Non-alcoholic steatohepatitis (NASH), the inflammatory subtype in the progression of non-alcoholic fatty liver disease, is becoming a serious burden threatening human health, but no approved medication is available to date. Mononoside is a natural active substance derived from Cornus officinalis and has been confirmed to have great potential in regulating lipid metabolism in our previous studies. However, its effect and mechanism to inhibit the progression of NASH remains unclear.

Anthracycline (ANT)-induced cardiotoxicity (AIC) is a particularly prominent form of cancer therapy-related cardiovascular toxicity leading to the limitations of ANTs in clinical practice. Even though AIC has drawn particular attention, the best way to treat it is remaining unclear. Updates to AIC therapy have been made possible by recent developments in research on the underlying processes of AIC. We review the current molecular pathways leading to AIC: 1) oxidative stress (OS) including enzymatic-induced and other mechanisms; 2) topoisomerase; 3) inflammatory response; 4) cardiac progenitor cell damage; 5) epigenetic changes; 6) renin-angiotensin-aldosterone system (RAAS) dysregulation. And we systematically discuss current prevention and treatment strategies and novel pathogenesis-based therapies for AIC: 1) dose reduction and change; 2) altering drug delivery methods; 3) antioxidants, dexrezosen, statina, RAAS inhibitors, and hypoglycemic drugs; 4) miRNA, natural phytochemicals, mesenchymal stem cells, and cardiac progenitor cells. We also offer a fresh perspective on the management of AIC by outlining the current dilemmas and challenges associated with its prevention and treatment.

Precision Treatment of Anthracycline-Induced Cardiotoxicity: An Updated Review.

Studies have substantiated that dietary-fat- and fructose-overconsumption-caused lipid metabolism disorders can trigger renal lipotoxicity to drive the progression of chronic kidney disease (CKD). This study was conducted to evaluate the efficacy of morroniside, a natural active substance extracted from the fruit of , in inhibiting the progression of CKD in high-fat and high-fructose-fed mice. Our results showed histological changes such as fatty degeneration of renal tubular cells, tubular dilatation, glomerular fibrosis, and abnormal renal function in the kidneys of high-fat- and high-fructose-fed mice, which was significantly improved after morroniside treatment. Mechanistically, morroniside maintained renal lipid metabolism homeostasis and inhibited NLRP3 inflammatory vesicle activation by activating AMPKα to promote TFEB nuclear translocation-mediated lipophagy. Consistent results were observed in palmitic acid-induced HK-2 cells. Notably, silencing AMPKα or TFEB both reversed the effects of morroniside in promoting lipophagy and inhibiting the activation of inflammatory responses in vivo and in vitro. Therefore, our study provides compelling evidence that morroniside delays CKD progression by promoting AMPK/TFEB-mediated lipophagy and inhibiting NLRP3 inflammasome activation, suggesting that dietary supplementation with morroniside and morroniside-rich foods (such as ) might be an effective strategy for the prevention of CKD.

Morroniside Ameliorates High-Fat and High-Fructose-Driven Chronic Kidney Disease by Motivating AMPK-TFEB Signal Activation to Accelerate Lipophagy and Inhibiting Inflammatory Response.

Esophageal cancer (EC) is a malignant tumor with a high recurrence and metastasis rate and poor prognosis. In 2024, China ranked first in the world in terms of new EC cases and deaths. Surgery is the main treatment method for EC, but the clinical difficulty is how to prevent recurrence and metastasis after surgery. Traditional Chinese medicine as a complementary therapy has played an important role in this regard. Preclinical studies have confirmed that Qizhu Yuling Prescription (QZYLP) has anticancer effects, reduces treatment side effects, and improves quality of life, except for the lack of long-term prognostic results. Therefore, this study aims to investigate whether QZYLP can reduce the recurrence and metastasis rates of EC after surgery, improve disease-free survival (DFS), prolong overall survival, and observe the safety of the drug.
