Benzidine, a known carcinogen, is closely associated with the development of bladder cancer (BC). Epithelial-mesenchymal transition (EMT) is a critical pathophysiological process in BC progression. The underlying molecular mechanisms of mitogen-activated protein kinase (MAPK) pathway, especially extracellular regulated protein kinases 5 (ERK5), in regulating benzidine-induced EMT remains unclarified. Hence, two human bladder cell lines, T24 and EJ, were utilized in our study. Briefly, cell migration was assessed by wound healing assay, and cell invasion was determined by Transwell assay. Quantitative PCR and western blot were utilized to determine both gene expressions as well as protein levels of EMT and MAPK, respectively. Small interfering RNA (siRNA) was transfected to further determine ERK5 function. As a result, the migration and invasion abilities were enhanced, epithelial marker expression was decreased while mesenchymal marker expression was increased in human BC cell lines. Meanwhile, benzidine administration led to activation of ERK5 and activator protein 1 (AP-1) proteins, without effective stimulation of the Jun N-terminal kinase (JNK) or p38 pathways. Moreover, Benzidine-induced EMT and ERK5 activation were completely suppressed by XMD8-92 and siRNAs specific to ERK5. Of note, ERK1/2 was activated in benzidine-treated T24 cells, while benzidine-induced EMT could not be reversed by U0126, an ERK1/2 inhibitor, as indicated by further study. Collectively, our findings revealed that ERK5-mediated EMT was critically involved in benzidine-correlated BC progression, indicating the therapeutic significance of ERK5 in benzidine-related BC.

Benzidine Induces Epithelial-Mesenchymal Transition of Human Bladder Cancer Cells through Activation of ERK5 Pathway.

The emerging infectious disease, coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), poses a serious threat in China and worldwide. Challenged by this serious situation, China has taken many measures to contain its transmission. This study aims to systematically review and record these special and effective practices, in hope of benefiting for fighting against the ongoing worldwide pandemic.

China's practice to prevent and control COVID-19 in the context of large population movement.

2020 was an unprecedented year, with rapid and drastic changes in human mobility due to the COVID-19 pandemic. To understand the variation in commuting patterns among the Chinese population across stable and unstable periods, we used nationwide mobility data from 318 million mobile phone users in China to examine the extreme fluctuations of population movements in 2020, ranging from the Lunar New Year travel season (), to the exceptional calm of COVID-19 lockdown, and then to the recovery period. We observed that cross-city movements, which increased substantially in  and then dropped sharply during the lockdown, are primarily dependent on travel distance and the socio-economic development of cities. Following the Lunar New Year holiday, national mobility remained low until mid-February, and COVID-19 interventions delayed more than 72.89 million people returning to large cities. Mobility network analysis revealed clusters of highly connected cities, conforming to the social-economic division of urban agglomerations in China. While the mass migration back to large cities was delayed, smaller cities connected more densely to form new clusters. During the recovery period after travel restrictions were lifted, the netflows of over 55% city pairs reversed in direction compared to before the lockdown. These findings offer the most comprehensive picture of Chinese mobility at fine resolution across various scenarios in China and are of critical importance for decision making regarding future public-health-emergency response, transportation planning and regional economic development, among others.

Mobility in China, 2020: a tale of four phases.

Although many therapeutic interventions have shown promise in treating spinal cord injury, focusing on a single aspect of repair cannot achieve successful and functional regeneration in patients following spinal cord injury . In this study, we applied a combinatorial approach for treating spinal cord injury involving neuroprotection and rehabilitation, exploiting cell transplantation and functional sensorimotor training to promote nerve regeneration and functional recovery. Here, we used a mouse model of thoracic contusive spinal cord injury to investigate whether the combination of bone marrow mesenchymal stem cell transplantation and exercise training has a synergistic effect on functional restoration. Locomotor function was evaluated by the Basso Mouse Scale, horizontal ladder test, and footprint analysis. Magnetic resonance imaging, histological examination, transmission electron microscopy observation, immunofluorescence staining, and western blotting were performed 8 weeks after spinal cord injury to further explore the potential mechanism behind the synergistic repair effect. In vivo, the combination of bone marrow mesenchymal stem cell transplantation and exercise showed a better therapeutic effect on motor function than the single treatments. Further investigations revealed that the combination of bone marrow mesenchymal stem cell transplantation and exercise markedly reduced fibrotic scar tissue, protected neurons, and promoted axon and myelin protection. Additionally, the synergistic effects of bone marrow mesenchymal stem cell transplantation and exercise on spinal cord injury recovery occurred via the PI3K/AKT/mTOR pathway. In vitro, experimental evidence from the PC12 cell line and primary cortical neuron culture also demonstrated that blocking of the PI3K/AKT/mTOR pathway would aggravate neuronal damage. Thus, bone marrow mesenchymal stem cell transplantation combined with exercise training can effectively restore motor function after spinal cord injury by activating the PI3K/AKT/mTOR pathway.

During the outbreak of COVID-19 in China, the travels of the population from Wuhan have furthered the spread of the virus as the period coincided with the world's largest population movement to celebrate the Chinese New Year.

Mobile Phone-Based Population Flow Data for the COVID-19 Outbreak in Mainland China.

Green innovation is imperative for the high-quality and sustainable development of the modern logistics industry. It is also key for achieving the goals of peak emissions and carbon neutrality. This study provides a way of thinking about the evaluation of the green innovation level of the logistics industry. The variance inflation factor-variance coefficient method was employed to construct an evaluation index system of the regional logistics green innovation level (RLGIL) from three dimensions. Empirical data were collected from statistical yearbooks covering 30 provinces in China from 2013 to 2017. Thereafter, the combination weighting-based GRA-TOPSIS method was applied to evaluate the RLGIL, and the spatial distribution differences and spatiotemporal evolution characteristics of inter-provincial green innovation levels were analyzed. The RLGILs in the 30 provinces were found to be generally unbalanced, and the differences between the eastern and western regions were significant. Guangdong, Jiangsu, and Zhejiang had stronger RLGILs, whereas most other provinces did not reach the average level. The RLGIL of the 30 provinces had a high positive spatial correlation and spatial aggregating effect. From a national perspective, the values for the RLGIL were generally higher in the eastern and southern regions and lower in the western and northern regions. Although significant differences were found in the RLGIL of these provinces, the overall development trend was stable.

Green Innovation in Regional Logistics: Level Evaluation and Spatial Analysis.

Respiratory syncytial virus (RSV) is the leading global cause of respiratory infections and is responsible for about 3 million hospitalizations and more than 100,000 deaths annually in children younger than 5 years, representing a major global healthcare burden. There is a great unmet need for new agents and universal strategies to prevent RSV infections in early life. A multidisciplinary consensus development group comprising experts in epidemiology, infectious diseases, respiratory medicine, and methodology aims to develop the current consensus to address clinical issues of RSV infections in children.

To trace the history of human-cat interactions and the arrival of domestic cats (Felis catus) in East Asia, we analyzed 22 small felid bones excavated from 14 archaeological sites across China spanning 5,000 years. Genomic and radiocarbon evidence revealed that commensal leopard cats (Prionailurus bengalensis) appeared in anthropogenic environments at least 5,400 years ago and persisted until 150 CE. After a gap of several centuries, the earliest known domestic cat in China (c. 730 CE), reconstructed as a fully or partially white cat, was identified in Shaanxi during the Tang Dynasty. Genomic analysis combining 130 modern and ancient Eurasian cat specimens suggested an origin of Chinese domestic cats from the Levant and a likely merchant-mediated dispersal via the Silk Road. Commensal leopard cats and domestic cats once independently inhabited ancient human settlements in China but followed divergent sociocultural paths with only domestic cats becoming fully domesticated and globally introduced.

Recent research has identified MdCoL, which encodes a putative 2-oxoglutarate (2OG)- and Fe(II)-dependent oxygenase, as a key regulator of the columnar growth habit in apple. However, its role in drought stress responses remains largely unexplored. In this study, shoot cultures of columnar apple showed increased drought tolerance compared to standard apple varieties when subjected to PEG-induced osmotic stress. MdCoL expression was significantly upregulated during PEG treatment. Ectopic expression of MdCoL in apple calli and transgenic poplar plants conferred increased drought tolerance. Physiological and biochemical assessments revealed that MdCoL-overexpressing lines maintained higher photosynthetic activity and more efficient reactive oxygen species (ROS) scavenging under drought conditions than wild-type (WT) counterparts. Moreover, MdCoL enhanced photosynthetic electron transport efficiency in poplar through interaction with PagFd1. Transgenic lines also developed epidermal surfaces with increased glandular trichome density. Leaf ultrastructure analysis showed pronounced leaf blades and palisade tissue thickening in transgenic plants relative to WT. Furthermore, MdCoL contributed to improved water retention and regulated stomatal behavior by modulating abscisic acid (ABA) levels. These findings underscore the role of MdCoL in increasing drought tolerance and highlight its potential utility in improving stress tolerance in woody plants.
