Mitochondrial Protein Quantification

Mitochondrial protein quantification is the measurement of protein abundance in mitochondria or mitochondrial fractions, a key approach for assessing organelle composition and function in biochemistry. It typically involves isolating mitochondria, extracting proteins, and determining their levels with methods such as immunoblotting, enzyme-linked assays, or mass spectrometry, using total protein or validated mitochondrial markers for normalization. These measurements help characterize respiratory-chain complexes, metabolic enzymes, and disease-associated changes in mitochondrial structure and activity. Quantitative protein profiles support studies of energy metabolism, apoptosis, aging, and inherited mitochondrial disorders, while improving evaluation of cellular responses to drugs, mutations, and environmental stress.
