Nmr Fragment Screening

NMR fragment screening is a biophysical method that identifies small chemical fragments capable of binding to a target protein, providing an early route to potential drug leads. In solution, nuclear magnetic resonance detects binding through changes such as chemical-shift perturbations, signal broadening, or altered ligand relaxation, while experiments including protein-observed and ligand-observed NMR distinguish specific interactions from nonbinding compounds. Because NMR can reveal weak binding and provide information about binding sites and interaction modes, it complements biochemical assays in drug discovery. The resulting fragment hits can guide structure-based design, fragment linking, and chemical optimization toward higher-affinity, more selective molecules.
