Porphysome nanovesicles &#x7531;&#x535F;&#x5549;&#x53CC;&#x5C42;&#x7684;&#x751F;&#x6210;&#x4F7F;&#x7528;&#x4F5C;&#x4E3A;&#x591A;&#x5F0F;&#x8054;&#x8FD0; biophotonic &#x9020;&#x5F71;&#x5242;&#x3002;

Optically active nanomaterials promise to advance a range of biophotonic techniques through nanoscale optical effects and integration of multiple imaging and therapeutic modalities. Here, we report the development of porphysomes; nanovesicles formed from self-assembled porphyrin bilayers that generated large, tunable extinction coefficients, structure-dependent fluorescence self-quenching and unique photothermal and photoacoustic properties. Porphysomes enabled the sensitive visualization of lymphatic systems using photoacoustic tomography. Near-infrared fluorescence generation could be restored on dissociation, creating opportunities for low-background fluorescence imaging. As a result of their organic nature, porphysomes were enzymatically biodegradable and induced minimal acute toxicity in mice with intravenous doses of 1,000 mg kg(-1). In a similar manner to liposomes, the large aqueous core of porphysomes could be passively or actively loaded. Following systemic administration, porphysomes accumulated in tumours of xenograft-bearing mice and laser irradiation induced photothermal tumour ablation. The optical properties and biocompatibility of porphysomes demonstrate the multimodal potential of organic nanoparticles for biophotonic imaging and therapy.

Nanomat&#xE9;riaux optiquement actifs promets de faire avancer une s&#xE9;rie de techniques biophotoniques travers l&#x26;#39;&#xE9;chelle nanom&#xE9;trique des effets optiques et l&#x26;#39;int&#xE9;gration de l&#x26;#39;imagerie multiple et les modalit&#xE9;s th&#xE9;rapeutiques. Nous rapportons ici le d&#xE9;veloppement de porphysomes; nanov&#xE9;sicules form&#xE9; &#xE0; partir de bicouches porphyrines auto-assembl&#xE9;es qui ont g&#xE9;n&#xE9;r&#xE9; de grands coefficients d&#x26;#39;extinction accordables, la structure d&#xE9;pendant de fluorescence &#xE0; auto-extinction et unique propri&#xE9;t&#xE9;s photothermiques et photoacoustique. Porphysomes permis la visualisation sensibles des syst&#xE8;mes lymphatiques en utilisant la tomographie photoacoustique. Dans le proche infrarouge g&#xE9;n&#xE9;ration de fluorescence peut &#xEA;tre restaur&#xE9; sur la dissociation, la cr&#xE9;ation d&#x26;#39;opportunit&#xE9;s pour les bas-fond l&#x26;#39;imagerie de fluorescence. Nanov&#xE9;sicules Porphysome g&#xE9;n&#xE9;r&#xE9;s par bicouches porphyrines pour utilisation comme agents de contraste multimodales biophotoniques

Optisch aktive Nanomaterialien Versprechen, eine Reihe von Biophotonische Techniken durch nanoskalige optische Effekte und die Integration von mehreren bildgebenden und therapeutischen Modalit&auml;ten. Hier berichten wir &uuml;ber die Entwicklung des Porphysomes; Nanovesicles gebildet von monomolekularen Porphyrin-Doppelschichten, die gro&szlig;e, einstellbare Aussterben Koeffizienten, Struktur-abh&auml;ngige Fluoreszenz self-quenching und einzigartige fotothermischen und Photoakustischen Eigenschaften generiert. Porphysomes aktiviert die sensible Visualisierung des lymphatischen Systems mit photoakustische Tomografie. Nah-Infrarot-Fluoreszenz-Generation konnte auf Dissoziation, Chancen f&uuml;r niedrig-Hintergrund-Fluoreszenz-Imaging wiederhergestellt werden. Aufgrund ihrer organischen Natur waren Porphysomes enzymatisch biologisch abbaubar und minimale akute Toxizit&auml;t bei M&auml;usen mit intraven&ouml;sen Dosen von 1.000 mg kg(-1) induziert. In &auml;hnlicher Weise zu Liposomen konnte der gro&szlig;e w&auml;ssrige Kern Porphysomes passiv oder aktiv geladen werden. Nach systemischer Verabreichung Porphysomes im Tumoren des Xenograft--Lagers M&auml;use und Bestrahlung induzierte fotothermischen Tumor Laserablation gesammelt. Die optischen Eigenschaften und Biokompatibilit&auml;t von Porphysomes demonstrieren das multimodale Potenzial der organische Nanopartikel f&uuml;r Biophotonische Bildgebung und Therapie.

Nanomateriali otticamente attivi promettono di avanzare una serie di tecniche di biophotonic attraverso effetti ottici su scala nanometrica e l&#39;integrazione di molteplici modalit&agrave; di imaging e terapeutica. Qui, segnaliamo lo sviluppo di porphysomes; nanovesicles formata da bilayer porfirina auto-assemblati che ha generato i coefficienti di estinzione grandi, sintonizzabile, fluorescenza dipendente dalla struttura unica e self-quenching fototermica e propriet&agrave; fotoacustico. Porphysomes abilitata la visualizzazione sensibile dei sistemi linfatico mediante tomografia fotoacustico. Generazione di fluorescenza vicino infrarosso potrebbe essere ripristinato sulla dissociazione, creando opportunit&agrave; per l&#39;imaging di fluorescenza di fondo basso. Aqu&#xED;, se presenta el desarrollo de porphysomes; nanoves&#xED;culas formado a partir de auto-ensambladas bicapas porfirina que generaron grandes coeficientes, la extinci&#xF3;n de sintonizables, dependiente de la estructura y propiedades fotot&#xE9;rmicas fotoac&#xFA;stica fluorescencia auto-apagado y &#xFA;nico. Porphysomes permiti&#xF3; la visualizaci&#xF3;n sensible de los sistemas linf&#xE1;ticos mediante tomograf&#xED;a fotoac&#xFA;stica. In photodynamic therapy (PDT), photosensitizers are activated by light of a specific wavelength and produce cytotoxic molecules to damage diseased tissues. Most photosensitizers are hydrophobic and easily aggregate in aqueous solution. To maintain their photodynamic activity and to enhance delivery efficiency of photosensitizers, various pharmaceutical carriers and delivery systems have been investigated for photosensitizers. This review will focus on liposomal nanostructures for the delivery of photosensitizing agents.

Enzymatic regioselection for the synthesis and biodegradation of porphysome nanovesicles.

Tumor hypoxia is increasingly being recognized as a characteristic feature of solid tumors and significantly complicates many treatments based on radio-, chemo-, and phototherapies. While photodynamic therapy (PDT) is based on photosensitizer interactions with diffused oxygen, photothermal therapy (PTT) has emerged as a new phototherapy that is predicted to be independent of oxygen levels within tumors. It has been challenging to meaningfully compare these two modalities due to differences in contrast agents and irradiation parameters, and no comparative in vivo studies have been performed until now. Here, by making use of recently developed nanostructured self-quenched porphysome nanoparticles, we were able to directly compare PDT and PTT using matched light doses and matched porphyrin photosensitizer doses (with the photosensitizer being effective for either PTT or PDT based on the existence of nanostructure or not). Therefore, we demonstrated the nanostructure-driven conversion from the PDT singlet oxygen generating mechanism of porphyrin to a completely thermal mechanism, ideal for PTT enhancement. Using a novel hypoxia tumor model, we determined that nanostructured porphyrin PTT enhancers are advantageous to overcome hypoxic conditions to achieve effective ablation of solid tumors.

Prostate cancer is the most common cancer among men and the second cause of male cancer-related deaths. There are currently three critical needs in prostate cancer imaging to personalize cancer treatment: (1) accurate intraprostatic imaging for multiple foci and extra-capsular extent; (2) monitoring local and systemic treatment response and predicting recurrence; and (3) more sensitive imaging of occult prostate cancer bone metastases. Recently, our lab developed porphysomes, inherently multimodal, all-organic nanoparticles with flexible and robust radiochemistry. Herein, we validate the first in vivo application of (64)Cu-porphysomes in clinically relevant orthotopic prostate and bony metastatic cancer models. We demonstrate clear multimodal delineation of orthotopic tumors on both the macro- and the microscopic scales (using both PET and fluorescence) and sensitively detected small bony metastases (<2 mm). The unique and multifaceted properties of porphysomes offers a promising all-in-one prostate cancer imaging agent for tumor detection and treatment response/recurrence monitoring using both radionuclide- and photonic-based strategies.
