Bone metabolic disorders, constituting a group of prevalent and grave conditions, currently have a scarcity of therapeutic alternatives. Over the recent past, exosomes have been at the forefront of research interest, owing to their nanoparticulate nature and potential for therapeutic intervention. ncRNAs are a class of heterogeneous transcripts that they lack protein-encoding capacity, yet they can modulate the expression of other genes through multiple mechanisms. Mounting evidence underscores the intricate role of exosomes as ncRNAs couriers implicated in the pathogenesis of bone metabolic disorders. In this review, we endeavor to elucidate recent insights into the roles of three ncRNAs - miRNAs, lncRNAs, and circRNAs - in bone metabolic ailments such as osteoporosis, osteoarthritis, and rheumatoid arthritis. Additionally, we examine the viability of exosomal ncRNAs as innovative, cell-free modalities in the diagnosis and therapeutic management of bone metabolic disorders. We aim to uncover the critical function of exosomal ncRNAs within the context of bone metabolic diseases.

Perspectives of exosomal ncRNAs in the treatment of bone metabolic diseases: Focusing on osteoporosis, osteoarthritis, and rheumatoid arthritis.

Ovarian ageing is a key factor in the decline of female fertility. It is primarily characterised by diminished oocyte quality, follicular depletion, and dysregulated hormone levels. In recent years, mitochondria-driven inflammation has emerged as a potential mechanism in ovarian ageing. Mitochondrial dysfunction results in the accumulation of reactive oxygen species (ROS) and the release of mitochondrial DNA (mtDNA), as well as the leakage of mitochondrial components and metabolites into the cytosol or extracellular space. These elements act as damage-associated molecular patterns (DAMPs), activating inflammasomes like NLRP3, thereby initiating and amplifying innate immune responses and contributing to sustained inflammation. Furthermore, an imbalance in mitochondrial quality control mechanisms can worsen the spread and persistence of inflammatory responses. In this study, we present a comprehensive overview of the signalling origins, molecular mechanisms of amplification, and key regulatory nodes involved in mitochondria-driven inflammation during ovarian ageing. Finally, we summarise potential therapeutic strategies targeting mitochondria-driven inflammation, offering novel perspectives and targets for delaying ovarian ageing and enhancing female reproductive health.

Mitochondria-driven inflammation: a new frontier in ovarian ageing.

Mitochondria are powerful multifunctional intracellular organelles involved in a wide array of cellular processes. Emerging evidence has highlighted mitochondria as pivotal activators of innate immunity and key participants in inflammatory responses, including in sterile inflammatory diseases, beyond their well-established role as the "powerhouse" of cells. Osteoporosis (OP) is a metabolic disorder characterized by chronic low-grade inflammation, suggesting potential therapeutic value for OP treatment by targeting mitochondria-driven inflammatory pathways. Current research on mitochondrial involvement in OP primarily focuses on its functions in energy metabolism, calcium signaling transduction, oxidative stress, and cell death. However, it remains elusive regarding the precise relationship between mitochondria-mediated inflammatory mechanisms and OP pathogenesis. Accordingly, this review aims to synthesize recent advancement in understanding mitochondria-mediated inflammatory mechanisms and their implications in OP development, to offer insights that may inform future mechanistic investigations and therapeutic strategy development.
