Am&#xE9;liorer l&#x26;#39;efficacit&#xE9; th&#xE9;rapeutique des cellules T exprimant un r&#xE9;cepteur d&#x26;#39;antig&#xE8;ne chim&#xE9;rique (CAR) repr&#xE9;sente un objectif important dans les efforts pour contr&#xF4;ler h&#xE9;mopathies &#xE0; cellules B. R&#xE9;cemment, une strat&#xE9;gie intrins&#xE8;que a &#xE9;t&#xE9; d&#xE9;velopp&#xE9; pour modifier la voiture elle-m&#xEA;me pour am&#xE9;liorer la signalisation des cellules T. Nous avons utilis&#xE9; &#xE9;lectrotransfert du syst&#xE8;me Belle au Bois Dormant &#xE0; introduire un transposon RCA et s&#xE9;lective propager CAR (+) des cellules T CD19 sur (+) artificiels cellules pr&#xE9;sentatrices d&#x26;#39;antig&#xE8;ne (VAMP). Lorsque l&#x26;#39;IL-21 &#xE9;tait pr&#xE9;sent, il n&#x26;#39;y avait pr&#xE9;f&#xE9;rentiel d&#x26;#39;expansion num&#xE9;rique des cellules T CD19-sp&#xE9;cifiques qui lys&#xE9;es et l&#x26;#39;IFN-&#x3B3; produit en r&#xE9;ponse &#xE0; CD19. Populations de ces RAC num&#xE9;riquement &#xE9;largi (+) des cellules T affich&#xE9;e une m&#xE9;moire ph&#xE9;notype d&#xE9;but de surface caract&#xE9;ris&#xE9; en CD62L (+) CD28 (+) et un profil de transcription des cellules T na&#xEF;fs. En revanche, les cellules T se propage avec seulement IL-2 exog&#xE8;ne ont tendance &#xE0; se traduire par une prolif&#xE9;ration de CD19-sp&#xE9;cifiques CD4 (+) des cellules T. En outre, le transfert adoptif de la RCA (+) des cellules T cultiv&#xE9;es avec IL-21 pr&#xE9;sentait un contr&#xF4;le am&#xE9;lior&#xE9; de CD19 (+) &#xE0; cellules B malignes chez la souris. Pour fournir des coordonn&#xE9;es de signalisation pour propager la RCA (+) des cellules T, nous avons d&#xE9;velopp&#xE9; une mut&#xE9;ine roman de l&#x26;#39;IL-21 li&#xE9;e &#xE0; la surface de la cellule de aAPC qui a remplac&#xE9; la n&#xE9;cessit&#xE9; d&#x26;#39;soluble IL-21. Verbesserung der therapeutischen Wirksamkeit von T-Zellen, die Ausdruck eines Chim&auml;ren Antigen-Rezeptors stellt (Auto) ein wichtiges Ziel in Bem&uuml;hungen zur Kontrolle von B-Zell-Tumoren. Vor kurzem wurde eine systeminterne Strategie entwickelt, um das Auto selbst zur Verbesserung der T-Zell-Signalisierung zu &auml;ndern. Hier berichten wir, dass ein zweiter extrinsischer Ansatz basierend auf Ver&auml;nderung des Kultur-Milieus um numerisch CAR(+) T-Zellen mit einem gew&uuml;nschten Ph&auml;notyp, f&uuml;r den Zusatz von Interleukin (IL)-21 zu Gewebekultur erweitern Auto-abh&auml;ngige T-Zell-Effektor-Funktionen verbessert. Wir fr&uuml;her Electrotransfer Sleeping Beauty Systems einzuf&uuml;hren, eine Auto-Transposon und CAR(+) T-Zellen auf CD19(+) k&uuml;nstliche Antigen-pr&auml;sentierenden Zellen (aAPC) selektiv zu verbreiten. Als IL-21 anwesend war, gab es bevorzugte numerische Erweiterung der CD19-spezifische T-Zellen, die Tauwetterprotokoll aufgel&ouml;st und IFN-&gamma; in Reaktion auf CD19 produziert. Populationen dieser numerisch erweiterten CAR(+) T-Zellen angezeigt eine fr&uuml;he Speicher Oberfl&auml;che Ph&auml;notyp als CD62L(+)CD28(+) und ein transkriptionelle Profil von naiven T-Zellen charakterisiert. Im Gegensatz dazu propagiert T-Zellen mit nur exogene Il tendenziell ein &uuml;berm&auml;&szlig;iges Wachstum von CD19-spezifische CD4(+) T-Zellen f&uuml;hren. Dar&uuml;ber hinaus stellte adoptiven Transfer CAR(+) T-Zellen kultiviert mit IL-21 verbesserte Kontrolle der CD19(+) B-Zell-Tumoren bei M&auml;usen. Um bieten koordinierte Signalisierung CAR(+) T-Zellen zu propagieren, entwickelten wir eine neuartige Mutein IL-21 gebunden an der Zelloberfl&auml;che von aAPC, die die Notwendigkeit der l&ouml;slichen IL-21 ersetzt. Improving the therapeutic efficacy of T cells expressing a chimeric antigen receptor (CAR) represents an important goal in efforts to control B-cell malignancies. Recently an intrinsic strategy has been developed to modify the CAR itself to improve T-cell signaling. Here we report a second extrinsic approach based on altering the culture milieu to numerically expand CAR(+) T cells with a desired phenotype, for the addition of interleukin (IL)-21 to tissue culture improves CAR-dependent T-cell effector functions. We used electrotransfer of Sleeping Beauty system to introduce a CAR transposon and selectively propagate CAR(+) T cells on CD19(+) artificial antigen-presenting cells (aAPC). When IL-21 was present, there was preferential numeric expansion of CD19-specific T cells which lysed and produced IFN-Î³ in response to CD19. Populations of these numerically expanded CAR(+) T cells displayed an early memory surface phenotype characterized as CD62L(+)CD28(+) and a transcriptional profile of naÃ¯ve T cells. In contrast, T cells propagated with only exogenous IL-2 tended to result in an overgrowth of CD19-specific CD4(+) T cells. Furthermore, adoptive transfer of CAR(+) T cells cultured with IL-21 exhibited improved control of CD19(+) B-cell malignancy in mice. To provide coordinated signaling to propagate CAR(+) T cells, we developed a novel mutein of IL-21 bound to the cell surface of aAPC that replaced the need for soluble IL-21. Our findings show that IL-21 can provide an extrinsic reprogramming signal to generate desired CAR(+) T cells for effective immunotherapy.

Migliorare l&#39;efficacia terapeutica di cellule T che esprimono un recettore chimerico antigene (auto) rappresentano un obiettivo importante negli sforzi per controllare i tumori maligni delle cellule di B. Recentemente una strategia intrinseca &egrave; stata sviluppata per modificare la macchina stessa per migliorare la segnalazione delle cellule T. Qui riportiamo un secondo approccio estrinseco basato su alterando il milieu di cultura per espandere numericamente CAR(+) T cellule con un fenotipo desiderato, per l&#39;aggiunta di interleuchina (IL) -21 per coltura tissutale migliora le funzioni effettrici delle cellule T auto-dipendente. Abbiamo utilizzato electrotransfer di sistema Sleeping Beauty per introdurre una trasposone auto e propagare selettivamente le cellule T CAR(+) su CD19(+) artificiale presentanti l&#39;antigene cellule (aAPC). Quando IL 21 era presente, ci fu espansione numerica preferenziale delle cellule CD19-specifiche T lisati e prodotto IFN-&gamma; in risposta a CD19. Popolazioni di queste cellule T CAR(+) numericamente espanse visualizzato un fenotipo di superficie memoria precoce caratterizzato come CD62L(+)CD28(+) e un profilo trascrizionale delle cellule T na&iuml;ve. Al contrario, le cellule T propagano con solo esogeni IL-2 tendono a causare una crescita eccessiva di cellule CD19-specifiche CD4(+) T. Inoltre, trasferimento adottivo di CAR(+) T cellule coltivate con IL 21 esposto migliorato controllo di malignit&agrave; delle cellule di B CD19(+) nei topi. La mejora de la eficacia terap&#xE9;utica de las c&#xE9;lulas T que expresan un receptor de ant&#xED;geno quim&#xE9;rico (CAR) representa un objetivo importante en los esfuerzos para controlar las c&#xE9;lulas B malignas. Recientemente una estrategia intr&#xED;nseca ha sido desarrollada para modificar el propio veh&#xED;culo para mejorar las c&#xE9;lulas T de se&#xF1;alizaci&#xF3;n. Aqu&#xED; se presenta un segundo enfoque extr&#xED;nseca basada en alterar el medio para expandir la cultura num&#xE9;ricamente CAR (+) las c&#xE9;lulas T con un fenotipo deseado, por la adici&#xF3;n de interleucina (IL) -21 al cultivo de tejidos mejora CAR-dependiente de c&#xE9;lulas T funciones efectoras. Se utiliz&#xF3; el sistema de electrotransfer de la Bella Durmiente para introducir un transpos&#xF3;n CAR y propagan selectivamente CAR (+) las c&#xE9;lulas T de CD19 (+) la presentaci&#xF3;n de ant&#xED;geno artificiales, las c&#xE9;lulas (AAPC). Cuando la IL-21 estaba presente, hubo expansi&#xF3;n preferencial num&#xE9;rico de las c&#xE9;lulas T CD19-espec&#xED;ficos que se lisaron y produjo IFN-&#x3B3; en respuesta a CD19. Las poblaciones de estos CAR num&#xE9;ricamente ampliado (+) las c&#xE9;lulas T muestran un fenotipo memoria superficie temprano caracterizado como CD62L (+) CD28 (+) y un perfil transcripcional de c&#xE9;lulas T naive. En contraste, las c&#xE9;lulas T propagan con s&#xF3;lo IL-2 ex&#xF3;gena tiende a resultar en un crecimiento excesivo de CD19 espec&#xED;ficos CD4 (+) las c&#xE9;lulas T. Adem&#xE1;s, la transferencia adoptiva de CAR (+) c&#xE9;lulas T cultivadas con IL-21 mostraron un mejor control de CD19 (+) de c&#xE9;lulas B en ratones malignidad. Para proporcionar coordinado de se&#xF1;alizaci&#xF3;n para propagar CAR (+) las c&#xE9;lulas T, hemos desarrollado una nueva mute&#xED;na de IL-21 unido a la superficie celular de AAPC que sustituye la necesidad de soluble de IL-21.
