&#x63A2;&#x6D4B; CXCR1 &#x548C; CXCR2 &#x53D7;&#x4F53;&#x7684;&#x6FC0;&#x6D3B;&#x548C; CXC &#x6BCD;&#x9898;&#x4E2D;&#x8D8B;&#x5316;&#x56E0;&#x5B50; CXCL8 &#x7684;&#x9AD8;&#x4EB2;&#x548C;&#x6027;&#x7ED3;&#x5408;&#x7684;&#x4F5C;&#x7528;&#x3002;

All chemokines share a common structural scaffold that mediate a remarkable variety of functions from immune surveillance to organogenesis. Chemokines are classified as CXC or CC on the basis of conserved cysteines, and the two subclasses bind distinct sets of GPCR class of receptors and also have markedly different quaternary structures, suggesting that the CXC/CC motif plays a prominent role in both structure and function. For both classes, receptor activation involves interactions between chemokine N-loop and receptor N-domain residues (Site-I), and between chemokine N-terminal and receptor extracellular/transmembrane residues (Site-II). We engineered a CC variant (labeled as CC-CXCL8) of the chemokine CXCL8 by deleting residue X (CXC â†’ CC), and found its structure is essentially similar to WT. In stark contrast, CC-CXCL8 bound poorly to its cognate receptors CXCR1 and CXCR2 (K(i) > 1 Î¼m). Further, CC-CXCL8 failed to mobilize Ca(2+) in CXCR2-expressing HL-60 cells or recruit neutrophils in a mouse lung model. However, most interestingly, CC-CXCL8 mobilizes Ca(2+) in neutrophils and in CXCR1-expressing HL-60 cells. Compared with the WT, CC-CXCL8 binds CXCR1 N-domain with only âˆ¼5-fold lower affinity indicating that the weak binding to intact CXCR1 must be due to its weak binding at Site-II. Nevertheless, this level of binding is sufficient for receptor activation indicating that affinity and activity are separable functions. We propose that the CXC motif functions as a conformational switch that couples Site-I and Site-II interactions for both receptors, and that this coupling is critical for high affinity binding but differentially regulates activation.

Probing the role of CXC motif in chemokine CXCL8 for high affinity binding and activation of CXCR1 and CXCR2 receptors.

Chimiokines tous partagent un &eacute;chafaudage structurels commun qui n&eacute;gocient une remarquable vari&eacute;t&eacute; de fonctions de surveillance du syst&egrave;me immunitaire &agrave; l&#39;organogen&egrave;se. Chimiokines sont class&eacute;s comme CXC ou CC sur la base des cyst&eacute;ines conserv&eacute;es, et les deux sous-classes lient des ensembles distincts de classe GPCR des r&eacute;cepteurs et ont &eacute;galement des structures quaternaires sensiblement diff&eacute;rentes, ce qui sugg&egrave;re que le motif CXC/CC joue un r&ocirc;le important dans la structure et la fonction. De plus, CC-CXCL8 n&#39;a pas Ca(2+) dans les cellules exprimant le CXCR2 HL-60 de mobiliser ou de recruter des neutrophiles dans un mod&egrave;le de poumons de souris. Cependant, plus int&eacute;ressant, le CC-CXCL8 mobilise Ca(2+) neutrophiles et dans les cellules exprimant le CXCR1 HL-60. Compar&eacute; avec le Tae, CC-CXCL8 lie CXCR1 N-domaine avec seulement &sim;5-pli plus faible affinit&eacute; indiquant que la liaison faible &agrave; CXCR1 intact doit &ecirc;tre &agrave; cause de sa faible liaison au Site-II. N&eacute;anmoins, ce niveau de liaison est suffisant pour r&eacute;cepteur activation indiquant que l&#39;affinit&eacute; et activit&eacute; sont s&eacute;parables fonctions. Nous proposons que le motif CXC fonctionne comme un commutateur conformationnels qui allie le Site-j&#39;et Site-II interactions pour les deux r&eacute;cepteurs, et que ce couplage est critique pour la liaison de haute affinit&eacute; mais r&eacute;glemente diff&eacute;remment d&#39;activation.

Tutte le chemochine condividono un&#39;impalcatura strutturale comune che mediano una notevole variet&agrave; di funzioni di sorveglianza immunitaria di organogenesi. Rapid mobilization of neutrophils from vasculature to the site of bacterial/viral infections and tissue injury is a critical step in successful resolution of inflammation. The chemokine CXCL8 plays a central role in recruiting neutrophils. A characteristic feature of CXCL8 is its ability to reversibly exist as both monomers and dimers, but whether both forms exist in vivo, and if so, the relevance of each form for in vivo function is not known. In this study, using a 'trapped' non-associating monomer and a non-dissociating dimer, we show that (i) wild type (WT) CXCL8 exists as both monomers and dimers, (ii) the in vivo recruitment profiles of the monomer, dimer, and WT are distinctly different, and (iii) the dimer is essential for initial robust recruitment and the WT is most active for sustained recruitment. Using a microfluidic device, we also observe that recruitment is not only dependent on the total amount of CXCL8 but also on the steepness of the gradient, and the gradients created by different CXCL8 variants elicit different neutrophil migratory responses. CXCL8 mediates its function by binding to CXCR2 receptor on neutrophils and glycosaminoglycans (GAGs) on endothelial cells. On the basis of our data, we propose that dynamic equilibrium between CXCL8 monomers and dimers and their differential binding to CXCR2 and GAGs mediates and regulates in vivo neutrophil recruitment. Our finding that both CXCL8 monomer and dimer are functional in vivo is novel, and indicates that the CXCL8 monomer-dimer equilibrium and neutrophil recruitment are intimately linked in health and disease.

In dieser Studie mit einem &#39;gefangen&#39; nicht zuordnen-Monomer und eine nicht-man distanziert-Dimer, zeigen wir, dass (i) Wildtyp (WT) CXCL8 existiert als Monomere und Dimere, (Ii) die in-vivo-Rekrutierung-Profile der Monomer, Dimer und WT sind deutlich anders, und (Iii) die Dimer unverzichtbar f&uuml;r die erste robuste Einstellung und WT ist aktivste f&uuml;r nachhaltige Einstellung. Mit einem mikrofluidische, beobachten wir auch, dass Einstellung nicht nur von den Gesamtbetrag der CXCL8 sondern auch die Steilheit des Farbverlaufs h&auml;ngt und die Farbverl&auml;ufe erstellt von CXCL8-Varianten verschiedene neutrophil migratorische entlocken. CXCL8 vermittelt seine Funktion durch Bindung an CXCR2 Rezeptor auf neutrophile und Glycosaminoglycans (GAG) in Endothelzellen. Auf Basis unserer Daten schlagen wir vor, dass dynamisches Gleichgewicht zwischen CXCL8-Monomere und Dimere und ihre differenzielle Bindung an CXCR2 und GAGs vermittelt und in-vivo neutrophil Rekrutierung regelt. Unsere Feststellung, dass CXCL8 Monomeren und dimeren in Vivo funktionsf&auml;hig sind Roman, und gibt an, dass die CXCL8 Monomer-Dimer Gleichgewicht und neutrophil Rekrutierung in Gesundheit und Krankheit eng verkn&uuml;pft sind.

La chemochina CXCL8 svolge un ruolo centrale nel reclutamento dei neutrofili. Una caratteristica della CXCL8 &egrave; la sua capacit&agrave; di reversibilmente esistere sia come monomeri e dimeri, ma se entrambe le forme esistono in vivo e se &egrave; cos&igrave;, non &egrave; nota la rilevanza di ogni forma di funzione in vivo. Utilizzando un dispositivo di microfluidica, osserviamo anche assunzione non &egrave; solo dipendente sulla quantit&agrave; totale di CXCL8, ma anche la ripidezza della sfumatura, che i gradienti creati da diverse varianti CXCL8 suscitano differenti risposte migratorie dei neutrofili. La r&#xE1;pida movilizaci&#xF3;n de los neutr&#xF3;filos de la vasculatura en el sitio de las infecciones bacterianas / viral y lesiones de tejidos es un paso cr&#xED;tico en la resoluci&#xF3;n satisfactoria de la inflamaci&#xF3;n. Las quimioquinas CXCL8 juega un papel central en los neutr&#xF3;filos de reclutamiento. Un rasgo caracter&#xED;stico de CXCL8 es su capacidad de existir como reversiblemente ambos mon&#xF3;meros y d&#xED;meros, pero si las dos formas existen in vivo, y si es as&#xED;, la importancia de cada forma de la funci&#xF3;n in vivo no se conoce. In the past decade the mouse has become the primary animal model of a variety of lung diseases. To assess various mechanisms underlying such pathologies, it is essential to make functional measurements that can reflect the developing pathology. In this regard, the diffusing capacity for carbon monoxide is a variable that directly reflects structural changes in the lung. Although measurement of single-breath diffusing capacity of the lung for carbon monoxide (DL(CO)) has also been previously reported in mice by a number of investigators, a number of technical issues have precluded routine and widespread use of this metric in mouse models. In the present report, we describe a means to quickly and simply measure a dimensionless variable closely related to the DL(CO) in mice, termed a diffusion factor for carbon monoxide (DF(CO)). The DF(CO) procedure involves a 9-s lung inflation with tracer gases in an anesthetized mouse, followed by a 1-min gas analysis time. We have tested the approach with two common models of lung pathology, elastase-induced emphysema and bleomycin-induced fibrosis. Results show a significant 15% reduction in DF(CO) in emphysema, and a 41% reduction in the fibrosis model. Repeat measurements within a mouse were found to be highly reproducible. This pulmonary function test can thus be used to detect structural changes with these pathological models. The method can also be used to measure changes in pulmonary blood volume, since the uptake of CO is highly dependent on this variable in addition to the gas exchange surface area.

En la &uacute;ltima d&eacute;cada el rat&oacute;n se ha convertido en el principal modelo animal de una variedad de enfermedades pulmonares. Evaluar diversos mecanismos subyacentes a estas patolog&iacute;as, es esencial para realizar mediciones funcionales que pueden reflejar la patolog&iacute;a en desarrollo. En este sentido, la capacidad de difusi&oacute;n de mon&oacute;xido de carbono es una variable que refleja directamente los cambios estructurales en el pulm&oacute;n. Aunque medida de aliento solo difunde capacidad del pulm&oacute;n para el mon&oacute;xido de carbono (DL(CO)) tambi&eacute;n se ha divulgado previamente en ratones por un n&uacute;mero de investigadores, una serie de problemas t&eacute;cnicos ha impedido uso sistem&aacute;tico y generalizado de esta m&eacute;trica en modelos de rat&oacute;n. En el presente informe, describimos a fin de r&aacute;pidamente y simplemente medida variable adimensional estrechamente relacionado con el DL(CO) en ratones, como un factor de difusi&oacute;n de mon&oacute;xido de carbono (DF(CO)). El procedimiento DF(CO) consiste en una inflaci&oacute;n de pulm&oacute;n 9-s con gases tracer en un rat&oacute;n anestesiado, seguido por un tiempo de an&aacute;lisis de gas de 1 min. Hemos probado el enfoque con dos modelos comunes de patolog&iacute;a pulmonar inducida por la elastasa enfisema y fibrosis inducida por bleomicina. KRAS mutant lung cancers are generally refractory to chemotherapy as well targeted agents. To date, the identification of drugs to therapeutically inhibit K-RAS have been unsuccessful, suggesting that other approaches are required. We demonstrate in both a novel transgenic mutant Kras lung cancer mouse model and in human lung tumors that the inhibition of Twist1 restores a senescence program inducing the loss of a neoplastic phenotype. The Twist1 gene encodes for a transcription factor that is essential during embryogenesis. Twist1 has been suggested to play an important role during tumor progression. However, there is no in vivo evidence that Twist1 plays a role in autochthonous tumorigenesis. Through two novel transgenic mouse models, we show that Twist1 cooperates with Kras(G12D) to markedly accelerate lung tumorigenesis by abrogating cellular senescence programs and promoting the progression from benign adenomas to adenocarcinomas. Moreover, the suppression of Twist1 to physiological levels is sufficient to cause Kras mutant lung tumors to undergo senescence and lose their neoplastic features. Finally, we analyzed more than 500 human tumors to demonstrate that TWIST1 is frequently overexpressed in primary human lung tumors. The suppression of TWIST1 in human lung cancer cells also induced cellular senescence. Hence, TWIST1 is a critical regulator of cellular senescence programs, and the suppression of TWIST1 in human tumors may be an effective example of pro-senescence therapy.

Los c&aacute;nceres de pulm&oacute;n mutante KRAS son generalmente refractarios a la quimioterapia como agentes bien dirigidos. Hasta la fecha, la identificaci&oacute;n de drogas para inhibir terap&eacute;uticamente K-RAS han sido infructuosas, sugiriendo que otros enfoques son necesarios. Demostramos en ambos un transg&eacute;nico mutante Kras pulm&oacute;n C&aacute;ncer rat&oacute;n modelo novedoso y en tumores de pulm&oacute;n humano que la inhibici&oacute;n de la Twist1 restaura un programa de senescencia inducir la p&eacute;rdida de un fenotipo neopl&aacute;sico. El gen Twist1 codifica para un factor de transcripci&oacute;n que es esencial durante la embriog&eacute;nesis. Twist1 se ha sugerido que juegan un papel importante durante la progresi&oacute;n del tumor. Por &uacute;ltimo, hemos analizado m&aacute;s de 500 tumores humanos para demostrar que TWIST1 con frecuencia se sobre expresa en tumores de pulm&oacute;n humano primario. La supresi&oacute;n de TWIST1 en c&eacute;lulas de c&aacute;ncer de pulm&oacute;n humano tambi&eacute;n induce la senescencia celular. Por lo tanto, TWIST1 es un regulador cr&iacute;tico de los programas de la senescencia celular, y la supresi&oacute;n del TWIST1 en tumores humanos puede ser un ejemplo eficaz de la terapia pro-senescence.
