The term short-term saccadic adaptation (STSA) captures our ability to unconsciously move the endpoint of a saccade to the final position of a visual target that has jumped to a new location during the saccade. STSA depends on the integrity of the cerebellar vermis. We tested the hypothesis that STSA reflects the working of a cerebellar mechanism needed to avoid 'fatigue', a gradual drop in saccade amplitude during a long series of stereotypic saccades. To this end we compared the kinematics of saccades of 14 patients suffering from different forms of cerebellar disease with those of controls in two tests of STSA and a test of saccadic resilience. Controls showed an increase in saccade amplitude (SA) for outward adaptation, prompted by outward target shifts, due to an increase in saccade duration (SD) in the face of constant peak velocity (PV). The decrease in SA due to inward adaptation was, contrariwise, accompanied by a drop in PV and SD. Whereas patients with intact vermis did not differ from controls, those with vermal pathology lacked outward adaptation: SD remained constant, as did SA and PV. In contrast, vermal patients demonstrated a significant decrease in SA, paralleled by a decrease in PV but mostly unaltered SD in the inward adaptation experiment as well as in the resilience test. These findings support the notion that inward adaptation is at least partially based on uncompensated fatigue. On the other hand, outward adaptation reflects an active mechanism for the compensation of fatigue, residing in the cerebellum.

L&#39;adaptation par saccades &agrave; court terme (STSA) capte notre capacit&eacute; inconsciemment vers le point de terminaison d&#39;une saccade la position finale d&#39;une cible visuelle qui a saut&eacute; vers un nouvel emplacement au cours de la saccade. STSA d&eacute;pend de l&#39;int&eacute;grit&eacute; du vermis c&eacute;r&eacute;belleux. Nous avons test&eacute; l&#39;hypoth&egrave;se que la STSA refl&egrave;te le fonctionnement d&#39;un m&eacute;canisme c&eacute;r&eacute;belleux n&eacute;cessaire pour &eacute;viter la &laquo; fatigue &raquo;, une baisse progressive en amplitude saccade pendant une longue s&eacute;rie de saccades st&eacute;r&eacute;otyp&eacute;s. &Agrave; cette fin, nous avons compar&eacute; la cin&eacute;matique des saccades 14 patients souffrant de diff&eacute;rentes formes de maladie c&eacute;r&eacute;belleuse avec ceux des contr&ocirc;les contenus dans deux tests de STSA et un test de r&eacute;silience par saccades. En revanche, les patients vermal a d&eacute;montr&eacute; une diminution significative de SA, accompagn&eacute;e d&#39;une diminution de PV mais essentiellement inchang&eacute;e SD dans l&#39;exp&eacute;rience d&#39;adaptation vers l&#39;int&eacute;rieur ainsi que dans l&#39;essai de r&eacute;silience. Ces r&eacute;sultats confortent l&#39;id&eacute;e qu&#39;adaptation vers l&#39;int&eacute;rieur est au moins partiellement issue de fatigue sans compensation. En revanche, l&#39;ext&eacute;rieur adaptation refl&egrave;te un m&eacute;canisme actif pour la compensation de la fatigue, r&eacute;sidant dans le cervelet.

Die Begriff kurzfristige saccadischen Anpassung (STSA) nimmt unsere F&auml;higkeit unbewusst den Endpunkt der ein Saccade in die Endposition der eine visuelle Ziel verschieben, die an einen neuen Speicherort w&auml;hrend der Saccade gesprungen ist. STSA h&auml;ngt die Integrit&auml;t des Kleinhirnwurms. Wir testeten die Hypothese, dass STSA das Funktionieren eines zerebell&auml;re Mechanismus erforderlich, um zu vermeiden, &quot;M&uuml;digkeit&quot;, einem allm&auml;hlichen R&uuml;ckgang der Saccade Amplitude w&auml;hrend einer langen Reihe von Stereotype Sakkaden widerspiegelt. Der R&uuml;ckgang der SA durch innere Anpassung wurde, umgekehrt, begleitet von einem R&uuml;ckgang der PV und SD w&auml;hrend Patienten mit intakten Vermis Unterschied sich nicht von Steuerelementen, die mit Vermal Pathologie fehlte nach au&szlig;en Anpassung: SD blieb konstant, wie SA und PV. Im Gegensatz dazu zeigte Vermal Patienten einen betr&auml;chtlicher R&uuml;ckgang der SA, in &auml;hnlicher Weise wurden durch eine Abnahme der PV aber meist unver&auml;ndert SD im Inneren Anpassung-Experiment sowie der Belastbarkeit-Test. Diese Befunde unterst&uuml;tzen das Konzept nach innen Anpassung beruht zumindest teilweise auf Unkompensiertes M&uuml;digkeit. Auf der anderen Seite spiegelt die &auml;u&szlig;erliche Anpassung ein aktiven Mechanismus zum Ausgleich der M&uuml;digkeit, wohnten im Kleinhirn.

STSA dipende dall&#39;integrit&agrave; del verme cerebellare. Abbiamo testato l&#39;ipotesi che la STSA riflette il funzionamento di un meccanismo cerebellare necessari per evitare la &#39;fatica&#39;, un graduale calo di ampiezza saccade durante una lunga serie di saccadi stereotipati. A tal fine abbiamo confrontato la cinematica delle saccadi di 14 pazienti affetti da diverse forme di malattia cerebellare con quelli dei controlli in due prove di STSA e una prova di resilienza saccadici. I controlli hanno mostrato un aumento in ampiezza saccade (SA) per adattamento esteriore, richiesto da mutamenti di destinazione verso l&#39;esterno, a causa di un aumento della durata saccade (SD) a fronte di velocit&agrave; di picco costante (PV). La diminuzione di SA a causa di adattamento verso l&#39;interno era, al contrario, accompagnata da una goccia in PV e SD. considerando che i pazienti con verme intatto non differiscono dai controlli, quelli con patologia vermal mancava di adattamento verso l&#39;esterno: SD &egrave; rimasto costante, come ha fatto SA e PV. Al contrario, vermal pazienti ha dimostrato una significativa diminuzione in SA, parallelizzata di una diminuzione in PV ma per lo pi&ugrave; inalterato SD l&#39;esperimento di adattamento verso l&#39;interno cos&igrave; come la prova di resilienza. Questi risultati supportano la nozione che adattamento verso l&#39;interno &egrave; almeno parzialmente basata sulla fatica non compensata. D&#39;altra parte, adattamento esteriore riflette un meccanismo attivo per la compensazione della fatica, che risiedono nel cervelletto.

La adaptaci&oacute;n sac&aacute;dicos a corto plazo (STSA) captura nuestra capacidad para mover inconscientemente el punto final de una sacudida a la posici&oacute;n final de un objetivo visual que ha saltado a una nueva ubicaci&oacute;n durante la sacudida. STSA depende de la integridad de los vermis cerebelosos. Probamos la hip&oacute;tesis que STSA refleja el funcionamiento de un mecanismo cerebeloso para evitar &#39;fatiga&#39;, una ca&iacute;da gradual en amplitud sacada durante una larga serie de movimientos sac&aacute;dicos estereotipados. Para ello se compar&oacute; la cinem&aacute;tica de los movimientos sac&aacute;dicos de 14 pacientes que padecen diversas formas de enfermedad cerebelosa con los controles en las dos pruebas de STSA y una prueba de resistencia sac&aacute;dicos. Controles mostraron un aumento en la amplitud de la sacudida (SA) de adaptaci&oacute;n exterior, motivada por cambios de destino hacia afuera, debido a un aumento en la duraci&oacute;n de la sacudida (SD) frente a velocidad constante de pico (PV). La disminuci&oacute;n en SA debido a la adaptaci&oacute;n interna fue, por el contrario, acompa&ntilde;ada de una ca&iacute;da en PV y SD mientras que pacientes con vermis intactos no difiri&oacute; de controles, aquellos con patolog&iacute;a vermal carec&iacute;an de adaptaci&oacute;n externa: SD se mantuvo constante, como lo hizo SA y PV. Por el contrario, vermal pacientes demostraron una disminuci&oacute;n significativa en SA, acompa&ntilde;ado por una disminuci&oacute;n en PV pero sobre todo inalterado SD en el experimento de adaptaci&oacute;n interna, as&iacute; como en la prueba de resistencia. Estos resultados apoyan la noci&oacute;n que la adaptaci&oacute;n interna al menos parcialmente se basa en fatiga no compensada. Por otro lado, la adaptaci&oacute;n exterior refleja un mecanismo activo para la compensaci&oacute;n de la fatiga, que reside en el cerebelo.

Prevailing concepts on the control of goal-directed hand movements (HM) have focused on a network of cortical areas whose early parieto-occipital stages are thought to extract and integrate information on target and hand location, involving subsequent stages in frontal cortex forming and executing movement plans. The substantial experimental results supporting this "cortical network" concept for hand movements notwithstanding, the concept clearly needs refinement to account for the surprisingly mild HM disturbances resulting from disconnecting the parieto-occipital from the frontal stages of the network. Clinical observations have suggested the cerebropontocerebellar projection as an alternative pathway for the sensory guidance of HM. As a first step in assessing its role, we explored the pontine nuclei (PN) of rhesus monkeys, trained to make goal-directed hand and eye movements guided by spatial memory. We were indeed able to delineate a distinct cluster of neurons in the rostrodorsal PN, activated by the preparation and the execution of hand reaches, close to but distinct from the region in which saccade-related neurons may be observed. The movement-related activity of HM-related neurons starts earlier than that of saccade-related neurons and both neuron types are usually effector specific, i.e., they respond only to the movement of the preferred effector. This is also the case when motor synergies involving both effectors are executed. Our findings support the notion of a distinct precerebellar, pontine visuomotor channel for hand reaches that is anatomically and functionally largely separated from the one serving eye movements.

Notions sur le contr&ocirc;le de main cibl&eacute; mouvements (HM) ont mis l&#39;accent sur un r&eacute;seau d&#39;aires corticales dont d&eacute;buts pari&eacute;to-occipitale sont cens&eacute;s d&#39;extraire et d&#39;int&eacute;grer des informations sur l&#39;emplacement cible et de la main, impliquant des &eacute;tapes ult&eacute;rieures dans le cortex frontal, formant et l&#39;ex&eacute;cution de plans de circulation en vigueur. Nous &eacute;tions en effet en mesure de d&eacute;limiter un cluster distinct des neurones dans le rostrodorsal PN, activ&eacute; par la pr&eacute;paration et l&#39;ex&eacute;cution de main atteint, pr&egrave;s de mais distincte de la r&eacute;gion dans laquelle les neurones associ&eacute;s au saccade peuvent &ecirc;tre observ&eacute;s. L&#39;activit&eacute; li&eacute;e au mouvement des neurones associ&eacute;s au HM commence plus t&ocirc;t que celui des neurones associ&eacute;s au saccade et les deux types de neurones sont g&eacute;n&eacute;ralement effecteurs sp&eacute;cifiques, c&#39;est-&agrave;-dire qu&#39;ils r&eacute;pondent seulement &agrave; la circulation de l&#39;effecteur pr&eacute;f&eacute;r&eacute;. C&#39;est &eacute;galement le cas lorsque des synergies moteurs impliquant les deux effecteurs sont ex&eacute;cut&eacute;es. Conceptos vigentes en el control de mano intencionadas movimientos (HM) se han centrado en una red de &aacute;reas corticales cuyas primeras etapas parieto-occipital se piensan extraer e integrar la informaci&oacute;n sobre la ubicaci&oacute;n de destino y de la mano, con etapas posteriores en la corteza frontal, formaci&oacute;n y ejecuci&oacute;n de planes del movimiento. Los resultados experimentales sustanciales apoyo a este concepto de &quot;red cortical&quot; para los movimientos de la mano a pesar, el concepto claramente necesita refinamiento para tener en cuenta los disturbios de HM sorprendentemente suaves resultantes de desconectar el parieto-occipital de las etapas frontales de la red. Observaciones cl&iacute;nicas han sugerido la proyecci&oacute;n de la cerebropontocerebellar como una v&iacute;a alternativa para la direcci&oacute;n sensorial de HM. Como un primer paso para evaluar su papel, exploramos los n&uacute;cleos del pontine (PN) de los monos rhesus, entrenados para hacer intencionadas mano y ojo movimientos guiados por la memoria espacial. De hecho hemos podido delinear un grupo distinto de neuronas en el rostrodorsal PN, activado por la preparaci&oacute;n y la ejecuci&oacute;n de mano alcanza, cerca pero distinto de la regi&oacute;n en la que pueden observarse las neuronas relacionadas con la sacudida. La actividad relacionada con el movimiento de neuronas relacionadas con HM comienza antes que el de las neuronas relacionadas con la sacudida y ambos tipos de neurona son generalmente efectoras espec&iacute;ficas, es decir, responden s&oacute;lo a la circulaci&oacute;n del efector preferido. Esto tambi&eacute;n es el caso cuando se ejecutan motores sinergias que ambos efectores. Nuestro resultados apoyan que la noci&oacute;n de un canal de visuomotor precerebellar, del pontine distinta para la mano alcanza se separa anat&oacute;micamente y funcionalmente en gran parte de los movimientos del ojo de una porci&oacute;n.

The pontine nuclei (PN) are the major intermediary elements in the corticopontocerebellar pathway. Here we asked if the PN may help to adapt the spatial reference frames used by cerebrocortical neurons involved in the sensory guidance of movement to a format potentially more appropriate for the cerebellum. To this end, we studied movement-related neurons in the dorsal PN (DPN) of monkeys, most probably projecting to the cerebellum, executing fixed vector saccades or, alternatively, fixed vector hand reaches from different starting positions. The 83 task-related neurons considered fired movement-related bursts before saccades (saccade-related) or before hand movements (hand movement-related). About 40% of the SR neurons were "oculocentric," whereas the others were modulated by eye starting position. A third of the HMR neurons encoded hand reaches in hand-centered coordinates, whereas the remainder exhibited different types of dependencies on starting positions, reminiscent in general of cortical responses. All in all, pontine reference frames for the sensory guidance of movement seem to be very similar to those in cortex. Specifically, the frequency of orbital position gain fields of SR neurons is identical in the DPN and in one of their major cortical inputs, lateral intraparietal area (LIP).

Dysfunction of the inner ear as caused by presbyacusis, injuries or noise traumata may result in subjective tinnitus, but not everyone suffering from one of these diseases develops a tinnitus percept and vice versa. The reasons for these individual differences are still unclear and may explain why different treatments of the disease are beneficial for some patients but not for others. Here we for the first time compare behavioral and neurophysiological data from hearing impaired Mongolian gerbils with (T) and without (NT) a tinnitus percept that may elucidate why some specimen do develop subjective tinnitus after noise trauma while others do not. Although noise trauma induced a similar permanent hearing loss in all animals, tinnitus did develop only in about three quarters of these animals. NT animals showed higher overall cortical and auditory brainstem activity before noise trauma compared to T animals; that is, animals with low overall neuronal activity in the auditory system seem to be prone to develop tinnitus after noise trauma. Furthermore, T animals showed increased activity of cortical neurons representing the tinnitus frequencies after acoustic trauma, whereas NT animals exhibited an activity decrease at moderate sound intensities by that time. Spontaneous activity was generally increased in T but decreased in NT animals. Plastic changes of tonotopic organization were transient, only seen in T animals and vanished by the time the tinnitus percept became chronic. We propose a model for tinnitus prevention that points to a global inhibitory mechanism in auditory cortex that may prevent tinnitus genesis in animals with high overall activity in the auditory system, whereas this mechanism seems not potent enough for tinnitus prevention in animals with low overall activity.

Plastica cambiamenti dell&#39;organizzazione tonotopico sono stati transitori, solo visto negli animali T e scomparso da tempo che la percezione dell&#39;acufene &egrave; diventato cronico. Vi proponiamo un modello per la prevenzione di acufene che punta ad un meccanismo inibitorio globale nella corteccia uditiva che possa impedire la genesi di acufene negli animali con alta attivit&agrave; complessiva del sistema uditivo, considerando che questo meccanismo non sembra abbastanza potente per la prevenzione dell&#39;acufene in animali con bassa attivit&agrave; complessiva.

Disfunci&oacute;n del o&iacute;do interno causado por la presbiacusia, lesiones o traumas de ruido puede causar tinnitus subjetivo, pero no todo el mundo sufren de una de estas enfermedades convierte un zumbido percept y viceversa. Las razones de estas diferencias individuales a&uacute;n no est&aacute;n claros y pueden explicar por qu&eacute; los diferentes tratamientos de la enfermedad son beneficiosos para algunos pacientes, pero no para otros. Aqu&iacute; estamos para el primeros tiempo compara comportamentales y neurofisiol&oacute;gicos datos de audiencia hab&iacute;an deteriorada jerbos de Mongolia con (T) y sin percept (NT) un zumbido que puede aclarar por qu&eacute; algunas muestras desarrollar tinnitus subjetivo despu&eacute;s del trauma de ruido mientras que otros no. Aunque el trauma de ruido inducido por una permanente similar p&eacute;rdida en todos los animales de la audici&oacute;n, zumbido se desarroll&oacute; s&oacute;lo en unas tres cuartas partes de estos animales. NT animales presentaron una mayor actividad en general cortical y auditiva del tronco encef&aacute;lico antes de trauma de ruido en comparaci&oacute;n con animales de T; es decir, animales con baja actividad general neuronal en el sistema auditivo parecen ser propensas a desarrollar zumbido despu&eacute;s del trauma de ruido. Adem&aacute;s, T animales presentaron aumento de la actividad de las neuronas corticales que representan las frecuencias de tinnitus despu&eacute;s de trauma ac&uacute;stico, mientras que los animales NT exhibieron una disminuci&oacute;n de la actividad moderada intensidades de sonido en ese momento. Actividad espont&aacute;nea generalmente se aument&oacute; en T pero disminuy&oacute; en animales de NT. Cambios pl&aacute;sticos de organizaci&oacute;n tonot&oacute;pica eran transitorios, s&oacute;lo visto en animales de T y desapareci&oacute; cuando el percept zumbido se convirti&oacute; en cr&oacute;nica. Se propone un modelo para la prevenci&oacute;n de zumbido que apunta a un mecanismo de inhibici&oacute;n global en la corteza auditiva que podr&iacute;a prevenir la g&eacute;nesis de zumbido en animales con alta actividad general en el sistema auditivo, Considerando que este mecanismo no parece lo suficientemente potente como para la prevenci&oacute;n de zumbido en animales con baja actividad general.
