During the past years in vivo transplantation experiments and in vitro colony-forming assays indicated that tumors arise only from rare cells. These cells were shown to bear self-renewal capacities and the ability to recapitulate all cell types within an individual tumor. Due to their phenotypic resemblance to normal stem cells, the term "cancer stem cells" is used. However, some pieces of the puzzle are missing: (a) a stringent definition of cancer stem cells in solid tumors (b) specific markers that only target cells that meet the criteria for a cancer stem cell in a certain type of tumor. These missing parts started an ongoing debate about which is the best method to identify and characterize cancer stem cells, or even if their mere existence is just an artifact caused by the experimental procedures. Recent findings query the cancer stem cell hypothesis for solid tumors itself since it was shown in xenograft transplantation experiments that under appropriate conditions tumor-initiating cells are not rare.In this review we critically discuss the challenges and prospects of the currently used major methods to identify cancer stem cells. Further on, we reflect the present discussion about the existence of cancer stem cells in solid tumors as well as the amount and characteristics of tumor-initiating cells and finally provide new perspectives like the correlation of cancer stem cells and induced pluripotent cells.

Cancer stem cells in solid tumors: elusive or illusive?

Au cours des derni&egrave;res ann&eacute;es des exp&eacute;riences de transplantation in vivo et des essais in vitro formatrices a indiqu&eacute; que les tumeurs proviennent uniquement des cellules rares. Ces cellules ont montr&eacute; &agrave; porter la capacit&eacute; d&#39;autorenouvellement et la capacit&eacute; de r&eacute;capituler tous les types de cellules au sein d&#39;une tumeur individuelle. En raison de leur ressemblance ph&eacute;notypique aux cellules souches normales, le terme &laquo; les cellules souches canc&eacute;reuses &raquo; est utilis&eacute;. Toutefois, certaines pi&egrave;ces du puzzle sont manquants: (a) une d&eacute;finition rigoureuse des cellules souches canc&eacute;reuses des marqueurs des tumeurs solides (b) sp&eacute;cifiques qui ciblent uniquement les cellules qui r&eacute;pondent aux crit&egrave;res d&#39;une cellule de tige de cancer dans un certain type de tumeur. Il manque des pi&egrave;ces a commenc&eacute; un d&eacute;bat en cours sur laquelle est la meilleure m&eacute;thode pour identifier et caract&eacute;riser les cellules souches canc&eacute;reuses, ou m&ecirc;me si leur simple existence est juste un artefact caus&eacute; par les proc&eacute;dures exp&eacute;rimentales. Des d&eacute;couvertes r&eacute;centes interroger l&#39;hypoth&egrave;se de cellules souches du cancer pour les tumeurs solides lui-m&ecirc;me puisqu&#39;il a &eacute;t&eacute; d&eacute;montr&eacute; dans des exp&eacute;riences de transplantation x&eacute;nogreffe que dans des conditions appropri&eacute;es les cellules tumorales qui lance ne sont pas rares.Dans cette revue, nous discutons critique les d&eacute;fis et les perspectives des principales m&eacute;thodes utilis&eacute;s actuellement pour identifier les cellules souches canc&eacute;reuses. De plus, nous refl&eacute;tons la pr&eacute;sente discussion quant &agrave; l&#39;existence de cellules souches du cancer dans les tumeurs solides ainsi que la quantit&eacute; et les caract&eacute;ristiques de la tumeur qui lance les cellules et enfin offrent de nouvelles perspectives comme la corr&eacute;lation entre les cellules souches canc&eacute;reuses et induit des cellules pluripotentes.

Les cellules souches canc&eacute;reuses des tumeurs solides : insaisissable ou illusoire ?

Negli ultimi anni gli esperimenti di trapianto in vivo e in vitro dosaggi formanti indicato che i tumori derivano solo da rare cellule. Queste cellule hanno dimostrate di sostenere la capacit&agrave; di auto-rinnovamento e la capacit&agrave; di riassumere tutti i tipi di cellule all&#39;interno di un singolo tumore. A causa della loro somiglianza fenotipica con le cellule staminali normali, viene utilizzato il termine &quot;cellule staminali tumorali&quot;. Tuttavia, mancano alcuni pezzi del puzzle: (a) una definizione rigorosa di cancro cellule staminali marcatori specifici (b) tumori solidi che colpiscono solo le celle che soddisfano i criteri per un cancro cellule in un determinato tipo di tumore. Queste parti ha iniziati un dibattito in corso sulla quale &egrave; il metodo migliore per identificare e caratterizzare le cellule staminali tumorali, o anche se mancanti loro mera esistenza &egrave; solo un artefatto causato delle procedure sperimentali. Recenti scoperte interrogare l&#39;ipotesi di cellule staminali del cancro per i tumori solidi se stesso poich&eacute; &egrave; stato dimostrato in esperimenti di trapianto xenoinnesto che in condizioni appropriate cellule tumorali di avvio non sono rare.In questa recensione discutiamo criticamente le sfide e le prospettive dei metodi principali attualmente usati per identificare le cellule staminali tumorali. Ulteriormente, ci riflettono la presente discussione circa l&#39;esistenza del cancro cellule staminali nei tumori solidi, nonch&eacute; la quantit&agrave; e le caratteristiche del tumore di avvio celle e infine fornire nuove prospettive come la correlazione delle cellule staminali tumorali e cellule pluripotenti indotte.

Durante los &uacute;ltimos a&ntilde;os experimentos de trasplante en vivo y ensayos in vitro que forman colonias indicaron que los tumores surgen s&oacute;lo de las c&eacute;lulas raras. Estas c&eacute;lulas fueron demostradas para tener capacidades de auto renovaci&oacute;n y la posibilidad de recapitular todos los tipos de c&eacute;lula dentro de un tumor individual. Debido a su semejanza fenot&iacute;pica a c&eacute;lulas madre normales, se utiliza el t&eacute;rmino &quot;c&eacute;lulas madre del c&aacute;ncer&quot;. Sin embargo, faltan algunas piezas del rompecabezas: (a) una definici&oacute;n estricta de las c&eacute;lulas madre de c&aacute;ncer en marcadores (b) espec&iacute;ficos de tumores s&oacute;lidos que s&oacute;lo atacan las c&eacute;lulas que cumplen los criterios para un c&aacute;ncer de c&eacute;lulas en un determinado tipo de tumor. &Eacute;stos faltan piezas iniciadas un debate sobre cual es el mejor m&eacute;todo para identificar y caracterizar las c&eacute;lulas madre de c&aacute;ncer, o incluso si su mera existencia es s&oacute;lo un artefacto causado por los procedimientos experimentales. Hallazgos recientes consultan la hip&oacute;tesis de c&eacute;lulas madre de c&aacute;ncer para tumores s&oacute;lidos s&iacute; mismo puesto que fue demostrado en experimentos de trasplante de xenoinjerto que bajo condiciones apropiadas c&eacute;lulas iniciadoras del tumor no son raras.En esta revisi&oacute;n se discuten cr&iacute;ticamente los retos y perspectivas de los m&eacute;todos m&aacute;s importantes utilizados en la actualidad para identificar las c&eacute;lulas madre tumorales. Adem&aacute;s, reflejamos la discusi&oacute;n actual sobre la existencia de c&eacute;lulas madre de c&aacute;ncer en tumores s&oacute;lidos, as&iacute; como la cantidad y caracter&iacute;sticas del tumor-iniciar c&eacute;lulas y finalmente proporcionar nuevas perspectivas como la correlaci&oacute;n de las c&eacute;lulas madre tumorales e inducida por c&eacute;lulas pluripotentes.

Human leukocyte antigens (HLA) bind peptides generated by limited proteolysis in cells and present them at the cell surfaces for recognition by T cells. Through this antigen presentation function they control the specificity of T cell responses and thereby adaptive immune responses. Knowledge of HLA-bound peptides is thus key to understanding adaptive immunity and to the development of vaccines and other specific immune intervention strategies. To gain insight into the antigenicity of melanomas, peptides were extracted from HLA isolated from the tumor cells, separated by two-dimensional HPLC, and sequenced by mass spectrometry. The spectra were analyzed by database-dependent MASCOT searches and database-independent de novo sequencing and, where required, confirmed with synthetic peptides, which were also used to determine their immunogenicity. Comparing four different melanoma cell lines, little overlap of the HLA-bound peptides was found, suggesting a high degree of individualization of the HLA peptidomes. This notwithstanding, the peptidomes were highly immunogenic in the patients from whom the tumor cells had been established and in unrelated patients. This broad cross-patient immunogenicity was only exceptionally related to individual peptides. The majority of the identified epitopes were derived from low to medium abundance proteins, mostly involved in sensitive cellular processes such as cell cycle control, DNA replication, control of gene expression, tumor suppressor function, and protein metabolism. The peptidomes thus provide insights into processes potentially related to tumorigenesis. Furthermore, analyses of the peptide sequences yield information on the specificity of peptide selection by HLA applicable to the developing prediction algorithms for T cell epitopes.

High immunogenicity of the human leukocyte antigen peptidomes of melanoma tumor cells.

Gr&acirc;ce &agrave; cette fonction de pr&eacute;sentation de l&#39;antig&egrave;ne, ils contr&ocirc;lent la sp&eacute;cificit&eacute; des lymphocytes T et des r&eacute;ponses immunes adaptatives ainsi. Connaissance de HLA-li&eacute; aux peptides est donc cl&eacute; pour comprendre l&#39;immunit&eacute; adaptative et au d&eacute;veloppement de vaccins et d&#39;autres strat&eacute;gies d&#39;intervention immunitaire sp&eacute;cifique. Ant&iacute;genos leucocitarios humanos (HLA) unen a p&eacute;ptidos generados por la prote&oacute;lisis limitada en las c&eacute;lulas y presentan en las superficies de la c&eacute;lula para el reconocimiento por las c&eacute;lulas T. A trav&eacute;s de esta funci&oacute;n de presentaci&oacute;n de ant&iacute;geno que controlan la especificidad de las respuestas de c&eacute;lulas T y tal modo adaptativas de las respuestas inmunitarias. Conocimiento de p&eacute;ptidos HLA-limite as&iacute; es clave para entender la inmunidad adaptativa y el desarrollo de vacunas y otras estrategias de intervenci&oacute;n inmune espec&iacute;fica. Para ganar la penetraci&oacute;n en la antigenicidad de los melanomas, p&eacute;ptidos fueron extra&iacute;dos de HLA aislado de las c&eacute;lulas del tumor, separados por HPLC bidimensional y ordenados por espectrometr&iacute;a de masas. Los espectros eran analizados por b&uacute;squedas de mascota de base de datos dependiente y la independiente de la base de datos de secuenciaci&oacute;n de novo y, cuando sea necesario, confirmados con p&eacute;ptidos sint&eacute;ticos, que tambi&eacute;n fueron usados para determinar su inmunogenicidad. Comparando cuatro l&iacute;neas celulares de melanoma diferentes, se encontr&oacute; escasa superposici&oacute;n de los p&eacute;ptidos HLA-limite, lo que sugiere un alto grado de individualizaci&oacute;n de la peptidomes HLA. Esto no obstante, los peptidomes eran altamente inmunog&eacute;nicas en los pacientes de quienes hab&iacute;an establecidas las c&eacute;lulas del tumor y en pacientes sin relaci&oacute;n. Este amplia inmunogenicidad Cruz-paciente s&oacute;lo excepcionalmente se relacion&oacute; con p&eacute;ptidos individuales. La mayor&iacute;a de los epitopos identificados fueron derivada de baja a las prote&iacute;nas de la abundancia media, en su mayor&iacute;a involucradas en procesos celulares sensibles como control del ciclo celular, replicaci&oacute;n del ADN, el control de la expresi&oacute;n g&eacute;nica, funci&oacute;n de supresor del tumor y el metabolismo de la prote&iacute;na. As&iacute;, las peptidomes proporcionar penetraciones en procesos potencialmente relacionados a la tumorig&eacute;nesis. Adem&aacute;s, an&aacute;lisis de las secuencias de p&eacute;ptidos producen informaci&oacute;n sobre la especificidad de la selecci&oacute;n de p&eacute;ptido por HLA aplicable a los algoritmos de predicci&oacute;n en desarrollo para epitopos T cell.
