Tyloxapol &#x6291;&#x5236;&#x5154;&#x548C;&#x6B7B;&#x4EA1;&#x7387;&#x4E2D;&#x7EC6;&#x83CC;&#x5185;&#x6BD2;&#x7D20;&#x7684;&#x75C5;&#x7406;&#x5F71;&#x54CD;&#x4E4B;&#x540E;&#x7684;&#x76F2;&#x80A0;&#x7ED3;&#x624E;&#x548C;&#x7A7F;&#x523A;&#x5927;&#x9F20;&#x5185;&#x6BD2;&#x7D20;&#x53D7;&#x4F53;-&#x914D;&#x4F53;&#x76F8;&#x4E92;&#x4F5C;&#x7528;&#x7684;&#x5C01;&#x9501;&#x3002;

We have previously demonstrated that the detergent Tyloxapol is effective in preventing reactions to endotoxin. We studied the effects of Tyloxapol on the morbidity and mortality from endotoxemia in rabbits and on the mortality in rats with sepsis. The effects of Tyloxapol on endotoxin binding and macrophage activation were studied in the macrophage cell line RAW264.7 and CHO cells expressing CD14. Isolated human leukocytes were used to study the effects of Tyloxapol on immune reactions, leukocyte motility, and phagocytosis. Intravenous Tyloxapol (200 mg/kg), given prior to or at the time of endotoxin infusion protected rabbits from developing shock. In rats with peritoneal sepsis, a lipid-rich diet and Tyloxapol given at the time of induction of peritonitis protected them from septic death. In vitro, Tyloxapol blocked the binding of endotoxin to murine macrophages and CHO cells expressing CD14, activation of macrophages, and also some antigen-antibody immune reactions (mediated by CD2, CD4, CD22, HLA-DR). Tyloxapol may prevent the reaction to endotoxin by desensitizing endotoxin-recognizing receptors.

Tyloxapol attenuates the pathologic effects of endotoxin in rabbits and mortality following cecal ligation and puncture in rats by blockade of endotoxin receptor-ligand interactions.

Nous avons d&eacute;montr&eacute; pr&eacute;c&eacute;demment que le d&eacute;tergent Tyloxapol est efficace pour pr&eacute;venir les r&eacute;actions &agrave; l&#39;endotoxine. Tyloxapol peut emp&ecirc;cher la r&eacute;action &agrave; l&#39;endotoxine de d&eacute;sensibilisation des r&eacute;cepteurs reconnaissant d&#39;endotoxine.

Tyloxapol att&eacute;nue les effets pathologiques de l&#39;endotoxine chez le lapin et la mortalit&eacute; apr&egrave;s la ligature du caecum et ponction chez le rat par le blocage des interactions de r&eacute;cepteur-ligand d&#39;endotoxine.

Tyloxapol kann verhindern, dass die Reaktion auf Endotoxin desensibilisieren Endotoxin-erkennen-Rezeptoren.

Tyloxapol d&auml;mpft die pathologischen Auswirkungen von Endotoxin im Kaninchen und Sterblichkeit nach cecal Verbindung und Punktion bei Ratten durch Blockade von Endotoxin-Rezeptor-Ligand-Interaktionen.

In vitro, Tiloxapol bloqueado el atascamiento de la endotoxina a macr&oacute;fagos murinos y c&eacute;lulas CHO expresando CD14, activaci&oacute;n de los macr&oacute;fagos y tambi&eacute;n algunas reacciones inmunes ant&iacute;geno-anticuerpo (mediadas por CD4, CD22, CD2, HLA-DR). Tiloxapol puede prevenir la reacci&oacute;n a la endotoxina desensibilizaci&oacute;n de receptores de reconocimiento de endotoxina.

Peripheral neuropathies are reported to arise as a result of the systemic inflammatory response produced by a full-thickness cutaneous burn injury. This study was designed to characterize the magnitude and time course of functional and morphological changes in peripheral axons that arise after a full-thickness dermal burn injury in an animal model. A 20% body surface area (20% BSA) full-thickness dermal burn was applied to the back of C57BL6 female mice. Longitudinal H- and M-wave recordings were used to determine the conduction velocities (CV) of large myelinated motor and sensory axons in the tibial nerve of sham control and burn-injured mice. Motor CVs were significantly reduced from 6 h to 28 days after the burn, and sensory CVs were significantly reduced from 7 to 14 days after the burn. Morphological evaluation also showed that the mean caliber of large axons in tibial nerves and L5 ventral and dorsal roots in burned mice was significantly decreased. The results demonstrate that both functional and morphological deficits may be produced in peripheral nerve axons at sites well removed from a full-thickness dermal burn injury. The neural deficits may contribute to changes in neuromuscular transmission and the development of limb and respiratory muscle weakness that also accompany burn injury.

The inwardly rectifying potassium channel Kir4.1 is widely expressed by astrocytes throughout the brain. Kir4.1 channels are absent in immature, proliferating glial cells. The progressive expression of Kir4.1 correlates with astrocyte differentiation and is characterized by the establishment of a negative membrane potential (> -70 mV) and an exit from the cell cycle. Despite some correlative evidence, a mechanistic interdependence between Kir4.1 expression, membrane hyperpolarization, and control of cell proliferation has not been demonstrated. To address this question, we used astrocyte-derived tumors (glioma) that lack functional Kir4.1 channels, and generated two glioma cell lines that stably express either AcGFP-tagged Kir4.1 channels or AcGFP vectors only. Kir4.1 expression confers the same K+ conductance to glioma membranes and a similar responsiveness to changes in [K+]o that characterizes differentiated astrocytes. Kir4.1 expression was sufficient to move the resting potential of gliomas from -50 to -80 mV. Importantly, Kir4.1 expression impaired cell growth by shifting a significant number of cells from the G2/M phase into the quiescent G0/G1 stage of the cell cycle. Furthermore, these effects could be nullified entirely if Kir4.1 channels were either pharmacologically inhibited by 100 microM BaCl2 or if cells were chronically depolarized by 20 mM KCl to the membrane voltage of growth competent glioma cells. These studies therefore demonstrate directly that Kir4.1 causes a membrane hyperpolarization that is sufficient to account for the growth attenuation, which in turn induces cell maturation characterized by a shift of the cells from G2/M into G0/G1.

Role of Kir4.1 channels in growth control of glia.

Kir4.1 expression conf&egrave;re la m&ecirc;me conductance K + aux membranes de gliome et une r&eacute;activit&eacute; similaire &agrave; l&#39;&eacute;volution de [K +] o qui caract&eacute;rise diff&eacute;renci&eacute;e des astrocytes. Kir4.1 expression ne suffisait pas &agrave; d&eacute;placer le potentiel de repos des gliomes de -50 &agrave; -80 mV. El canal de potasio interiormente rectificaci&oacute;n Kir4.1 se expresa ampliamente por los astrocitos en el cerebro. Kir4.1 canales est&aacute;n ausentes en inmaduras, proliferaci&oacute;n de c&eacute;lulas gliales. La expresi&oacute;n progresiva de Kir4.1 se correlaciona con la diferenciaci&oacute;n de astrositos y se caracteriza por la creaci&oacute;n de un potencial de membrana negativo (&gt; -70 mV) y una salida del ciclo celular. A pesar de algunas pruebas correlativos, no se ha demostrado una mecanicista interdependencia entre la expresi&oacute;n Kir4.1, hiperpolarizaci&oacute;n de la membrana y el control de la proliferaci&oacute;n celular. Para abordar esta cuesti&oacute;n, se utilizan tumores derivados de Astrocito (glioma) que carecen de canales de Kir4.1 funcionales y generan dos l&iacute;neas de c&eacute;lulas de glioma que expresan estable Kir4.1 AcGFP-etiquetado canales o vectores de AcGFP solamente. Kir4.1 expresi&oacute;n confiere la misma conductancia del K + a membranas de glioma y una respuesta similar a los cambios en la [K +] o que caracteriza a distinguido astrocitos. Kir4.1 expresi&oacute;n era suficiente para mover el potencial de reposo de los gliomas de -50 a -80 mV. Importante, Kir4.1 expresi&oacute;n deteriorada crecimiento celular cambiando un n&uacute;mero significativo de c&eacute;lulas de la fase G2/M en la etapa quieta G0/G1 del ciclo celular. Adem&aacute;s, estos efectos podr&iacute;an anularse totalmente si Kir4.1 canales farmacol&oacute;gicamente tampoco fueron inhibidos por 100 microM BaCl2 o si las c&eacute;lulas fueron cr&oacute;nicamente depolarized 20 mm KCl a la tensi&oacute;n de la membrana de las c&eacute;lulas de glioma competente de crecimiento. Estos estudios muestran por lo tanto directamente que Kir4.1 causa una hiperpolarizaci&oacute;n de la membrana que es suficiente para tener en cuenta para la atenuaci&oacute;n del crecimiento, que a su vez induce la maduraci&oacute;n celular caracterizada por un cambio de las c&eacute;lulas de G2/M en G0/G1.

The systemic inflammatory response produced by a full-thickness dermal burn injury is associated with a peripheral motor neuropathy. We previously reported that a 20% body surface area (BSA) full-thickness dermal burn in C57BL6 mice produced structural and functional deficits in motor axons at a distance from the burn site. The etiology of the neuropathy, however, is not well characterized. Burn injury leads to an increase in production of a number of proinflammatory mediators, including nitric oxide (NO). We tested the hypothesis that dermal burn-induced motor neuropathy is mediated by increased production of NO. NO synthase (NOS) activity was inhibited following a 20% BSA full-thickness burn by injection of non-specific NOS inhibitor, nitro-L-arginine methyl ester or inducible NOS (iNOS) inhibitors, L-N6-(1-iminoethyl) lysine, and aminoguanidine. NOS inhibitors also prevented the reduction in ventral roots mean axon caliber and the decrease in a motor nerve conduction velocity (MCV) following burn. iNOS knockout mice prevented MCV decrease in the first 3 days post-burn, but iNOS knockout MCV was significantly reduced at 7-14 days post-burn. These results suggest that an increase in NO production generated by systemic inflammatory response pathways after burn injury contributes to the development of structural and functional deficits in peripheral motor axons.

Epoxyeicosatrienoic acids (EETs), synthesized and released by astrocytes in response to glutamate, are known to play a pivotal role in neurovascular coupling. In vascular smooth muscle cells (VSMC), EETs activate large-conductance, Ca(2+)-activated K(+) (BK) channels resulting in hyperpolarization and vasodilation. However, the functional role and mechanism of action for glial-derived EETs are still to be determined. In this study, we evaluated the effect of the synthetic EET analog 11-nonyloxy-undec-8(Z)-enoic acid (NUD-GA) on outward K(+) currents mediated by calcium-activated K(+) channels. Addition of NUD-GA significantly increased intracellular Ca(2+) and outward K(+) currents in perivascular astrocytes. NUD-GA-induced currents were significantly inhibited by BK channel blockers paxilline and tetraethylammonium (TEA) (23.4 Â± 2.4%; P < 0.0005). Similarly, NUD-GA-induced currents were also significantly inhibited in the presence of the small-conductance Ca(2+)-activated K(+) channel inhibitor apamin along with a combination of blockers against glutamate receptors (12.8 Â± 2.70%; P < 0.05). No changes in outward currents were observed in the presence of the channel blocker for intermediate-conductance K(+) channels TRAM-34. Blockade of the endogenous production of EETs with N-methylsulfonyl-6-(2-propargyloxyphenyl)hexanamide (MS-PPOH) significantly blunted (dl)-1-aminocyclopentane-trans-1,3-dicarboxylic acid (t-ACPD)-induced outward K(+) currents (P < 0.05; n = 6). Both NUD-GA and t-ACPD significantly increased BK channel single open probability; the later was blocked following MS-PPOH incubation. Our data supports the idea that EETs are potent K(+) channel modulators in cortical perivascular astrocytes and further suggest that these metabolites may participate in NVC by modulating the levels of K(+) released at the gliovascular space.

Role of epoxyeicosatrienoic acids as autocrine metabolites in glutamate-mediated K+ signaling in perivascular astrocytes.

Les acides &eacute;poxy&eacute;icosatri&eacute;no&iuml;ques (EETs), synth&eacute;tis&eacute;e et lib&eacute;r&eacute;e par les astrocytes en r&eacute;ponse au glutamate, sont connus pour jouer un r&ocirc;le crucial dans le couplage neurovasculaire. Dans les cellules du muscle lisse vasculaire (CMLV), EETs activent de grande conductance, canaux activ&eacute;s par Ca(2+) k (BK) ce qui entra&icirc;ne une hyperpolarisation et une vasodilatation. Cependant, le r&ocirc;le fonctionnel et le m&eacute;canisme d&#39;action d&#39;EETs gliales d&eacute;riv&eacute;s sont encore &agrave; d&eacute;terminer. Ebenso waren hat-GA-induzierte Str&ouml;me auch erheblich gehemmt in Anwesenheit der klein-Leitwert K(+) Ca(2+) aktivierte Kanal Inhibitor Apamin zusammen mit einer Kombination von Blocker gegen Glutamat-Rezeptoren (12,8 &plusmn; 2.70 %; P &lt; 0,05). Acidi Epoxyeicosatrienoic (EETs), sintetizzato e rilasciato da astrociti in risposta al glutammato, sono noti per giocare un ruolo fondamentale nell&#39;accoppiamento neurovascolare. Nelle cellule muscolari lisce vascolari (VSMC), EETs attivare grandi-conduttanza, canali attivati da Ca(2+) K(+) (BK) con conseguente hyperpolarization e vasodilatazione. Tuttavia, il ruolo funzionale e meccanismo d&#39;azione per EETs gliali derivati sono ancora da stabilirsi. &#xC870;&#xBBF8;&#xB8CC; &#xC911;&#xC7AC; K + perivascular &#xC774;&#xB2E4;&#xC5D0;&#xC11C; &#xC2E0;&#xD638;&#xC5D0; autocrine &#xB300;&#xC0AC; &#xC0B0;&#xBB3C;&#xC73C;&#xB85C; epoxyeicosatrienoic &#xC0B0;&#xC758; &#xC5ED;&#xD560;.

Epoxyeicosatrienoic &aacute;cidos (EETs), sintetizado y liberado por los astrocitos en respuesta al glutamato, se saben que desempe&ntilde;an un papel fundamental en el acoplamiento neurovascular. En c&eacute;lulas de m&uacute;sculo liso vascular (VSMC), EETs activan grande-conductancia, canales de k Ca(2+)-activado (BK) dando por resultado la hiperpolarizaci&oacute;n y vasodilataci&oacute;n. Sin embargo, el papel funcional y mecanismo de acci&oacute;n de origen glial EETs est&aacute;n todav&iacute;a por determinar. En este estudio, se evalu&oacute; el efecto de la EET sint&eacute;tico an&aacute;logo 11-nonyloxy-undec-8 (Z)-&aacute;cido enoic (NUD-GA) en corrientes de capacidad exterior mediada por calcio activado k canales. Adici&oacute;n de NUD-GA aument&oacute; significativamente CA intracelular y corrientes de k hacia fuera en los astrocitos perivasculares. Corrientes NUD-GA-inducida fueron significativamente inhibidas por paxilline de los bloqueadores de los canales BK y Tetraetilamonio (TEA) (23.4 &plusmn; 2.4%; P &lt; 0.0005). Asimismo, corrientes NUD-GA-inducida fueron tambi&eacute;n significativamente inhibidas en presencia del peque&ntilde;o-conductancia activados por Ca(2+) k canal inhibidor apamin junto con una combinaci&oacute;n de bloqueadores de los receptores de glutamato (12,8 &plusmn; 2.70%; P &lt; 0.05). Cambios en las corrientes hacia fuera no se observaron en presencia del bloqueador de los canales para los canales de k intermedio-conductancia tranv&iacute;a-34. Bloqueo de la producci&oacute;n end&oacute;gena de EETs con N-methylsulfonyl - 6-(2-propargyloxyphenyl) hexanamide (MS-PPOH) blunted significativamente (&aacute;cido dl)-1-aminocyclopentane-trans-1,3-dicarboxylic (t-ACPD)-inducida hacia fuera k corrientes (P &lt; 0.05; n = 6). Tanto NUD-GA y t-CONTINU&Eacute; aumentaron significativamente la probabilidad de abrir solo canal BK; m&aacute;s adelante fue bloqueado a ra&iacute;z de la incubaci&oacute;n de MS-PPOH. Nuestros datos apoyan la idea que EETs son moduladores de canal k potentes en astrocitos perivasculares cortical y m&aacute;s sugieren que estos metabolitos pueden participar en NVC modulando los niveles de k en el espacio gliovascular.
