[Therapeutic effect of enhancer of Zeste homolog 2 inhibitor GSK343 on periodontitis by regulating macrophage differentiation].

Oral squamous cell carcinoma (OSCC) is an aggressive malignant tumor. Bupivacaine (Bupi), a local anesthetic drug, has been shown to display anti-tumor activity against a variety of tumors.

Bupivacaine inhibits the malignant biological behavior of oral squamous cell carcinoma cells by inhibiting the activation of ERK1/2 and STAT3.

A radicular groove is an anatomic malformation that usually initiates at the central fossa, extending along the root at varying lengths and depths and predisposes the involved tooth to a severe periodontal defect. Severe grooves that extend to the root apex often lead to complex combined periodontal-endodontic lesions. They are a serious challenge for doctors to diagnose and treat.

Intentional replantation combined root resection therapy for the treatment of type III radicular groove with two roots: A case report.

The development of a versatile antibacterial coating, irrespective of material characteristics, is greatly attractive but still a challenge. In this work, mussel-inspired dopamine-modified sodium alginate (SA-DA) was successfully synthesized as the adhesion layer, and antibacterial coatings on three types of substrates, namely cotton fabric, aluminum sheet, and polyurethane membrane, were constructed through the layer-by-layer (LbL) deposition of polyhexamethylene guanidine and sodium alginate. Among the coated materials, the coated cotton fabric was systematically characterized, and the results showed that it still exhibited ideal hydrophilicity, and its liquid absorption capacity increased with an increase in the coating layers. The growth of  and  was notably inhibited on the coated cotton fabric, and 10 coating bilayers achieved 100% inhibition of bacterial growth within 10 min. Furthermore, an ideal antibacterial ability maintained after 10 cycles of antibacterial trials or 50 washing or soaping cycles.  evaluation of the hemostatic effect indicated that the coated cotton fabric could promote blood clotting by concentrating the components of blood and activating the platelets, and no significant hemolysis and cytotoxicity were observed in the coated cotton fabric. Moreover, the coated aluminum and polyurethane film also displayed an obvious antibacterial effect, which proved that the constructed coating could successfully adhere to the metal and polymer surfaces. Therefore, this work provided a proper way for the progress of a current antibacterial coating tactics for different substrate surfaces.

A universal coating strategy for inhibiting the growth of bacteria on materials surfaces.

Disruption of the symbiosis of extra/intratumoral metabolism is a good strategy for treating tumors that shuttle resources from the tumor microenvironment. Here, we report a precision treatment strategy for enhancing pyruvic acid and intratumoral acidosis to destroy tumoral metabolic symbiosis to eliminate tumors; this approach is based on PEGylated gold and lactate oxidase-modified aminated dendritic mesoporous silica with lonidamine and ferrous sulfide loading (PEG-Au@DMSNs/FeS/LND@LOX). In the tumor microenvironment, LOX oxidizes lactic acid to produce pyruvate, which represses tumor cell proliferation by inhibiting histone gene expression and induces ferroptosis by partial histone monoubiquitination. In acidic tumor conditions, the nanoparticles release HS gas and Fe ions, which can inhibit catalase activity to promote the Fenton reaction of Fe, resulting in massive ·OH production and ferroptosis via Fe. More interestingly, the combination of HS and LND (a monocarboxylic acid transporter inhibitor) can cause intracellular acidosis by lactate, and protons overaccumulate in cells. Multiple intracellular acidosis is caused by lactate-pyruvate axis disorders. Moreover, HS provides motive power to intensify the shuttling of nanoparticles in the tumor region. The findings confirm that this nanomedicine system can enable precise antitumor effects by disrupting extra/intratumoral metabolic symbiosis and inducing ferroptosis and represents a promising active drug delivery system candidate for tumor treatment.

HS-Powered Nanomotors for Active Therapy of Tumors by Inducing Ferroptosis and Lactate-Pyruvate Axis Disorders.

The disruptions caused by ice crystal formation during the cryopreservation of cells and tissues can cause cell and tissue damage. Thus, preventing such damage during cryopreservation is an important but challenging goal. Here, a hibernating/awakening nanomotor with magnesium/palladium covering one side of a silica platform (Mg@Pd@SiO) is proposed. This nanomotor is used in the cultivation of live NCM460 cells to demonstrate a new method to actively limit ice crystal formation and enable highly efficient cryopreservation. Cooling Mg@Pd@SiO in solution releases Mg/H and promotes the adsorption of H at multiple Pd binding sites on the cell surface to inhibit ice crystal formation and cell/tissue damage; additionally, the Pd adsorbs and stores H to form a hibernating nanomotor. During laser-mediated heating, the hibernating nanomotor is activated (awakened) and releases H, which further suppresses recrystallization and decreases cell/tissue damage. These hibernating/awakening nanomotors have great potential for promoting highly efficient cryopreservation by inhibiting ice crystal formation.

Hibernating/Awakening Nanomotors Promote Highly Efficient Cryopreservation by Limiting Ice Crystals.

Oral aphthous ulcers are common mucosal lesions that cause pain and discomfort. There are diverse biomaterials and drug treatments for oral ulcers used in both research and clinical settings. However, the complex oral environment often results in low adhesion and short drug retention times, which lead to poor drug availability and treatment outcomes. In this study, a mussel-inspired adhesive hydrogel was developed by grafting catechol onto hyaluronic acid (C-HA), and dopamine was added for oxidative pre-polymerization to form modified hyaluronic acid (M-HA), which remarkably increased the adhesion of the hydrogels. Then, M-HA was interpenetrated into the gelatin methacryloyl (GelMA) network. Chlorhexidine gluconate (CHG) was then incorporated into the hydrogel to enhance its availability and therapeutic effect through its sustained-release capability. The GelMA/M-HA hydrogel demonstrated strong adhesion to wet tissues, antibacterial and anti-inflammatory properties, and good biocompatibility. In both rat oral ulcers and infected wounds, the adhesive hydrogel significantly accelerated the healing of the ulcers and infected wounds. These results indicated that this adhesive hydrogel offers a promising new strategy for the treatment of oral ulcers in clinical practice. STATEMENT OF SIGNIFICANCE: Oral ulcers are a common and high-incidence mucosal condition that seriously affect people's daily lives, often making it difficult for patients to chew and speak. However, a dynamic oral environment with various types of bacteria influences drug availability and treatment effects in clinical settings. To address this challenge, an adhesive, mussel-inspired, drug-loaded hydrogel was constructed using natural macromolecules (hyaluronic acid and gelatin) with good biocompatibility. Chlorhexidine gluconate (CHG), with its broad-spectrum antibacterial activity, has been incorporated to synergistically promote oral ulcer healing. The splendid adhesion, antibacterial, and therapeutic effects of this hydrogel demonstrated a new strategy for treating oral ulcers.

Mussel-inspired adhesive drug-loaded hydrogels for oral ulcers treatment.

Oral health problems, particularly tooth defects, can significantly affect people's quality of life and overall well-being. The development of titanium (Ti) dental implants has largely replaced natural tooth roots to prevent periodontal and gastrointestinal diseases. However, challenges such as postoperative bacterial infections and poor osseointegration continue to hinder progress in dental implant technology. To tackle these issues, we used hydroxypropyl trimethylammonium chloride chitosan (HACC) and gallic acid-modified gelatin (GAG) to create extracellular matrix (ECM) coatings on titanium using layer-by-layer self-assembly. GAG showed better water solubility at room temperature, being over 99.0 times more soluble than regular gelatin.  and  analyses of the ECM coatings revealed their antibacterial properties and their ability to promote osteogenic differentiation, resulting in over 31.5 times more calcareous deposits than Ti. This strategy shows potential for improving oral health and reducing the complications associated with dental implants in clinical settings.

Antibacterial and Osteogenesis Promotion of Bionic Extracellular Matrix Implant Coating Based on Gallic Acid Self-Assembly.

Head and Neck Squamous Cell Carcinoma (HNSCC) is an aggressive malignancy with poor prognosis and low five-year survival rates. Ferroptosis, an iron-dependent form of cell death, has emerged as a potential factor in cancer therapy. This study aimed to identify differentially expressed genes (DEGs) associated with HNSCC, ferroptosis, and radiotherapy, and to develop a diagnostic model to establish novel biomarkers, early diagnosis and individualized therapy.

Ferroptosis and radiotherapy-related genes in head and neck squamous cell carcinoma: diagnostic and prognostic significance.

Acute kidney injury (AKI) is characterized by a rapid loss of renal metabolic function and a high mortality rate. Although significant progress has been made in developing targeted drugs for AKI treatment, issues such as inadequate antioxidant effects and poor renal enrichment efficiency remain. Nanomotors can enhance drug delivery efficiency in AKI treatments through self-propulsion in the microenvironment or via external stimuli. We reviewed recent progress in the targeted treatment of AKI with nanomotors, focusing on their contribution to targeted drug delivery at different stages and combined treatments. Current limitations and future development directions are also discussed.
