Circular RNAs (circRNAs) play crucial biological functions in various tumors, including bladder cancer (BCa). However, the roles and underlying molecular mechanisms of circRNAs in the malignant proliferation of BCa are yet unknown. CircKDM1A was observed to be downregulated in BCa tissues and cells. Knockdown of circKDM1A promoted the proliferation of BCa cells and bladder xenograft growth, while the overexpression of circKDM1A exerts the opposite effect. The dual-luciferase reporter assay revealed that circKDM1A was directly bound to miR-889-3p, acting as its molecular sponge to downregulate CPEB3. In turn, the CPEB3 was bound to the CPE signal in p53 mRNA 3'UTR to stabilize its expression. Thus, circKDM1A-mediated CPEB3 downregulation inhibits the stability of p53 mRNA and promotes BCa malignant progression. In conclusion, circKDM1A functions as a tumor suppressor in the malignant proliferation of BCa via the miR-889-3p/CPEB3/p53 axis. CircKDM1A may be a potential prognostic biomarker and therapeutic target of BCa.

N-acetyltransferase 10 (NAT10), an enzyme responsible for ac4C acetylation, is implicated in cancer progression, though its specific biological function in prostate cancer remains insufficiently understood. This study clarifies NAT10's role in prostate cancer and its effects on the tumor immune microenvironment. NAT10 expression and clinical relevance were assessed through bioinformatics, RT-qPCR, and IHC analyses, comparing prostate cancer tissues with normal controls. The impact of NAT10 on tumor cell proliferation, migration, and invasion was investigated via in vitro assays-including CCK-8, EdU, wound healing, and 3D-Transwell-as well as in vivo mouse xenograft models and organoid studies. Further, NAT10's influence on immune cell infiltration was examined using flow cytometry, IHC, cell co-culture assays, and ELISA to elucidate downstream chemokine effects, specifically targeting CD8 T cells. Findings indicated significant upregulation of NAT10 in prostate cancer cells, enhancing their proliferative and invasive capacities. Notably, NAT10 suppresses CD8 T cell recruitment and cytotoxicity through the CCL25/CCR9 axis, fostering an immunosuppressive microenvironment that exacerbates tumor progression. An ac4C modification score was also devised based on NAT10's downstream targets, providing a novel predictive tool for evaluating immune infiltration and forecasting immunotherapy responses in patients with prostate cancer. This study underscores NAT10's pivotal role in modulating the prostate cancer immune microenvironment, offering insights into the immune desert phenomenon and identifying NAT10 as a promising therapeutic target for improving immunotherapy efficacy.

Acetyltransferase NAT10 promotes an immunosuppressive microenvironment by modulating CD8 T cell activity in prostate cancer.

Robot-assisted surgical techniques have enhanced surgical precision, control, stability, and vision, particularly in cardiac interventions. However, the relationship between hospital volume and the clinical and readmission outcomes of robot-assisted cardiac surgery remains undefined. This retrospective cohort study analyzed Nationwide Readmissions Database (NRD) data from 2010 to 2020. Patients who underwent various cardiac procedures were categorized into three groups based on hospital volume of robot-assisted cardiac surgery: low-volume centers (LVCs), medium-volume centers (MVCs), and high-volume centers (HVCs). Our primary outcome variables of interest were the incidence of perioperative complications, 30-day readmission, and in-hospital mortality. 10,543 patients were included in the analysis. Compared with those treated in LVCs, the adjusted OR of 30-day readmission [OR, 0.70; (95% CI 0.53-0.92), P = 0.014] was lower in HVCs. However, higher perioperative bleeding rates were observed in MVCs (OR, 1.38; (95% CI 1.14-1.66), P = 0.001) and HVCs (OR, 1.84; (95% CI 1.52-2.21), P = 0.001). No significant differences were found in in-hospital mortality. Our research demonstrated that higher hospital surgical volume was associated with better outcomes in robot-assisted cardiac surgery, despite higher perioperative bleeding rates.

Accurate differentiation of prostate cancer at various stages provides a reliable basis for clinical decision-making in cancer treatment. However, there is still no effective protocol to enable the rapid and accurate identification of cancer progression. With the aim to improve the diagnostic efficiency of prostate cancer at different stages, we systematically analyzed exosomes derived from prostate cancer at different stages of disease progression by integrating surface-enhanced Raman spectroscopy (SERS) with principal component analysis (PCA) in this work. Firstly, using a simple two-step electrodeposition strategy, we optimized deposition voltage and time to obtain a non-metallic SERS platform of g-CN@MXene with fascinating uniformity and stability. Particularly, this platform not only enhanced molecular enrichment by providing abundant adsorption sites, but also excited prominent SERS spectra of prostate cancer exosomes by promoting charge transfer within the heterostructure of g-CN and MXene. Furthermore, PCA was employed to conduct a detailed analysis of spectral features among the exosomes from different disease stages including early, intermediate, and late cancers. Specially, all these corresponding exosomes were accurately classified, which highlighted the potential of the SERS-PCA technique for exosome-based diagnostics and its utility in precision oncology.

Radiotherapy and brachytherapy are integral treatments for breast cancer, yet the influence of hospital characteristics on patient outcomes remains underexplored. This study evaluated the influence of hospital teaching status (teaching vs non-teaching) on radiation-related complications, in-hospital mortality, readmission rate and healthcare resource utilization (HCRU) in breast cancer patients receiving these therapies. We conducted a retrospective cohort study using the US Nationwide Readmissions Database (2016-2021), including female breast cancer patients aged ≥18 years who underwent radiotherapy or brachytherapy. Multivariable logistic regression models evaluated the impact of hospital teaching status (teaching vs non-teaching) on clinical outcomes. Among 9030 patients, 7177 (79.5%) were treated at teaching hospitals. These patients were younger, had a higher prevalence of metastatic disease, and were less likely to undergo breast-conserving surgery or mastectomy. In-hospital mortality was higher for radiotherapy than for brachytherapy (4.2% vs 0.3%), with no unadjusted difference by hospital type. Radiation-related complications were more frequent in non-teaching hospitals for both radiotherapy (OR: 1.11 [1.01, 1.21]). Radiotherapy readmission rates were lower at non-teaching hospitals at 30 (OR: 0.82 [0.70, 0.95]) and 90 days (OR: 0.84 [0.74, 0.95]), with reduced costs and shorter length of stay compared to patients receiving radiotherapy at teaching hospitals. Our study demonstrates that breast cancer patients undergoing radiotherapy at non-teaching hospitals experienced better prognosis, with lower readmission rates, reduced healthcare costs, and shorter lengths of stay compared to those treated at teaching hospitals. These findings highlight important differences in patient outcomes and healthcare resource utilization between teaching and non-teaching hospitals, emphasizing the need for further research to understand the underlying causes and optimize care delivery.

Hidden blood loss (HBL) is a critical but understudied component of perioperative blood loss in laparoscopic myomectomy, with limited data on its magnitude and associated factors.
