We have previously reported an expression system based on the capsid protein gene (CP) of cucumber mosaic virus (CMV) placed under transcriptional control of a potato virus X (PVX)-based vector. PVX-expressed CMV CP formed virus-like particles, which served as carriers for heterologous antigens of the Newcastle disease virus (NDV). In this work, we applied our expression tool toward the development of plant-derived vaccine candidate against avian influenza A virus. Twenty-three amino acid-long extracellular domain of the viral M2 protein (M2e) was engineered into the internal motif 5 of CMV CP and the recombinant gene then was transiently expressed in plants through a PVX vector. Chimeric CMV capsids reacted with specific antibodies produced to synthetic M2e epitope of the H5N1 strain of the virus. In addition, CMV CP-M2e protein was expressed to high levels in Escherichia coli bacterial cells and was recognized by antibodies to both CMV and M2e. This initial study demonstrates the feasibility of using plant virus-based vectors for expression of antigenic epitopes of H5N1 avian influenza in plants.

Transient expression of the ectodomain of matrix protein 2 (M2e) of avian influenza A virus in plants.

Nous avons d&eacute;j&agrave; rapport&eacute; qu&#39;un syst&egrave;me d&#39;expression fond&eacute;e sur le g&egrave;ne de la prot&eacute;ine de capside (CP) du virus de la mosa&iuml;que du concombre (CMV) plac&eacute; sous le contr&ocirc;le transcriptionnel d&#39;un virus de la pomme de terre X PVX vectoriel. Exprim&eacute;s PVX CMV CP form&eacute; Pseudo-particules virales, qui servait de transporteurs pour les antig&egrave;nes h&eacute;t&eacute;rologues du virus de la maladie de Newcastle (NDV). Dans ce travail, nous avons appliqu&eacute; notre outil d&#39;expression vers le d&eacute;veloppement des v&eacute;g&eacute;taux vaccin candidat contre le virus de l&#39;influenza aviaire A. Vingt-trois acides amin&eacute;s long domaine extracellulaire de la prot&eacute;ine virale de M2 (M2e) a &eacute;t&eacute; con&ccedil;u dans le motif interne 5 du CP CMV et le g&egrave;ne recombinant puis a &eacute;t&eacute; exprim&eacute; transitoirement chez les plantes &agrave; travers un vecteur PVX. Chim&eacute;riques capsides CMV r&eacute;agissent avec des anticorps sp&eacute;cifiques produits synth&eacute;tiques M2e &eacute;pitope de la souche H5N1 du virus. En outre, CMV CP-M2e prot&eacute;ine a &eacute;t&eacute; exprim&eacute;e &agrave; des niveaux &eacute;lev&eacute;s dans les cellules bact&eacute;riennes Escherichia coli et a &eacute;t&eacute; reconnu par l&#39;anticorps anti-CMV tant M2e. Wir haben bereits berichtet, eine Ausdruck-System basierend auf das Kapsid-Protein-gen (CP) des Cucumber Mosaic Virus (CMV) transkriptionelle Kontrolle eines Kartoffel-Virus Vektor X PVX-basierte. CMV-CP PVX-ausgedr&uuml;ckt gebildet virus&auml;hnliche Partikel, die als Tr&auml;ger f&uuml;r heterologe Antigene von Newcastle Disease Virus (NDV) serviert. In dieser Arbeit angewendet wir unser Ausdruck Tool zur Entwicklung von pflanzlichen Impfstoff-Kandidat gegen avi&auml;re Influenza-A-Virus. Dreiundzwanzig Aminos&auml;ure langen extrazellul&auml;ren Dom&auml;ne des viralen M2 Proteins (M2e) wurde entwickelt, in das innere Motiv 5 der CMV-CP und das rekombinante gen war dann vor&uuml;bergehend in Pflanzen durch einen Vektor PVX ausgedr&uuml;ckt. Chim&auml;ren CMV Capsids reagierte mit spezifischen Antik&ouml;rpern zu synthetischen M2e-Epitop der H5N1-Stamm des Virus produziert. Dar&uuml;ber hinaus CMV CP-M2e Protein dr&uuml;ckte hohen in Escherichia coli Bakterienzellen und wurde von Antik&ouml;rper gegen CMV und M2e anerkannt. Dieser ersten Studie veranschaulicht die Machbarkeit mit Pflanze Virus-basierte Vektoren zur Interessenbekundung Antigen Epitopes von H5N1-Vogelgrippe in Pflanzen.

In precedenza abbiamo segnalato che un sistema di espressione basata su gene (CP) della proteina del capside del virus del mosaico del cetriolo (CMV) posizionato sotto il controllo trascrizionale di un virus della patata PVX X-base vettoriale. Espresso PVX CMV CP formano particelle virus-simili, che serviva come vettori per eterologo antigeni del virus della malattia di Newcastle (NDV). In questo lavoro, abbiamo applicato il nostro strumento di espressione verso lo sviluppo del candidato vegetali vaccino contro il virus a dell&#39;influenza aviaria. Hemos divulgado previamente que un sistema de expresi&oacute;n basada en el gene de la prote&iacute;na c&aacute;pside (CP) del virus del mosaico del pepino (CMV) puestas bajo el control transcripcional de un virus de patata vector X PVX-basado. PVX-expres&oacute; CMV CP forman part&iacute;culas similares a los virus, que sirvi&oacute; como portadores de ant&iacute;genos heter&oacute;logos de los virus de la enfermedad de Newcastle (NDV). En este trabajo, aplicamos nuestra herramienta de expresi&oacute;n hacia el desarrollo del candidato de origen vegetal de la vacuna contra el virus de la gripe aviar. Veintitr&eacute;s amino&aacute;cidos de largo dominio extracelular de la prote&iacute;na viral M2 (M2e) fue dise&ntilde;ado en el motivo interno 5 de CP de CMV y luego el gen recombinante transitoriamente se expres&oacute; en las plantas a trav&eacute;s de un vector PVX. Quim&eacute;rica c&aacute;psida de CMV reaccion&oacute; con anticuerpos espec&iacute;ficos producidos a sint&eacute;tico M2e epitopo de la cepa H5N1 del virus. Adem&aacute;s, la prote&iacute;na de CMV CP-M2e expres&oacute; a altos niveles en c&eacute;lulas bacterianas de Escherichia coli y fue reconocida por los anticuerpos frente al CMV tanto M2e. Este estudio inicial demuestra la viabilidad del uso de plantas basadas en virus vectores para la expresi&oacute;n de ep&iacute;topos antig&eacute;nicos de gripe aviar H5N1 en las plantas.

We have previously reported that Potato virus X-expressed coat protein of Cucumber mosaic virus (CMV) formed virus-like particles (VLPs), which served as carriers for display of different neutralizing epitopes of Newcastle disease virus (NDV). In this work, we further modified the purification protocol of recombinant VLPs carrying short neutralizing epitopes of the NDV proteins and demonstrated that self-contained capsid protein subunits of CMV transiently expressed from heterologous virus packaged into individual virions morphologically resembling and/or indistinguishable from wild type CMV particles. Homogeneity of the final preparation represents an advance over our previous study, where VLPs were found to be of variable size. Chickens immunized with purified VLPs developed antigen-specific response.

Improvement of PVX/CMV CP expression tool for display of short foreign antigens.

Nous avons d&eacute;j&agrave; rapport&eacute; que Potato virus X exprim&eacute;s prot&eacute;ine du virus de la mosa&iuml;que du concombre (CMV) form&eacute; des Pseudo-particules virales (VLP), qui a servi comme porteurs pour l&#39;affichage des diff&eacute;rents &eacute;pitopes neutralisants du virus de la maladie de Newcastle (NDV). Dans ce travail, nous encore modifi&eacute; le protocole de purification du PPV recombinants transportant des &eacute;pitopes neutralisants courts des prot&eacute;ines NDV et a d&eacute;montr&eacute; que les sous-unit&eacute;s de prot&eacute;ines de capside autonome du CMV exprim&eacute; transitoirement de virus h&eacute;t&eacute;rologue emball&eacute;s dans des virions individuelles ressemblant morphologiquement ou indiscernable de type sauvage particules CMV. Homog&eacute;n&eacute;it&eacute; de la pr&eacute;paration finale repr&eacute;sente une avanc&eacute;e au cours de notre &eacute;tude pr&eacute;c&eacute;dente, o&ugrave; VLP s&#39;est av&eacute;r&eacute; pour &ecirc;tre de taille variable. Wir haben bereits berichtet, dass Virus X-ausgedr&uuml;ckt Mantel Kartoffelprotein Cucumber Mosaic Virus (CMV) virus&auml;hnliche Partikel (VLPs), gebildet, die als Tr&auml;ger f&uuml;r die Anzeige von verschiedenen neutralisierende Epitopes von Newcastle Disease Virus (NDV) serviert. In dieser Arbeit wir weiter ver&auml;ndert das Reinigung-Protokoll des rekombinanten VLPs tragen kurze neutralisierende Epitope der NDV-Proteine und gezeigt, dass eigenst&auml;ndige Kapsid Protein-Untereinheiten der CMV vor&uuml;bergehend heterologe Virus verpackt in einzelnen Virionen morphologisch &auml;hnlich und/oder nicht von Wildtyp CMV Partikel ausgedr&uuml;ckt. Homogenit&auml;t der endg&uuml;ltigen Zubereitung stellt einen Vorschuss gegen&uuml;ber unserer fr&uuml;heren Studie, in der VLPs gefunden wurden, von variabler Gr&ouml;&szlig;e sein. H&uuml;hner geimpft mit gereinigtem VLPs entwickelt Antigen-spezifische Reaktion.

In precedenza abbiamo segnalato che Potato virus X-espresso cappotto proteina del virus del mosaico del cetriolo (CMV) formata particelle virus-simili (VLP), che serviva come vettori per la visualizzazione di diversi epitopi neutralizzanti del virus della malattia di Newcastle (NDV). In questo lavoro, abbiamo ulteriormente modificato il protocollo di purificazione di VLP ricombinante che trasportano gli epitopi neutralizzanti brevi delle proteine NDV e dimostrato che subunit&agrave; proteiche del capside autonomo del CMV transitoriamente espresso da virus eterologo indistinguibile da wild type o confezionati in singoli virioni morfologicamente simile a particelle di CMV. Omogeneit&agrave; della preparazione finale rappresenta un anticipo sul nostro studio precedente, dove le VLP sono state trovate per essere di dimensioni variabili. Polli immunizzati con VLP purificata sviluppato risposta antigene-specifica.

Maize rayado fino virus (MRFV) virus-like-particles (VLPs) produced in tobacco plants were examined for their ability to serve as a novel platform to which a variety of peptides can be covalently displayed when expressed through a Potato virus X (PVX)-based vector. To provide an anchor for chemical modifications, three Cys-MRFV-VLPs mutants were created by substituting several of the amino acids present on the shell of the wild-type MRFV-VLPs with cysteine residues. The mutant designated Cys 2-VLPs exhibited, under native conditions, cysteine thiol reactivity in bioconjugation reactions with a fluorescent dye. In addition, this Cys 2-VLPs was cross-linked by NHS-PEG4-Maleimide to 17 (F) and 8 (HN) amino acid long peptides, corresponding to neutralizing epitopes of Newcastle disease virus (NDV). The resulting Cys 2-VLPs-F and Cys 2-VLPs-HN were recognized in Western blots by antibodies to MRFV as well as to F and HN. The results demonstrated that plant-produced MRFV-VLPs have the ability to function as a novel platform for the multivalent display of surface ligands.

Mutational analysis of the two in-frame start codons and of the putative proteolytic-cleavage site showed that the N-terminal sequence is crucial for efficient cell-to-cell movement, functional systemic movement, homologous CP interactions and particle formation, but is not required for virus replication. Blocking production of the 28 kDa CP by internal initiation shows no major outcome, whereas additional mutation to prevent proteolytic cleavage at the chloroplast membrane has a dramatic effect on virus infection.
