Optical projection tomography (OPT) imaging is a powerful tool for three-dimensional imaging of gene and protein distribution patterns in biomedical specimens. We have previously demonstrated the possibility, by this technique, to extract information of the spatial and quantitative distribution of the islets of Langerhans in the intact mouse pancreas. In order to further increase the sensitivity of OPT imaging for this type of assessment, we have developed a protocol implementing a computational statistical approach: contrast limited adaptive histogram equalization (CLAHE). We demonstrate that this protocol significantly increases the sensitivity of OPT imaging for islet detection, helps preserve islet morphology and diminish subjectivity in thresholding for tomographic reconstruction. When applied to studies of the pancreas from healthy C57BL/6 mice, our data reveal that, at least in this strain, the pancreas harbors substantially more islets than has previously been reported. Further, we provide evidence that the gastric, duodenal and splenic lobes of the pancreas display dramatic differences in total and relative islet and Î²-cell mass distribution. This includes a 75% higher islet density in the gastric lobe as compared to the splenic lobe and a higher relative volume of insulin producing cells in the duodenal lobe as compared to the other lobes. Altogether, our data show that CLAHE substantially improves OPT based assessments of the islets of Langerhans and that lobular origin must be taken into careful consideration in quantitative and spatial assessments of the pancreas.

An improved protocol for optical projection tomography imaging reveals lobular heterogeneities in pancreatic islet and Î²-cell mass distribution.

Imagerie de tomographie (OPT) projection optique est un outil puissant pour l&#39;imagerie en trois dimensions des patrons de distribution des g&egrave;nes et des prot&eacute;ines chez les sp&eacute;cimens biom&eacute;dicaux. Nous avons d&eacute;j&agrave; d&eacute;montr&eacute; la possibilit&eacute;, par cette technique, pour extraire des informations de la distribution spatiale et quantitative des &icirc;lots de Langerhans dans le pancr&eacute;as de souris intactes. Afin d&#39;accro&icirc;tre encore la sensibilit&eacute; de l&#39;imagerie OPT pour ce type d&#39;&eacute;valuation, nous avons d&eacute;velopp&eacute; un protocole de mise en &oelig;uvre d&#39;une approche statistique computationnelle : contraste limit&eacute; d&#39;&eacute;galisation adaptative histogramme (CLAHE). Nous d&eacute;montrons que ce protocole augmente consid&eacute;rablement la sensibilit&eacute; de l&#39;imagerie OPT pour la d&eacute;tection de l&#39;&icirc;lot, contribue &agrave; pr&eacute;server la morphologie de l&#39;&icirc;lot et diminue la subjectivit&eacute; en seuillage pour reconstruction tomographique. Lorsqu&#39;elle est appliqu&eacute;e &agrave; l&#39;&eacute;tude du pancr&eacute;as de chez la souris C57BL/6, nos donn&eacute;es indiquent que, au moins dans cette souche, le pancr&eacute;as rec&egrave;le des &icirc;lots beaucoup plus importante qu&#39;on a d&eacute;j&agrave; signal&eacute;. De plus, nous d&eacute;montrons que les lobes spl&eacute;niques, gastriques et duod&eacute;nales du pancr&eacute;as pr&eacute;sentent des diff&eacute;rences dramatiques en totale et relative &icirc;lot et cellules &beta; distribution massive. Il s&#39;agit d&#39;une densit&eacute; sup&eacute;rieure &agrave; 75 % &icirc;lot dans le lobe gastrique par comparaison avec le lobe spl&eacute;nique et un volume relatif plus &eacute;lev&eacute; de cellules dans le lobe duod&eacute;nal par rapport &agrave; d&#39;autres les lobes productrices d&#39;insuline. Au total, nos donn&eacute;es montrent que CLAHE am&eacute;liore sensiblement OPT fond&eacute; &eacute;valuations des &icirc;lots de Langerhans et cette origine lobulaire doit &ecirc;tre prise en consid&eacute;ration dans les &eacute;valuations quantitatives et spatiales du pancr&eacute;as.

Optische Projektion Tomographie (OPT) Imaging ist ein leistungsf&auml;higes Werkzeug f&uuml;r dreidimensionale Bildgebung der Gene und Proteine Verteilungsmuster in biomedizinischen Proben. Zuvor haben wir die M&ouml;glichkeit, durch diese Technik, zum Extrahieren von Informationen &uuml;ber die r&auml;umliche und mengenm&auml;&szlig;ige Verteilung der Langerhans-Inseln der Bauchspeicheldr&uuml;se intakt Maus gezeigt. Um die Empfindlichkeit des OPT-Imaging f&uuml;r diese Art der Bewertung zu steigern, haben wir ein Protokoll implementieren einen computational statistischen Ansatz entwickelt: Kontrast begrenzt adaptive Histogramm Entzerrung (CLAHE). Wir zeigen, dass dieses Protokoll erh&ouml;ht deutlich die Sensitivit&auml;t des OPT-Imaging f&uuml;r Insel-Erkennung, hilft bewahren Inselchen Morphologie und Subjektivit&auml;t in Thresholding tomographischen Wiederaufbau zu verringern. Bei Anwendung auf Studien des Pankreas von gesunden C57BL/6 M&auml;usen zeigen unsere Daten, dass zumindest dieser Belastung die Bauchspeicheldr&uuml;se birgt wesentlich mehr Inseln als zuvor berichtet wurde. Des weiteren bieten wir Hinweise, dass die Milz, Magen und Zw&ouml;lffingerdarm Lappen der Bauchspeicheldr&uuml;se dramatische Unterschiede im Gesamt und relative Islet und &beta;-Zellen Massenverteilung anzeigen. Dies umfasst eine 75 % h&ouml;here Inselchen Dichte in den Magen Lappen im Vergleich zu der Milz Lobus und eine h&ouml;here relative Lautst&auml;rke der Insulin produzierenden Zellen in den Zw&ouml;lffingerdarm Lappen im Vergleich zu den anderen Lappen. Insgesamt unsere Daten zeigen, dass CLAHE wesentlich, OPT verbessert basierte Einsch&auml;tzungen der Langerhans-Inseln und diesen lobul&auml;res Ursprung muss ber&uuml;cksichtigt werden vorsichtig in quantitativer und r&auml;umliche Einsch&auml;tzungen der Bauchspeicheldr&uuml;se.

Proiezione ottica tomografia (OPT) imaging &egrave; uno strumento potente per l&#39;imaging tridimensionale di modelli di distribuzione gene e proteina in campioni biomedici. In precedenza abbiamo dimostrato la possibilit&agrave;, da questa tecnica, per estrarre informazioni della distribuzione spaziale e quantitativa delle isole di Langerhans del pancreas intatto del mouse. Al fine di aumentare ulteriormente la sensibilit&agrave; dell&#39;imaging OPT per questo tipo di valutazione, abbiamo sviluppato un protocollo di attuazione di un approccio statistico computazionale: contrasto limited equalizzazione adattativa istogramma (CLAHE). Dimostreremo che questo protocollo aumenta notevolmente la sensibilit&agrave; dell&#39;imaging OPT per il rilevamento di isolotto, aiuta a preserve la morfologia isolotto e diminuisce la soggettivit&agrave; nella soglia per la ricostruzione tomografica. Quando viene applicato agli studi del pancreas da topi C57BL/6 sani, i nostri dati rivelano che, almeno in questo sforzo, il pancreas porti sostanzialmente pi&ugrave; isolotti che precedentemente &egrave; stata segnalata. Inoltre, forniamo prove che i lobi splenici, gastrici e duodenali del pancreas mostrano differenze drammatiche in totale e relativa isoletta e &beta;-cellula distribuzione di massa. Questo include una maggiore densit&agrave; di isolotto del 75% nel lobo gastrico rispetto al lobo splenico e un maggiore volume relativo di insulina producendo le cellule del lobo duodenale rispetto ad altri lobi. Proyecci&oacute;n de imagen de proyecci&oacute;n &oacute;ptica tomograf&iacute;a (OPT) es una poderosa herramienta para obtener im&aacute;genes tridimensionales de patrones de distribuci&oacute;n de genes y prote&iacute;nas en muestras biom&eacute;dicas. Previamente hemos demostrado la posibilidad, mediante esta t&eacute;cnica, para extraer informaci&oacute;n de la distribuci&oacute;n espacial y cuantitativa de los islotes de Langerhans en el p&aacute;ncreas de rat&oacute;n intacto. Para aumentar a&uacute;n m&aacute;s la sensibilidad de la proyecci&oacute;n de imagen de OPT para este tipo de evaluaci&oacute;n, hemos desarrollado un protocolo de implementaci&oacute;n de un enfoque estad&iacute;stico computacional: contraste limitada ecualizaci&oacute;n de histograma adaptativos (CLAHE). Demostramos que este protocolo aumenta significativamente la sensibilidad de la proyecci&oacute;n de imagen de OPT para detecci&oacute;n de islote, ayuda a preservar la morfolog&iacute;a del islote y disminuir la subjetividad en el umbral de reconstrucci&oacute;n tomogr&aacute;fica. Cuando se aplica a los estudios del p&aacute;ncreas de saludables ratones C57BL/6, nuestros datos revelan que, al menos en esta cepa, el p&aacute;ncreas alberga substancialmente m&aacute;s islotes que se ha divulgado previamente. Adem&aacute;s, Proporcionamos evidencia que los l&oacute;bulos g&aacute;stricos y duodenales y espl&eacute;nicos del p&aacute;ncreas presentan diferencias abismales en distribuci&oacute;n masiva del islote y &beta;-celular total y relativa. Since it was first presented in 2002, optical projection tomography (OPT) has emerged as a powerful tool for the study of biomedical specimen on the mm to cm scale. In this paper, we present computational tools to further improve OPT image acquisition and tomographic reconstruction. More specifically, these methods provide: semi-automatic and precise positioning of a sample at the axis of rotation and a fast and robust algorithm for determination of postalignment values throughout the specimen as compared to existing methods. These tools are easily integrated for use with current commercial OPT scanners and should also be possible to implement in "home made" or experimental setups for OPT imaging. They generally contribute to increase acquisition speed and quality of OPT data and thereby significantly simplify and improve a number of three-dimensional and quantitative OPT based assessments.

In diesem Beitrag stellen wir computational Tools zur OPT-Bilderfassung und computertomographische Wiederaufbau weiter zu verbessern. Genauer gesagt, diese Methoden bieten: halbautomatische und pr&auml;zise Positionierung einer Probe an der Achse der Drehung und eine schnelle und robuste Algorithmus zur Bestimmung des postalignment Werte w&auml;hrend der Probe im Vergleich zu bestehenden Methoden. Diese Werkzeuge sind leicht f&uuml;r die Verwendung mit aktuellen kommerziellen OPT-Scanner integriert und sollte auch m&ouml;glich sein, implementieren in &quot;home made&quot; oder Versuchsaufbauten f&uuml;r OPT-Imaging. Puesto que primero fue presentado en 2002, tomograf&iacute;a de proyecci&oacute;n &oacute;ptica (OPT) ha surgido como una poderosa herramienta para el estudio del esp&eacute;cimen biom&eacute;dica en la mm a escala de cm. En este trabajo presentamos herramientas computacionales para mejorar a&uacute;n m&aacute;s la adquisici&oacute;n de im&aacute;genes OPT y reconstrucci&oacute;n tomogr&aacute;fica. M&aacute;s concretamente, estos m&eacute;todos proporcionan: semiautom&aacute;tico y precisa colocaci&oacute;n de una muestra en el eje de rotaci&oacute;n y un algoritmo r&aacute;pido y robusto para la determinaci&oacute;n de valores postalignment a lo largo de la muestra en comparaci&oacute;n con los m&eacute;todos existentes. Estas herramientas se integran f&aacute;cilmente para el uso con esc&aacute;neres OPT comerciales actuales y tambi&eacute;n deber&iacute;an ser posibles implementar en &quot;caseras&quot; o configuraciones experimentales para la proyecci&oacute;n de imagen de OPT. Generalmente contribuyen a aumentar la velocidad y calidad de los datos de los territorios palestinos ocupados y as&iacute; significativamente simplificar y mejorar varias evaluaciones de OPT base tridimensionales y cuantitativas.
