Pleckstrin Homology Domain

A pleckstrin homology (PH) domain is a compact protein module that helps target signaling proteins to specific cellular membranes, making it important for understanding spatial control of cell signaling. Many PH domains recognize phosphoinositides, charged lipid molecules enriched on particular membrane surfaces, while some also interact with proteins or soluble ligands. These interactions recruit enzymes, adaptors, and cytoskeletal regulators to defined locations, where they influence pathways controlling membrane trafficking, cell growth, and cytoskeletal organization. In biological techniques, PH domains are studied using lipid-binding assays, fluorescence-based localization, mutational analysis, and structural methods to map membrane interactions and investigate signaling dysfunction.

Homologous recombination techniques greatly advance Drosophila genetics by enabling the creation of molecularly precise mutations. The recent adoption of recombineering allows one to manipulate large pieces of DNA and transform them into Drosophila6.
