamydia vaccine based on the recombinant proteins PmpG-1, PmpE/F-2, and MOMP delivered in a DDA/TDB adjuvant conferred protection against infection that correlated with IFN-gamma/TNF-alpha and IFN-gamma/IL-17 double-positive CD4(+) T cells.

Des obstacles majeurs au d&eacute;veloppement d&#39;un vaccin contre la Chlamydia se trouvent dans l&#39;identification des antig&egrave;nes de T-cellules immunologiquement comp&eacute;tentes et livraison de mani&egrave;re &agrave; stimuler l&#39;immunit&eacute; protectrice. Gro&szlig;e Hindernisse f&uuml;r die Entwicklung eines Impfstoffes Chlamydien liegen bei der Ermittlung von immunologisch relevanten T-Zell-Antigene und Lieferung in einer Weise zu sch&uuml;tzende Immunit&auml;t zu stimulieren. Immunoproteomic herantreten, haben wir bereits drei immundominante Chlamydien T-Zell-Antigene (PmpG-1, PmpE/F-2 und RplF) identifiziert. Da RplF hohe Homologie zu einem menschlichen Ortholog verf&uuml;gt, kann es nicht f&uuml;r menschliche Impfstoffentwicklung geeignet. Daher in dieser Studie ausgewertet wir gegen eine Chlamydien Infektion in den Genitaltrakt bei C57BL/6 M&auml;usen mit Chlamydien-spezifische Membranproteine PmpG-1 PmpE/F-2 und gro&szlig;en &auml;u&szlig;eren Membranprotein geimpft (MOMP; als Referenz) oder eine Kombination davon mit einem der drei Adjuvantien, CpG Oligodeoxynucleotide (CpG-ODN), AbISCO-100 (AbISCO) oder DDA/TDB formuliert (Dimethyldioctadecylammonium Bromid/D-(+)-Trehalose 6, 6&#39;-Dibehenate). Die Ergebnisse zeigen, dass die Immunisierung mit der CpG-ODN-Formulierung konnte gegen eine Chlamydien-Infektion zur Verf&uuml;gung zu stellen; die AbISCO-Formulierung &uuml;bertragenen m&auml;&szlig;iger Schutz und die DDA/TDB-Formulierung zeigte den h&ouml;chsten Grad der Schutzwirkung. Principali ostacoli allo sviluppo di un vaccino clamidia si trovano nell&#39;identificare gli antigeni cellule T immunologicamente pertinenti e consegna in modo da stimolare l&#39;immunit&agrave; protettiva. Utilizzando un approccio di immunoproteomic, abbiamo precedentemente individuato tre antigeni di Chlamydia T-cellula immunodominanti (PmpG-1, PmpE/F-2 e RplF). Perch&eacute; RplF ha elevata omologia ad un ortholog umano, pu&ograve; non essere adatto per lo sviluppo del vaccino umano. Despite the reports of dysfunction of the lytic abilities of CD8(+) T cells during human immunodeficiency virus-1 (HIV-1) disease progression, the effects of infection on the noncytolytic functions of CD8(+) T cells have not been well characterized to date. We examined the effect of HIV-1 infection on the cytokine and chemokine responses of peripheral blood-derived CD8(+) T cells in an in vitro system. Activation of HIV-1-infected CD8(+) T cells with phytohemagglutinin resulted in a 4- to 8-fold increase in the production of macrophage inflammatory protein (MIP)-1Î±, MIP-1Î², regulated on activation normal T-cell expressed and secreted, and interleukin (IL)-16. Treatment of activated HIV-1-infected CD8(+) T cells with anti-CD3 monoclonal (M) antibody (Ab) and IL-15 induced strong production of interferon-Î³ (IFN-Î³). Treatment of cells with anti-IL-12 MAb and IL-4 to induce a Tc1-to-Tc2 shift resulted in no change in viral production levels or IFN-Î³ production within the HIV-1-infected CD8(+) T cell population. Initiation of a Tc2-to-Tc1 shift resulted in a 6-fold increase in HIV-1 replication and 2- to 3-fold higher levels of IFN-Î³, demonstrating that infection can protect against a Tc1-to-Tc2 shift in CD8(+) T cells.

Tratamiento de activado infectadas por VIH-1 CD8 T c&eacute;lulas con anticuerpo monoclonal de anti-CD3 (M) (Ab) y IL-15 inducida por la producci&oacute;n de interfer&oacute;n-&gamma; (IFN-&gamma;). Tratamiento de c&eacute;lulas con MAb anti-IL-12 e IL-4 para inducir un cambio Tc1-a-Tc2 dio lugar a ning&uacute;n cambio en los niveles de producci&oacute;n viral o la producci&oacute;n de IFN-&gamma; dentro de la poblaci&oacute;n de c&eacute;lulas T CD8 de infectadas por VIH-1. Iniciaci&oacute;n de un cambio de Tc2-a-Tc1 result&oacute; en un incremento de seis en la replicaci&oacute;n del VIH-1 y 2 a 3 veces mayores niveles de IFN-&gamma;, demostrando que la infecci&oacute;n puede proteger contra un cambio de Tc1-a-Tc2 en linfocitos T CD8.

T cells with the specialized ability to suppress both adaptive and innate immune responses have been identified and called T regulatory cells (Tregs). The primary function of Tregs is to maintain a balance between immunity (foreign Ag) and tolerance (self Ag) to tissues. Tregs prevent autoimmune disease, maintain immune homeostasis and modulate protective responses against infection. Tregs function in two ways; 1) limiting the magnitude of effector responses which influence the adequate control of infection and 2) control collateral tissue damage caused by vigorous antimicrobial responses against pathogens. Initially, the immune suppressive ability of CD4 T cells was predicted by expression of the forkhead box p3 (Foxp3) transcription factor. However, many reports have demonstrated immune suppressive function in an array of other T cells which include iT(R)35, CD8+, NKT cells, especially in mucosal tissues. The immune suppressive mechanisms of Tregs include contact-dependent, cytokine secretion and regulation of immune cell migration. The expanded group of Tregs is crucial for protecting the function of mucosal tissues such as the gut, respiratory and genital tracts, as these tissues are routinely exposed to foreign pathogens.
