We describe azimuthal light scattering spectroscopy (phi/LSS), a novel technique for assessing epithelial-cell nuclear morphology. The difference between the spectra measured at azimuthal angles phi = 0 degrees and phi = 90 degrees preferentially isolates the single backscattering contribution due to large (approximately 10 microm) structures such as epithelial cell nuclei by discriminating against scattering from smaller organelles and diffusive background. We demonstrate the feasibility of using phi/LSS for cancer detection by showing that spectra from cancerous colon tissue exhibit significantly greater azimuthal asymmetry than spectra from normal colonic tissues.

Assessing epithelial cell nuclear morphology by using azimuthal light scattering spectroscopy.

This report presents a model-independent method of assessing contributions to the light scattering from individual organelles in single intact cells. We first measure the 3D index map of a living cell, and then modify the map in such a way so as to eliminate contrast due to a particular intracellular organelle. By calculating and comparing the light scattering distributions calculated from the original and modified index maps using the Rytov approximation, we extract the light scattering contribution from the particular organelle of interest. The relative contributions of the nucleus and nucleolus to the scattering of the entire cell are thus determined, and the applicability of the homogeneous spherical model to non-spherical and heterogeneous organelles in forward scattering is evaluated.

Assessing light scattering of intracellular organelles in single intact living cells.

We employ a novel interferometer to measure the angular distribution of light backscattered by a turbid medium. Through comparison of the measured data with the predictions of Mie theory, we are able to determine the size of the scatterers comprising the medium with subwavelength precision. As the technique is based on low-coherence interferometry, we are able to examine the evolution of the angular distribution of scattered light as it propagates into the medium. The effects of multiple scattering as a function of penetration depth in the medium are analyzed. We also present various considerations for extending this technique to determining structural information in biological tissues, such as the effects of a distribution of particle sizes and the need to average out speckle contributions.

Determination of particle size by using the angular distribution of backscattered light as measured with low-coherence interferometry.

Light scattered from biological tissues can exhibit an inverse power law spectral component. We develop a model based on the Born approximation and von Karman (self-affine) spatial correlation of submicron tissue refractive index to account for this. The model is applied to light scattering spectra obtained from excised esophagi of normal and carcinogen-treated rats. Power law exponents used to fit dysplastic tissue site spectra are significantly smaller than those from normal sites, indicating that changes in tissue self-affinity can serve as a potential biomarker for precancer.

Tissue self-affinity and polarized light scattering in the born approximation: a new model for precancer detection.

We report a method of assessing the contribution of whole cell body and its nucleus to the clinically most relevant backward light scattering. We first construct an experimental system that can measure forward scattering and use the system to precisely extract the optical properties of a specimen such as the refractive index contrast, size distribution, and their density. A system that can simultaneously detect the backscattered light is installed to collect the backscattering for the same specimen. By comparing the measured backscattering spectrum with that estimated from the parameters determined by the forward scattering experiment, the contribution of cell body and nucleus to the backward light scattering is quantitatively assessed. For the HeLa cells in suspension, we found that the cell body contributes less than 10% and cell nucleus on the order of 0.1% to the total backscattering signal. Quantitative determination of the origin of backscattered light may help design a system that aims for detecting particular structure of biological tissues.

The emergence and spread of bacterial resistance to ever increasing classes of antibiotics intensifies the need for fast phenotype-based clinical tests for determining antibiotic susceptibility. Standard susceptibility testing relies on the passive observation of bacterial growth inhibition in the presence of antibiotics. In this paper, we present a novel microfluidic platform for antibiotic susceptibility testing based on stress-activation of biosynthetic pathways that are the primary targets of antibiotics. We chose Staphylococcus aureus (S. aureus) as a model system due to its clinical importance, and we selected bacterial cell wall biosynthesis as the primary target of both stress and antibiotic. Enzymatic and mechanical stresses were used to damage the bacterial cell wall, and a Î²-lactam antibiotic interfered with the repair process, resulting in rapid cell death of strains that harbor no resistance mechanism. In contrast, resistant bacteria remained viable under the assay conditions. Bacteria, covalently-bound to the bottom of the microfluidic channel, were subjected to mechanical shear stress created by flowing culture media through the microfluidic channel and to enzymatic stress with sub-inhibitory concentrations of the bactericidal agent lysostaphin. Bacterial cell death was monitored via fluorescence using the Sytox Green dead cell stain, and rates of killing were measured for the bacterial samples in the presence and absence of oxacillin. Using model susceptible (Sanger 476) and resistant (MW2) S. aureus strains, a metric was established to separate susceptible and resistant staphylococci based on normalized fluorescence values after 60 min of exposure to stress and antibiotic. Because this ground-breaking approach is not based on standard methodology, it circumvents the need for minimum inhibitory concentration (MIC) measurements and long wait times. We demonstrate the successful development of a rapid microfluidic-based and stress-activated antibiotic susceptibility test by correctly designating the phenotypes of 16 additional clinically relevant S. aureus strains in a blinded study. In addition to future clinical utility, this method has great potential for studying the effects of various stresses on bacteria and their antibiotic susceptibility.

Die Entstehung und Ausbreitung der bakteriellen Resistenz gegen zunehmende Antibiotikaklassen verst&auml;rkt die Notwendigkeit f&uuml;r schnelle Ph&auml;notyp-basierte klinische Tests zur Bestimmung der antibiotischen Anf&auml;lligkeit. Norm Resistenztestung st&uuml;tzt sich auf die passive Beobachtung von Bakterienwachstum Hemmung in Anwesenheit von Antibiotika. In diesem Beitrag pr&auml;sentieren wir eine neuartige Microfluidic-Plattform f&uuml;r antibiotische Anf&auml;lligkeit Tests basierend auf Stress-Aktivierung der biosynthetischen Bahnen, die die prim&auml;ren Ziele der Antibiotika sind. Staphylococcus Aureus (S. Aureus) w&auml;hlten wir als Modellsystem wegen seiner klinischen Bedeutung und wir bakteriellen Zellwand Biosynthese als das prim&auml;re Ziel von Stress und Antibiotikum ausgew&auml;hlt. Enzymatische und mechanische Beanspruchungen wurden verwendet, um die bakterielle Zellwand besch&auml;digen, und ein &beta;-Lactam-Antibiotikum mischte sich mit dem Reparaturprozess, was zu raschen Zelltod der St&auml;mme, die keinen Widerstand-Mechanismus zu beherbergen. La aparici&oacute;n y propagaci&oacute;n de la resistencia bacteriana a cada vez m&aacute;s clases de antibi&oacute;ticos se intensifica la necesidad de pruebas cl&iacute;nicas basadas en fenotipo r&aacute;pido para determinar la sensibilidad a los antibi&oacute;ticos. Las pruebas de sensibilidad est&aacute;ndar se basa en la observaci&oacute;n pasiva de inhibici&oacute;n del crecimiento bacteriano en presencia de antibi&oacute;ticos. En este trabajo presentamos una plataforma novedosa microflu&iacute;dicos para las pruebas de susceptibilidad antibi&oacute;tica basada en estr&eacute;s-activaci&oacute;n de v&iacute;as biosint&eacute;ticas que son los objetivos principales de los antibi&oacute;ticos. Elegimos Staphylococcus aureus (S. aureus) como un modelo de sistema debido a su importancia cl&iacute;nica, y seleccionamos la bios&iacute;ntesis de la pared celular bacteriana como el objetivo principal de tanto estr&eacute;s y antibi&oacute;tico. Enzim&aacute;tica y mec&aacute;nica fueron utilizadas para da&ntilde;ar la pared celular bacteriana, y un antibi&oacute;tico &beta;-lactam interfiri&oacute; con el proceso de reparaci&oacute;n, dando por resultado muerte celular r&aacute;pida de cepas que no albergan ning&uacute;n mecanismo de resistencia. En cambio, las bacterias resistentes a segu&iacute;a siendo viables en las condiciones de ensayo. Bacterias, enlazado covalentemente a la parte inferior del canal microflu&iacute;dicos, fueron sometidas a tensiones mec&aacute;nicas de shear creado por medios de cultivo que fluye a trav&eacute;s del canal de microfluidos y al estr&eacute;s enzim&aacute;tico con concentraciones subinhibitorias de la lysostaphin de agente bactericida. Muerte de la c&eacute;lula bacteriana se control&oacute; mediante fluorescencia mediante la tinci&oacute;n de c&eacute;lulas muertas de Sytox Green, y las tasas de homicidio se midieron las muestras bacterianas en presencia y ausencia de oxacilina. Utilizando el modelo susceptible (Sanger 476) y resistencias (MW2) S. aureus, se estableci&oacute; una m&eacute;trica separar Estafilococos resistentes y susceptibles, basados en normalizados valores de fluorescencia despu&eacute;s de 60 minutos de exposici&oacute;n a estr&eacute;s y antibi&oacute;tico. Porque este enfoque innovador no se basa en la metodolog&iacute;a est&aacute;ndar, evita la necesidad de medidas de concentraci&oacute;n m&iacute;nima inhibitoria (CMI) y tiempos de espera larga. Demostramos el desarrollo exitoso de una prueba r&aacute;pida basada en microfluidos y activada por estr&eacute;s de susceptibilidad antibi&oacute;tica designando correctamente los fenotipos de las 16 adicionales cl&iacute;nicamente relevantes S. aureus cepas en un estudio ciego. Adem&aacute;s de utilidad cl&iacute;nica futura, este m&eacute;todo tiene un gran potencial para el estudio de los efectos de diversas tensiones en bacterias y su sensibilidad a los antibi&oacute;ticos.
