Considering that pathological hyperactive mTOR also occurs in individuals carrying no genetic mutations, we examined whether increasing mTOR activity in neonatal NPCs of wild-type mice would recapitulate the above phenotypes. Electroporation of a plasmid encoding constitutively active Ras-homolog enriched in brain (Rheb(CA)) into subventricular zone NPCs increased mTOR activity in newborn cells. At 19 d post-electroporation (dpe), heterotopia and ectopic cells with a neuronal morphology were observed along the migratory path [rostral migratory stream (RMS)] and in the olfactory bulb (OB). These ectopic cells displayed action potentials and received synaptic inputs identifying them as synaptically integrated neurons. RMS heterotopias contained astrocytes, neurons, and entrapped neuroblasts. Immunostaining at 3 dpe revealed the presence of Mash1(+) Olig2(-) cells in the migratory route accompanied by ectopic neuronal differentiation and altered direction and speed of neuroblast migration at 7 dpe, suggesting a non-cell-autonomous disruption of migration. At >19 dpe, newborn Rheb(CA)-expressing neurons displayed altered distribution and formed micronodules in the OB. In addition, they displayed increased dendritic complexity along with altered membrane biophysics and increased frequency of GABAergic synaptic inputs. OB heterotopia, micronodules, and dendrite hypertrophy were notably prevented by rapamycin treatment, suggesting their mTOR dependence. Collectively, these data show that increasing mTOR activity in neonatal NPCs of wild-type mice recapitulate the pathologies observed in Tsc1 mutant mice. In addition, increased mTOR activity in individuals without known mutations could significantly impact neurogenesis and circuit formation.

Blanco mam&iacute;fero de rapamicina (mTOR) hiperactividad en c&eacute;lulas progenitoras neurales perinatal (PNJ) del rat&oacute;n de heterocigoto (Tsc1) complejo de 1 de la esclerosis tuberosa conduce a heterotopia y morfog&eacute;nesis neuronal anormal en pacientes con esclerosis tuberosa. Teniendo en cuenta que mTOR hiperactivo patol&oacute;gico tambi&eacute;n ocurre en individuos no que portan mutaciones gen&eacute;ticas, examinamos si aumentar la actividad de mTOR en NPCs neonatales de ratones de tipo salvaje recapitular los fenotipos anteriores. Electroporaci&oacute;n de un pl&aacute;smido codificaci&oacute;n constitutivamente activa Ras-hom&oacute;logo enriquecido en el cerebro (Rheb(CA)) en la zona subventricular PNJs aumentadas actividad de mTOR en c&eacute;lulas del reci&eacute;n nacidas. A 19 d post-electroporaci&oacute;n (dpe), heterotopia y c&eacute;lulas ect&oacute;picas con una morfolog&iacute;a neuronal se observaron a lo largo de la ruta migratoria [corriente migratoria rostral (RMS)] y en el bulbo olfatorio (OB). Estas c&eacute;lulas ect&oacute;picas muestran potenciales de acci&oacute;n y recibieron entradas sin&aacute;pticas identific&aacute;ndolos como neuronas CCIS integradas. Heterotop&iacute;as RMS figuran astrocitos, neuronas y atrap&oacute; neuroblastos. Immunostaining en dpe 3 revel&oacute; la presencia de c&eacute;lulas de Mash1(+) Olig2(-) en la ruta migratoria acompa&ntilde;ada de diferenciaci&oacute;n neuronal ect&oacute;pica y altera la direcci&oacute;n y velocidad de migraci&oacute;n neurobl&aacute;stica en 7 dpe, sugiriendo una interrupci&oacute;n no-aut&oacute;nomos-c&eacute;lula de la migraci&oacute;n. En &gt; 19 dpe, reci&eacute;n neuronas Rheb CA-expresi&oacute;n aparece alterado distribuci&oacute;n y formado micronodules en el OB. Adem&aacute;s, exhibi&oacute; mayor complejidad dendr&iacute;tica junto con Biof&iacute;sica de la membrana alterado y mayor frecuencia de entradas sin&aacute;pticas GABA&eacute;rgica. Heterotopia de OB, micronodules e hipertrofia de la dendrita en particular fueron prevenidos por el tratamiento de la rapamicina, sugiriendo su dependencia de mTOR. Colectivamente, estos datos demuestran creciente actividad de mTOR en NPCs neonatales de ratones de tipo salvaje Recapitulemos las patolog&iacute;as observadas en ratones mutantes Tsc1. Adem&aacute;s, mTOR mayor actividad en individuos sin mutaciones sabidas podr&iacute;a afectar significativamente la neurog&eacute;nesis y circuito de formaci&oacute;n.

Rheb activaci&oacute;n en los progenitores de la zona subventricular conduce a heterotopia, ect&oacute;pica diferenciaci&oacute;n neuronal y sensible a la rapamicina olfativa micronodules e hipertrofia de la dendrita de neuronas reci&eacute;n.
