The recent elucidation of molecular regulators of apoptosis and their roles in cellular oncogenesis has motivated the development of biomacromolecular anticancer therapeutics that can activate intracellular apoptotic signaling pathways. Pharmaceutical scientists have employed a variety of classes of biologics toward this goal, including antisense oligodeoxynucleotides, small interfering RNA, proteins, antibodies, and peptides. However, stability in the in vivo environment, tumor-specific biodistribution, cell internalization, and localization to the intracellular microenvironment where the targeted molecule is localized pose significant challenges that limit the ability to directly apply intracellular-acting, pro-apoptotic biologics for therapeutic use. Thus, approaches to improve the pharmaceutical properties of therapeutic biomacromolecules are of great significance and have included chemically modifying the bioactive molecule itself or formulation with auxiliary compounds. Recently, promising advances in delivery of pro-apoptotic biomacromolecular agents have been made using tools such as peptide "stapling", cell penetrating peptides, fusogenic peptides, liposomes, nanoparticles, smart polymers, and synergistic combinations of these components. This review will discuss the molecular mediators of cellular apoptosis, the respective mechanisms by which these mediators are dysregulated in cellular oncogenesis, the history and development of both nucleic-acid and amino-acid based drugs, and techniques to achieve intracellular delivery of these biologics. Finally, recent applications where pro-apoptotic functionality has been achieved through delivery of intracellular-acting biomacromolecular drugs will be highlighted.

La reciente aclaraci&oacute;n de reguladores moleculares de la apoptosis y sus funciones en la oncog&eacute;nesis celular ha motivado el desarrollo de la biomacromoleculares terap&eacute;utica contra el c&aacute;ncer que puede activar v&iacute;as de se&ntilde;alizaci&oacute;n intracelular de la apoptosis. Los cient&iacute;ficos farmac&eacute;uticos han empleado una variedad de clases de productos biol&oacute;gicos hacia esta meta, incluyendo oligodeoxynucleotides antisentido, ARN interferente peque&ntilde;o, prote&iacute;nas, anticuerpos y p&eacute;ptidos. Sin embargo, la estabilidad en el medio ambiente in vivo, tumor-espec&iacute;ficos biodistribuci&oacute;n, internalizaci&oacute;n celular y localizaci&oacute;n para el microambiente intracelular donde se localiza la mol&eacute;cula espec&iacute;fica plantean retos importantes que limitan la capacidad de aplicar directamente la acci&oacute;n intracelular, pro-apopt&oacute;tica productos biol&oacute;gicos para uso terap&eacute;utico. As&iacute;, enfoques para mejorar las propiedades farmac&eacute;uticas de biomacromol&eacute;culas terap&eacute;uticos son de gran importancia y han incluido modificar qu&iacute;micamente la mol&eacute;cula bioactivo s&iacute; mismo o formulaci&oacute;n con compuestos auxiliares. Recientemente, se han logrado avances prometedores en la entrega de los favorables-apoptotic biomacromoleculares agentes utilizando herramientas como p&eacute;ptido &quot;grapar&quot;, p&eacute;ptidos de penetraci&oacute;n celular, p&eacute;ptidos fusog&eacute;nica, liposomas, nanopart&iacute;culas, pol&iacute;meros inteligentes y combinaciones sin&eacute;rgicas de estos componentes. Esta revisi&oacute;n examinar&aacute; los mediadores moleculares de la apoptosis celular, los mecanismos respectivos por que estos mediadores son dysregulated en oncog&eacute;nesis celular, la historia y el desarrollo de drogas basado en &aacute;cidos nucleicos y amino&aacute;cidos y t&eacute;cnicas para lograr el transporte intracelular de estos productos biol&oacute;gicos. Finalmente, se resaltar&aacute; las &uacute;ltimas aplicaciones donde pro-apopt&oacute;tica funcionalidad se ha logrado a trav&eacute;s de la entrega de medicamentos de acci&oacute;n intracelular biomacromoleculares.
