Increasing reports demonstrate that long noncoding RNAs participate in the regulation of numerous malignancies, cervical cancer included. Although lncRNA LOXL1 antisense RNA 1 has been commonly accepted to be an oncogene in many cancers. Here, the role of LOXL1-AS1 in CC still need to be explored. In this study, LOXL1-AS1 was found elevated in CC tissues and cells. LOXL1-AS1 depletion restrained CC cell proliferation, migration, invasion, and angiogenesis in vivo. Furthermore, we found that LOXL1-AS1 upregulated Lysophospholipase 1 expression via sequestering miR-526b-5p. Rescue assays revealed that overexpression of LYPLA1 reversed the LOXL1-AS1 silencing-induced inhibitory effects on the malignant phenotypes of CC cells. To conclude, this study showed that LOXL1-AS1 facilitates cellular process in CC via functioning as a miR-526b-5p sponge.

Cervical cancer (CC), as a serious menace to the health of women, has long been one of the most lethal gynecologic neoplasms throughout the world. Long non-coding RNA (LncRNA)  has been documented to exert crucial functions in many malignant tumors. Nonetheless, the function and molecular mechanism of  in CC remain completely unknown.

Delayed childbearing leads to increased assisted reproductive technology use by women of advanced maternal age (AMA). It is unclear whether fresh or frozen embryo transfer (FET) is the better option. We aimed to assess maternal and neonatal outcomes in patients having their first FET after a freeze-all cycle versus those having their first fresh embryo transfer (ET). We reviewed 720 women of AMA undergoing a first fresh ET (n = 375) or FET (n = 345) between January 2016 and April 2021. No significant difference in the live birth rate was found between FET and fresh ET (19.7% vs. 24.3%,  = 0.141). The clinical pregnancy rate was significantly lower in the FET group than in the fresh ET group (26.4 % (91/345) vs. 33.6% (126/375),  = 0.035), but FET resulted in higher birthweights (3217.16 ± 734.44 vs. 3003.37 ± 635.00,  = 0.037) and was associated with a lower incidence of preterm births (2.6% vs. 5.6%,  = 0.046). The risks of other maternal and neonatal outcomes did not differ significantly between the groups. Among women of AMA, the transfer of frozen embryos did not result in significantly higher rates of live birth than fresh embryos did; however, a freeze-all strategy may not be beneficial for the women of AMA.

Ovarian cancer (OC) is a common malignant tumor in females with high recurrence and poor prognosis. Cisplatin is commonly used for OC clinical treatment, but its efficacy is usually challenged by the chemotherapy resistance of cancer cells. MicroRNAs (miRNAs), including miR-30a-5p, were identified to modulate drug resistance in numerous tumors. However, molecular mechanisms of miR-30a-5p in OC chemoresistance need more illumination.

Pelvic masses frequently originate from the pelvic cavity and are often associated with uterine, ovarian, or intestinal disorders. This report describes the case of a patient with a pelvic mass diagnosed as a retroperitoneal dermoid cyst at our hospital. We analyzed this case and conducted a literature review, to mitigate the risk of misdiagnosis and enhance the treatment of retroperitoneal masses.

Impact of surgical compliance on survival prognosis of patients with ovarian cancer and associated influencing factors: A propensity score matching analysis of the SEER database.

Our study aimed to investigate the expression characteristics of the homeobox B13 (HOXB13) gene in placental tissues of gestational diabetes mellitus (GDM) and its impact on placental metabolic function. Placental tissues from 30 GDM patients and normal pregnancy cases were collected. The expression of HOX family genes (including HOXB13 and HOXA2 among seven genes) was screened using quantitative polymerase chain reaction, and the protein expression of HOXB13 was validated by Western blot and immunohistochemistry. Untargeted metabolomics analysis using ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry was performed to examine metabolic differences in placental tissues, and Kyoto Encyclopedia of Genes and Genomes pathway enrichment was applied to interpret metabolic regulation networks. The results showed that the mRNA and protein expression of HOXB13 were significantly lower in the GDM placentas compared to the normal pregnancy group. A total of 337 differential metabolites were identified, with the highest proportion being lipids and lipid-like molecules (37.96%). The levels of glucose and galactose in the GDM group were significantly reduced. Kyoto Encyclopedia of Genes and Genomes pathway analysis showed that these differential metabolites were enriched in pathways related to steroid hormone biosynthesis, retinol metabolism, and arachidonic acid metabolism. We conclude that HOXB13 is downregulated in GDM placentas, and its expression is closely associated with dysregulated glucose-lipid metabolism and abnormalities in key metabolic pathways. HOXB13 may serve as a potential biomarker and therapeutic target for GDM. Notre étude visait à étudier les caractéristiques d'expression du gène homéobox B13 (HOXB13) dans les tissus placentaires de patients atteints de diabète gestationnel (DG) et son impact sur la fonction métabolique placentaire. Des tissus placentaires de 30 patientes atteintes de DG et de cas de grossesse normale ont été prélevés. L'expression des gènes de la famille HOX (dont HOXB13 et HOXA2 parmi sept gènes) a été criblée par réaction en chaîne par polymérase quantitative, et l'expression protéique de HOXB13 a été validée par Western blot et immunohistochimie. Une analyse métabolomique non ciblée par chromatographie liquide à ultra haute performance couplée à une spectrométrie de masse à temps de vol quadripolaire a été réalisée afin d'examiner les différences métaboliques dans les tissus placentaires. L'enrichissement des voies de signalisation de l'Encyclopédie de Kyoto des gènes et des génomes a été appliqué pour interpréter les réseaux de régulation métabolique. Les résultats ont montré que l'expression de l'ARNm et des protéines de HOXB13 était significativement plus faible dans les placentas atteints de DG que dans le groupe de grossesse normale. Au total, 337 métabolites différentiels ont été identifiés, la proportion la plus élevée étant des lipides et des molécules de type lipidique (37,96 %). Les taux de glucose et de galactose dans le groupe GDM ont été significativement réduits. L'analyse des voies de signalisation de l'Encyclopédie des gènes et génomes de Kyoto a montré que ces métabolites différentiels étaient enrichis dans les voies liées à la biosynthèse des hormones stéroïdes, au métabolisme du rétinol et au métabolisme de l'acide arachidonique. Nous concluons que HOXB13 est sous-régulé dans les placentas du GDM, et que son expression est étroitement associée à un métabolisme glucose-lipides dérégulé et à des anomalies des principales voies métaboliques. HOXB13 pourrait servir de biomarqueur potentiel et de cible thérapeutique pour le GDM.

Expression of homeobox B13 gene and metabolomics study in placental tissues of gestational diabetes mellitus.

Transferrin receptor (TFRC) is essential for iron uptake and may regulate ferroptosis, a form of iron-dependent cell death implicated in cervical cancer (CC). Bioinformatic analyses (GEPIA, UALCAN, SRAMP, starBase) were combined with in vitro and in vivo experiments. CC cell lines were transfected with shRNAs or overexpression plasmids targeting TFRC and LOXL1-AS1. Proliferation was assessed by colony formation, EdU staining, and Ki-67 immunostaining. Ferroptosis was evaluated by measuring malondialdehyde (MDA), lipid ROS, Fe⁺, and ferroptosis-related proteins. RNA pull-down, RIP, and MeRIP assays were used to explore m6A-dependent regulation, and actinomycin D assays assessed mRNA stability. TFRC and LOXL1-AS1 were upregulated in CC and associated with poor prognosis. TFRC promoted CC cell proliferation and inhibited ferroptosis. LOXL1-AS1 positively regulated TFRC by stabilizing its mRNA via an m6A-IGF2BP2-dependent mechanism. Rescue experiments confirmed that TFRC overexpression reversed the effects of LOXL1-AS1 knockdown. In vivo, LOXL1-AS1 depletion suppressed tumor growth and enhanced ferroptosis. LOXL1-AS1 promoted CC progression by stabilizing TFRC mRNA through m6A-IGF2BP2 interaction, suppressing ferroptosis. Targeting the LOXL1-AS1/IGF2BP2/TFRC axis may offer a potential therapeutic strategy for CC.
