A set of monolithic stationary phases representing a broad span of monomers and porogens have been characterized directly in their capillary chromatographic format by computational assessment of their pore structure from transmission electron micrographs obtained after in situ embedment of the monoliths in contrast resin, followed by dissolution of the fused-silica tubing, further encasement of the resin-embedded monolith, and microtomy. This technique has been compared to mercury intrusion, a more conventional technique for macroporosity estimation. Supplementing the embedding resin by lead methacrylate gave a negative staining, and the resulting micrographs showed a good contrast between the polymeric monoliths and the embedding resin that allowed studies on the pore formation and polymer development. The technique was also applied to a commercial monolithic silica column.

Una serie di fasi stazionarie monolitici che rappresenta un ampio arco di monomeri e porogens sono state caratterizzate direttamente nel loro formato cromatografica capillare di valutazione computazionale della loro struttura di poro da transmission electron micrografie ottenute dopo infissione in situ dei monoliti in contrasto resina, seguita dalla dissoluzione del tubo di silice fusa, ulteriormente encasement del monolite resina incorporati e Microtomia. Questa tecnica &egrave; stata paragonata all&#39;intrusione di mercurio, una tecnica pi&ugrave; convenzionale per la stima di macroporosit&agrave;. Un conjunto de fases estacionarias monol&iacute;ticos que representa un amplio lapso de mon&oacute;meros y por&oacute;genos han sido caracterizados directamente en su formato de cromatograf&iacute;a capilar mediante evaluaci&oacute;n computacional de su estructura de poro de transmisi&oacute;n micrograf&iacute;as obtenidos despu&eacute;s de incrustaci&oacute;n in situ de los monolitos en cambio resina, seguida de la disoluci&oacute;n de la tuber&iacute;a de S&iacute;lice fundida, m&aacute;s protector del monolito embebido en resina y microtom&iacute;a. Esta t&eacute;cnica ha sido comparada con la intrusi&oacute;n de mercurio, una t&eacute;cnica m&aacute;s convencional para la estimaci&oacute;n de la macroporosidad. Complementando la resina empotrar por plomo metacrilato dio una tinci&oacute;n negativa y las micrograf&iacute;as resultantes mostraron un buen contraste entre los monolitos polim&eacute;ricos y la resina de inclusi&oacute;n que permite estudios sobre la formaci&oacute;n del poro y el desarrollo de pol&iacute;meros. Since it was first presented in 2002, optical projection tomography (OPT) has emerged as a powerful tool for the study of biomedical specimen on the mm to cm scale. In this paper, we present computational tools to further improve OPT image acquisition and tomographic reconstruction. More specifically, these methods provide: semi-automatic and precise positioning of a sample at the axis of rotation and a fast and robust algorithm for determination of postalignment values throughout the specimen as compared to existing methods. These tools are easily integrated for use with current commercial OPT scanners and should also be possible to implement in "home made" or experimental setups for OPT imaging. They generally contribute to increase acquisition speed and quality of OPT data and thereby significantly simplify and improve a number of three-dimensional and quantitative OPT based assessments.

Depuis sa premi&egrave;re pr&eacute;sentation en 2002, la tomographie optique de projection (OPT) a &eacute;merg&eacute; comme un outil puissant pour l&#39;&eacute;tude de sp&eacute;cimens biom&eacute;dicaux sur le mm &agrave; cm &eacute;chelle. Dans cet article, nous pr&eacute;sentons les outils informatiques pour am&eacute;liorer davantage l&#39;acquisition d&#39;images OPT et reconstruction tomographique. Plus sp&eacute;cifiquement, ces m&eacute;thodes fournissent : positionnement semi-automatique et pr&eacute;cis d&#39;un &eacute;chantillon &agrave; l&#39;axe de rotation et un algorithme rapide et robuste pour la d&eacute;termination des valeurs de postalignment dans l&#39;ensemble de l&#39;&eacute;chantillon par rapport aux m&eacute;thodes existantes. Da es im Jahr 2002 erstmals pr&auml;sentiert wurde, hat optische Projektion Tomographie (OPT) als ein leistungsf&auml;higes Werkzeug f&uuml;r das Studium der biomedizinischen Muster auf die mm bis cm Umfang entstanden. In diesem Beitrag stellen wir computational Tools zur OPT-Bilderfassung und computertomographische Wiederaufbau weiter zu verbessern. Genauer gesagt, diese Methoden bieten: halbautomatische und pr&auml;zise Positionierung einer Probe an der Achse der Drehung und eine schnelle und robuste Algorithmus zur Bestimmung des postalignment Werte w&auml;hrend der Probe im Vergleich zu bestehenden Methoden. Diese Werkzeuge sind leicht f&uuml;r die Verwendung mit aktuellen kommerziellen OPT-Scanner integriert und sollte auch m&ouml;glich sein, implementieren in &quot;home made&quot; oder Versuchsaufbauten f&uuml;r OPT-Imaging. Sie tragen im Allgemeinen zur Akquisition Geschwindigkeit und Qualit&auml;t der OPT-Daten zu erh&ouml;hen und dadurch erheblich Vereinfachung und Verbesserung verschiedener Bewertungen in den dreidimensionalen und quantitative OPT basiert.

Dal momento che esso &egrave; stato presentato nel 2002, Tomografia ottica di proiezione (OPT) &egrave; emerso come un potente strumento per lo studio del campione biomedica sulla mm a scala cm. In questo libro, vi presentiamo gli strumenti computazionali per migliorare ulteriormente la OPT immagine acquisizione e ricostruzione tomografica. Puesto que primero fue presentado en 2002, tomograf&iacute;a de proyecci&oacute;n &oacute;ptica (OPT) ha surgido como una poderosa herramienta para el estudio del esp&eacute;cimen biom&eacute;dica en la mm a escala de cm. En este trabajo presentamos herramientas computacionales para mejorar a&uacute;n m&aacute;s la adquisici&oacute;n de im&aacute;genes OPT y reconstrucci&oacute;n tomogr&aacute;fica. M&aacute;s concretamente, estos m&eacute;todos proporcionan: semiautom&aacute;tico y precisa colocaci&oacute;n de una muestra en el eje de rotaci&oacute;n y un algoritmo r&aacute;pido y robusto para la determinaci&oacute;n de valores postalignment a lo largo de la muestra en comparaci&oacute;n con los m&eacute;todos existentes. Estas herramientas se integran f&aacute;cilmente para el uso con esc&aacute;neres OPT comerciales actuales y tambi&eacute;n deber&iacute;an ser posibles implementar en &quot;caseras&quot; o configuraciones experimentales para la proyecci&oacute;n de imagen de OPT. Generalmente contribuyen a aumentar la velocidad y calidad de los datos de los territorios palestinos ocupados y as&iacute; significativamente simplificar y mejorar varias evaluaciones de OPT base tridimensionales y cuantitativas.
