Metabolic Activity Assay

A metabolic activity assay measures biochemical activity in living cells by tracking the conversion of substrates into a detectable signal. In these assays, enzymes in metabolically active cells reduce or oxidize indicator reagents, generating a colorimetric, fluorescent, or luminescent output that reflects activity under defined conditions. In bioengineering, metabolic activity assays help evaluate cell viability, proliferation, and responses to biomaterials, engineered tissues, drugs, or environmental conditions. Because metabolic activity is an indirect readout, standardized controls and calibration are important for distinguishing changes in cell number from altered cellular function and for supporting reproducible material and tissue characterization.

Sebolai11Department of Microbiology and Biochemistry, University of the Free State. The video demonstrates a technique for evaluating the influence of photodynamic treatment (PDT) with a photosensitizer on the metabolic activity of murine macrophages. The cells undergo treatment with the photosensitizer followed by exposure to UV light, initiating PDT. This process triggers the photosensitizer to generate reactive oxygen species...

2023

This video describes the colorimetric assay, methyl thiazole tetrazolium (MTT) assay, to measure drug-induced cytotoxicity in cancer cells and determine their metabolic activity. This assay is based on the metabolic conversion of tetrazolium salts into colored products, reflecting the mitochondrial activity of cells.

Isothermal Microcalorimetry to Monitor Metabolic Activity in Bacteria

Take serial dilutions of a bacterial culture and add aliquots into calorimetric vials containing growth medium.Load the sealed vials into the multi-well plate of an isothermal microcalorimeter to measure metabolic heat production. The device is held at a constant temperature to ensure the detected heat originates solely from bacterial metabolism. As bacteria metabolize nutrients and proliferate, they release heat that sensors detect in real time.In parallel, incubate identical vials in an...
