The A Disintegrin And Metalloprotease (ADAM) family of metalloproteases affects a variety of proteins with important roles in development and disease, including growth factors and adhesion molecules. We have analyzed the expression patterns of ADAMs 9, 10, and 17 during pancreas ontogeny. All ADAMs investigated were expressed in the pancreatic anlagen but invariably became restricted to divergent pancreatic compartments. ADAM9 and 17 became restricted to the insulin-producing beta-cells and all islet cells, respectively. During embryogenesis, ADAM10 was detected predominantly in acinar cells, but in the adult, it was localized to the cell surface membrane of both endocrine and exocrine cells. In addition to ADAM9, a potential prognostic factor for ductal cancers, we describe the expression of ADAM10 and ADAM17 in the pancreatic ductal epithelium. Altogether, the dynamic expression profile of the ADAM proteases described here may reflect a functional divergence of these as mediators of pancreas biology.

Developmental expression of metalloproteases ADAM 9, 10, and 17 becomes restricted to divergent pancreatic compartments.

Die Familie A Disintegrin und Metalloproteinase (ADAM) von Metalloproteinasen betrifft eine Vielzahl von Proteinen mit wichtigen Rollen in der Entwicklung und Krankheit, einschlie&szlig;lich Wachstumsfaktoren und Adh&auml;sionsmolek&uuml;le. Wir haben die Muster von ADAMs, 9, 10 und 17 w&auml;hrend der Ontogenese des Pankreas analysiert. Tutti ADAMs indagati sono stati espressi nella anlagen pancreatico ma invariabilmente &egrave; diventato limitati agli scomparti del pancreas divergenti. La familia A conclusi&oacute;n y Metaloproteinasa (ADAM) de metaloproteasas afecta a una variedad de prote&iacute;nas con funciones importantes en el desarrollo y la enfermedad, incluyendo factores de crecimiento y mol&eacute;culas de adhesi&oacute;n. Hemos analizado los patrones de expresi&oacute;n de ADAMs 9, 10 y 17 durante la ontogenia del p&aacute;ncreas. Members of the Nkx family of homeodomain proteins are involved in a variety of developmental processes such as cell fate determination in the CNS and in the pancreas. Here we describe the cloning and developmental expression pattern of Nkx6.3, a new member of the Nkx6 subfamily of homeodomain proteins. Nkx6.3 is expressed in the developing CNS and gastro-intestinal tract. In contrast to Nkx6.1 and Nkx6.2 that are broadly expressed in ventral positions of the developing CNS, Nkx6.3 shows a remarkably selective expression in a subpopulation of differentiating V2 neurons at caudal hindbrain levels. The expression of Nkx6.3 at this level depends on the activity of other Nkx6 proteins. In the gut, Nkx6.3 is expressed in duodenal and glandular stomach endoderm and at the end of gestation Nkx6.3 became restricted to the base of the gastric units in the glandular stomach. The expression of Nkx6.3 overlapped with the expression of Nkx6.2 both in the CNS and in the gut. Transient Nkx6.2 expression was also detected in the developing pancreas. However, analysis of Nkx6.2(-/-) mice did not display any obvious aberrations of pancreatic or stomach development.

Transiente Nkx6.2 Ausdruck wurde auch in den Entwicklungsl&auml;ndern Bauchspeicheldr&uuml;se festgestellt. A predicament when assessing the mechanisms underlying the pathogenesis of type-1 diabetes (T1D) has been to maintain simultaneous global and regional information on the loss of insulin-cell mass and the progression of insulitis. We present a procedure for high-resolution 3-D analyses of regions of interest (ROIs), defined on the basis of global assessments of the 3-D distribution, size, and shape of molecularly labeled structures within the full volume of the intact mouse pancreas. We apply a refined protocol for optical projection tomography (OPT)-aided whole pancreas imaging in combination with confocal laser scanning microscopy of site-directed pancreatic microbiopsies. As such, the methodology provides a useful tool for detailed cellular and molecular assessments of the autoimmune insulitis in T1D. It is anticipated that the same approach could be applied to other areas of research where 3-D molecular distributions of both global and regional character is required.

Un dilema cuando evaluar los mecanismos subyacentes a la patog&eacute;nesis de la diabetes de tipo 1 (T1D) ha sido mantener simult&aacute;nea informaci&oacute;n global y regional sobre la p&eacute;rdida de c&eacute;lulas insulina masa y la progresi&oacute;n de insulitis. Presentamos un procedimiento de an&aacute;lisis tridimensionales de alta resoluci&oacute;n de regiones de inter&eacute;s (ROIs), definido a partir de evaluaciones globales de la distribuci&oacute;n tridimensional, tama&ntilde;o y forma de estructuras molecularmente etiquetadas dentro del volumen completo del p&aacute;ncreas rat&oacute;n intacto. Aplicamos un protocolo refinado para tomograf&iacute;a de proyecci&oacute;n &oacute;ptica (OPT)-con la ayuda de im&aacute;genes de p&aacute;ncreas entero en combinaci&oacute;n con l&aacute;ser confocal de microscop&iacute;a de microbiopsies pancre&aacute;tico dirigida. Como tal, la metodolog&iacute;a proporciona una herramienta &uacute;til para las evaluaciones detalladas de celulares y moleculares de la insulitis autoinmune en T1D. Se prev&eacute; que lo mismo podr&iacute;a aplicarse a otras &aacute;reas de investigaci&oacute;n donde se requiere la distribuci&oacute;n molecular tridimensional de car&aacute;cter regional y mundial.
