The Hinsberg test is a method to identify and distinguish between primary, secondary, and tertiary amines. Amines are treated with benzenesulfonyl chloride, also known as the Hinsberg reagent, in the presence of aqueous base. Based on the amine type, different observable changes occur, allowing chemists to classify amines by their reactivity and solubility patterns in the resulting products.

Primary amines react with benzenesulfonyl chloride to form N-substituted sulfonamides. The electron-withdrawing sulfonyl group makes the N–H proton acidic enough to react with base, producing a water-soluble salt. This acidic proton enables deprotonation in base, causing the precipitate to dissolve into a clear solution.

Secondary amines react with benzenesulfonyl chloride to form N,N-disubstituted sulfonamides. Unlike primary amine products, these compounds lack an N–H proton, so they cannot be deprotonated by base. The resulting N,N-disubstituted sulfonamide remains insoluble in the added base, distinguishing secondary amines from primary amines.

Tertiary amines do not react with benzenesulfonyl chloride because they lack N–H protons. No precipitate forms when tertiary amines are exposed to the Hinsberg reagent. Upon acidification, tertiary amines are protonated and become water-soluble, but this occurs independently of the reagent reaction.

Benzenesulfonyl chloride is named the Hinsberg reagent after Oscar Hinsberg, who pioneered this classification method. The reagent selectively reacts with primary and secondary amines but not tertiary amines, making it an effective tool for distinguishing amine types based on their structural differences and reactivity.

After the Hinsberg test, the N-substituted sulfonamide salt dissolves in aqueous base to form a clear solution. Adding acid to this solution precipitates the N-substituted sulfonamide, allowing recovery of the product. This acid-base extraction process demonstrates the reversible nature of the sulfonamide formation and deprotonation.

The electron-withdrawing sulfonyl group attached to the nitrogen makes the N–H proton significantly more acidic than in typical amines. This increased acidity allows the N–H proton to be readily deprotonated by aqueous base, enabling the formation of water-soluble salts that distinguish primary amines from secondary and tertiary amines in the test.
