Nav1.5 channel is crucial for the proliferation and migration of breast cancer cells. In this study, we investigated the anticancer effect of JZTX-14, a natural peptide considered an effective antagonist of Nav1.5. First, we successfully isolated and purified the 31 amino acid peptide JZTX-14 containing three pairs of disulfide bonds from spider venom and synthesised JZTX-14 by solid phase synthesis. We then predicted their physiochemical properties and structures in the peptide database. Further, we investigated the effects of natural and synthetic JZTX-14 on the proliferation and migration of MDA-MB-231 breast cancer cells  modulation of sodium current through the Nav1.5 channel. The results showed that both synthetic and natural JZTX-14 inhibited Nav1.5 currents, indicating the successful synthesis of JZTX-14. However, JZTX-14 did not affect MDA-MB-231 cell proliferation but inhibited its migration. Transcriptome analysis revealed that JZTX-14 downregulated S100A4 and FBXO2 and upregulated SERPINB2 in MDA-MB-231 cells. Western blot analysis demonstrated an increased level of the epithelial marker, E-cadherin, and decreased levels of the mesenchymal markers, N-cadherin and vimentin, and matrix metalloproteinase (MMP2), indicating the possible underlying mechanism of the inhibition of MDA-MB-231 cell migration by JZTX-14. This study provides a new target for inhibiting breast cancer metastasis and identifies a potent natural peptide for treating Triple-negative breast cancer.

Heat shock factors (Hsfs) play a crucial role in plant defense processes. However, the distribution and functional characteristics of Hsf genes in the relict plant  are still unclear. In this study, a total of 19  were identified and divided into three separate subgroups, comprising 10 , 7 , and 2  genes, respectively, based on their phylogenetic tree and the presence/absence of conserved protein domains. Whole-genome duplication and segmental duplication led to an expansion of the  gene family. The promoters of  genes are enriched for different types of -acting elements, including hormone responsive and abiotic-stress-responsive elements. The expression of , , , , and  increased significantly as a result of both cold and drought treatments. , , and  act as important genes whose expression levels correlate strongly with the expression of the , , and  genes under heat stress. In addition, we found that transiently transformed  seedlings showed significantly lower levels of hydrogen peroxide (HO) after heat stress and showed a stronger thermotolerance. This study sheds light on the possible functions of  genes under abiotic stress and identifies potentially useful genes to target for molecular breeding, in order to develop more stress-resistant varieties.

Genome-Wide Analysis of the  Gene Family under Abiotic Stress and Characterization of the  Gene.

 Djak (Lepidoptera, Geometridae) is a leaf-feeding pest unique to Mongolia. Outbreaks of this pest can cause larch needles to shed slowly from the top until they die, leading to a serious imbalance in the forest ecosystem. In this work, to address the need for the low-cost, fast, and effective identification of this pest, we used field survey indicators and UAV images of larch forests in Binder, Khentii, Mongolia, a typical site of  Djak pest outbreaks, as the base data, calculated relevant multispectral and red-green-blue (RGB) features, used a successive projections algorithm (SPA) to extract features that are sensitive to the level of pest damage, and constructed a recognition model of  Djak pest damage by combining patterns in the RGB vegetation indices and texture features (RGB) with the help of random forest (RF) and convolutional neural network (CNN) algorithms. The results were compared and evaluated with multispectral vegetation indices (MS) to explore the potential of UAV RGB images in identifying needle pests. The results show that the sensitive features extracted based on SPA can adequately capture the changes in the forest appearance parameters such as the leaf loss rate and the colour of the larch canopy under pest damage conditions and can be used as effective input variables for the model. The RGB-RF and RGB-CNN models have the best performance, with their overall accuracy reaching more than 85%, which is a significant improvement compared with that of the RGB model, and their accuracy is similar to that of the MS model. This low-cost and high-efficiency method can excel in the identification of  Djak-infested regions in small areas and can provide an important experimental theoretical basis for subsequent large-scale forest pest monitoring with a high spatiotemporal resolution.

Potential of Unmanned Aerial Vehicle Red-Green-Blue Images for Detecting Needle Pests: A Case Study with  Djak (Lepidoptera, Geometridae).

The mucosal system serves as the primary barrier against respiratory diseases and plays a crucial role in combating viral infections through mucosal immunity. The resident microbial community constitutes the main component of the mucosal system and exerts a significant inhibitory impact on the invasion of exogenous agents. However, the precise relationship between resident microbiota, mucosal immunity, and viral infections remains incomplete. This review aims to summarize the regulatory interactions between the resident microbiota of the mucosal system and innate immune components such as mucosal immunity and trained immunity. By clarifying these complex relationships, this review seeks to identify potential targets for augmenting respiratory disease prevention strategies and developing novel vaccine formulations. Furthermore, we propose the possibility of integrating the fields of microbiome-based therapeutics and vaccine development to create multifunctional vaccine formulations capable of targeting mucosal immunity induction. Such an approach holds great potential in offering novel pathways and strategies for the prevention and treatment of respiratory diseases.

Boreal coniferous forests play important roles in global ecological and economic processes. Mongolia, rich in forest resources and part of the boreal ecosystem, faces significant deforestation due to  Djak (Lepidoptera: Geometridae), a rapidly spreading needle pest in coniferous forests. This study aims to provide with rapid and precise pest occurrence data, enabling timely and effective control measures to preserve and enhance the agroforestry ecological environment. In vegetation disturbance detection, UAV remote sensing exhibits operational performance with unique spatiotemporal advantages (notably cm-resolution data acquisition and flexible revisit cycles) unattainable through traditional ground surveys or satellite platforms. Therefore, we used unmanned aerial vehicle (UAV) imagery from representative areas affected by , calculated conventional and red edge spectral indices, extracted features sensitive to pest infestation levels, detected disturbances using machine-learning algorithms, and analyzed the pest's spatial distribution. The sequential forward selection (SFS) and successive projection algorithms (SPA) can effectively extract features sensitive to the response to pest disturbance, in which the red edge (RE) features have a greater effect than the conventional (CONV) features in detecting the pest. The detection models developed using machine learning all achieved accuracy rates above 82%, with the Back Propagation Neural Network (BPNN) performing the best. Spatial distribution analysis based on the model revealed that  primarily exhibited a pattern of outward diffusion from the center of aggregation during the outbreak period.
