Leaf Powder Preparation

Leaf powder preparation is the process of converting fresh or dried leaves into a uniform, stable material for biochemical analysis and laboratory use. It typically involves removing excess moisture, drying the tissue under conditions that limit degradation, and grinding it into a fine powder, which increases surface area and promotes consistent sampling and extraction. The resulting powder can support the recovery and measurement of proteins, pigments, enzymes, metabolites, and other cellular constituents. Careful handling helps preserve biochemical integrity, improve reproducibility, and enable comparisons among plant samples in research involving nutrition, physiology, natural products, and environmental responses.

Source: Tamara M. Powers, Department of Chemistry, Texas A&M University X-ray crystallography is a technique that uses X-rays to study the structure of molecules. X-ray diffraction (XRD) experiments are routinely carried out with either single-crystal or powdered samples. Single-crystal XRD: Single-crystal XRD allows for absolute structure determination. With single-crystal XRD data, the exact atomic positions can be observed, and thus bond lengths and angles can be determined. This...

Source: Kerry M. Dooley and Michael G. Benton, Department of Chemical Engineering, Louisiana State University, Baton Rouge, LA Surface area and pore size distribution are attributes used by adsorbent and catalyst manufacturers and users to ensure quality control and to determine when products are at the end of their useful lives. The surface area of a porous solid is directly related to its adsorption capacity or catalytic activity. The pore size distribution of an adsorbent or catalyst is...

Source: Gulla, S., et al. Multi-locus Variable-number Tandem-repeat Analysis of the Fish-pathogenic Bacterium Yersinia ruckeri by Multiplex PCR and Capillary Electrophoresis. J. Vis. Exp. (2019).This protocol extracts genomic DNA from Yersinia ruckeri by suspending isolated colonies in ultrapure water and applying heat to lyse the cells. Centrifugation separates the DNA-containing supernatant from cell debris, yielding template DNA suitable for PCR-based genotyping and epidemiological analysis.
