&#x79CD;&#x7CFB; Bicyclus anynana &#x8774;&#x8776;&#x7684;&#x8F6C;&#x5316;&#x3002;

Ecological and evolutionary theory has frequently been inspired by the diversity of colour patterns on the wings of butterflies. More recently, these varied patterns have also become model systems for studying the evolution of developmental mechanisms. A technique that will facilitate our understanding of butterfly colour-pattern development is germline transformation. Germline transformation permits functional tests of candidate gene products and of cis-regulatory regions, and provides a means of generating new colour-pattern mutants by insertional mutagenesis. We report the successful transformation of the African satyrid butterfly Bicyclus anynana with two different transposable element vectors, Hermes and piggyBac, each carrying EGFP coding sequences driven by the 3XP3 synthetic enhancer that drives gene expression in the eyes. Candidate lines identified by screening for EGFP in adult eyes were later confirmed by PCR amplification of a fragment of the EGFP coding sequence from genomic DNA. Flanking DNA surrounding the insertions was amplified by inverse PCR and sequenced. Transformation rates were 5% for piggyBac and 10.2% for Hermes. Ultimately, the new data generated by these techniques may permit an integrated understanding of the developmental genetics of colour-pattern formation and of the ecological and evolutionary processes in which these patterns play a role.

La th&#xE9;orie &#xE9;cologique et &#xE9;volutive a souvent &#xE9;t&#xE9; inspir&#xE9; par la diversit&#xE9; des mod&#xE8;les de couleur sur les ailes des papillons. Plus r&#xE9;cemment, ces motifs vari&#xE9;s sont &#xE9;galement devenus des syst&#xE8;mes mod&#xE8;les pour &#xE9;tudier l&#x26;#39;&#xE9;volution des m&#xE9;canismes du d&#xE9;veloppement. Une technique qui facilitera notre compr&#xE9;hension de papillon de couleur mod&#xE8;le de d&#xE9;veloppement est la transformation germinale. Transformation germinale permet des tests fonctionnels de produits de g&#xE8;nes candidats et des r&#xE9;gions cis-r&#xE9;gulatrices, et fournit un moyen de g&#xE9;n&#xE9;rer de nouvelles couleurs de mod&#xE8;le mutants par mutagen&#xE8;se insertionnelle. Nous rapportons la transformation r&#xE9;ussie de l&#x26;#39;Afrique satyrid papillon anynana Bicyclus avec deux vecteurs diff&#xE9;rents &#xE9;l&#xE9;ments transposables, Herm&#xE8;s et piggyBac, chaque EGFP portant les s&#xE9;quences codant entra&#xEE;n&#xE9;s par le 3XP3 enhancer synth&#xE9;tique qui dirige l&#x26;#39;expression du g&#xE8;ne dans les yeux. ADN flanquant entourant les insertions a &#xE9;t&#xE9; amplifi&#xE9; par PCR inverse et s&#xE9;quenc&#xE9;s. Les taux de transformation &#xE9;taient de 5% pour piggyBac et 10,2% pour Herm&#xE8;s. En fin de compte, les nouvelles donn&#xE9;es g&#xE9;n&#xE9;r&#xE9;es par ces techniques peut permettre une compr&#xE9;hension int&#xE9;gr&#xE9;e de la g&#xE9;n&#xE9;tique du d&#xE9;veloppement de la couleur-mod&#xE8;le de formation et des processus &#xE9;cologiques et &#xE9;volutifs dans lesquels ces mod&#xE8;les jouent un r&#xF4;le.

Teoria ecologica ed evolutiva frequentemente &egrave; stata ispirata dalla diversit&agrave; dei modelli di colore sulle ali delle farfalle. Pi&ugrave; recentemente, questi modelli vario sono diventati sistemi modello per studiare l&#39;evoluzione dei meccanismi di sviluppo. Una tecnica che faciliter&agrave; la comprensione dello sviluppo di colore-pattern farfalla &egrave; la trasformazione di germline. DNA che circonda gli inserimenti di accompagnamento &egrave; stato amplificato dalla PCR inversa e sequenziato. Tassi di trasformazione erano per piggyBac il 5% e 10,2% per Hermes. Until now, the implication of candidate genes in the differentiation of color patterns has relied primarily on correlational evidence, i.e., gene expression patterns in a developing wing mapping closely to the adult color pattern. The use of transgenic techniques in the Lepidoptera, including the manipulation of gene expression, will finally allow researchers to test hypotheses of gene function at various levels of the patterning hierarchy, from signaling ligands and transcription factors to pigment enzymes. Here we present an overview of transgenic techniques employed in lepidopteran systems and highlight areas where current and future research will provide exciting opportunities to deepen our understanding of the mechanisms of morphological evolution.

Los patrones de lepid&#xF3;pteros muy diversas alas construir modelos &#xFA;tiles para estudiar la evoluci&#xF3;n del desarrollo y los cambios moleculares que permiten. Hasta ahora, la implicaci&#xF3;n de los genes candidatos en la diferenciaci&#xF3;n de los patrones de color se ha basado principalmente en pruebas de correlaci&#xF3;n, es decir, los patrones de expresi&#xF3;n g&#xE9;nica de una asignaci&#xF3;n de ala en desarrollo en estrecha colaboraci&#xF3;n con el patr&#xF3;n de coloraci&#xF3;n adulta. Butterfly eyespots represent novel complex traits that display substantial diversity in number and size within and across species. Correlative gene expression studies have implicated a large suite of transcription factors, including Distal-less (Dll), Engrailed (En), and Spalt (Sal), in eyespot development in butterflies, but direct evidence testing the function of any of these proteins is still missing. Here we show that the characteristic two-eyespot pattern of wildtype Bicyclus anynana forewings is correlated with dynamic progression of Dll, En, and Sal expression in larval wings from four spots to two spots, whereas no such decline in gene expression ensues in a four-eyespot mutant. We then conduct transgenic experiments testing whether over-expression of any of these genes in a wild-type genetic background is sufficient to induce eyespot differentiation in these pre-patterned wing compartments. We also produce a Dll-RNAi transgenic line to test how Dll down-regulation affects eyespot development. Finally we test how ectopic expression of these genes during the pupal stages of development alters adults color patters. We show that over-expressing Dll in larvae is sufficient to induce the differentiation of additional eyespots and increase the size of eyespots, whereas down-regulating Dll leads to a decrease in eyespot size. Furthermore, ectopic expression of Dll in the early pupal wing led to the appearance of ectopic patches of black scales. We conclude that Dll is a positive regulator of focal differentiation and eyespot signaling and that this gene is also a possible selector gene for scale melanization in butterflies. J. Exp. Zool. Papillon ocelles repr&eacute;sentent des caract&egrave;res nouveaux complexes qui t&eacute;moignent de la diversit&eacute; importante en nombre et en taille dans et entre les esp&egrave;ces. &Eacute;tudes d&#39;expression de g&egrave;ne corr&eacute;latives ont mis en cause une grande suite de facteurs de transcription, y compris les moins Distal (Dll), Engrailed (fr) et Spalt (Sal), dans le d&eacute;veloppement du pi&eacute;tin-verse en papillons, mais il manque encore une preuve directe, tester le fonctionnement de le quelconque de ces prot&eacute;ines. Farfalla ocelli rappresentano romanzo tratti complessi che mostrano notevoli diversit&agrave; nel numero e dimensioni all&#39;interno e fra specie. Studi di espressione genica correlativo hanno implicato una grande suite di fattori di trascrizione, compreso meno distali (Dll), Engrailed (En) e Spalt (Sal), nello sviluppo di eyespot in farfalle, ma prove dirette test della funzione di una qualsiasi di queste proteine sono ancora mancante. Qui ci mostra che il modello di due-eyespot caratteristico di wildtype Bicyclus anynana mandibole &egrave; correlato con la progressione dinamica dell&#39;espressione Dll, En e Sal in Ali larvale da quattro punti a due punti, considerando che nessun tale declino nell&#39;espressione del gene deriva in un mutante quattro-eyespot. Seasonal polyphenism demonstrates an organism's ability to respond to predictable environmental variation with alternative phenotypes, each presumably better suited to its respective environment. However, the molecular mechanisms linking environmental variation to alternative phenotypes via shifts in development remain relatively unknown. Here we investigate temporal gene expression variation in the seasonally polyphenic butterfly Bicyclus anynana. This species shows drastic changes in eyespot size depending on the temperature experienced during larval development. The wet season form (larvae reared over 24°C) has large ventral wing eyespots while the dry season form (larvae reared under 19°C) has much smaller eyespots. We compared the expression of three proteins, Notch, Engrailed, and Distal-less, in the future eyespot centers of the two forms to determine if eyespot size variation is associated with heterochronic shifts in the onset of their expression. For two of these proteins, Notch and Engrailed, expression in eyespot centers occurred earlier in dry season than in wet season larvae, while Distal-less showed no temporal difference between the two forms. These results suggest that differences between dry and wet season adult wings could be due to a delay in the onset of expression of these eyespot-associated genes. Early in eyespot development, Notch and Engrailed may be functioning as repressors rather than activators of the eyespot gene network. Alternatively, temporal variation in the onset of early expressed genes between forms may have no functional consequences to eyespot size regulation and may indicate the presence of an 'hourglass' model of development in butterfly eyespots.

Temporal gene expression variation associated with eyespot size plasticity in Bicyclus anynana.

Student learning style modality preferences, in preclinical classes, were assessed using the visual-aural-read/write-kinesthetic (VARK) inventory. Preferences were assessed for 137 preclinical students, including those in nursing, physician's assistant, physical therapy, athletic training, and natural science programs using the online VARK inventory. All classes contained a majority of multimodal and a significantly high proportion of kinesthetic learners. No correlations were noted between modality preference strength and assessment performance in general biology classes; significant correlations were discovered for kinesthetic preference among the same cohort in subsequent human anatomy (negative correlation) and general physiology (positive correlation) classes. Assessment performance of nursing students in an anatomy and physiology class resulted in correlations with aural (negative correlation) and visual (positive correlation) preference strengths. Study findings are used to evaluate the efficacy of non-omnimodal delivery of content-focused science classes, before the students have developed the background knowledge or skills required to contextualize the learning.
