Amphibian Model

An amphibian model is a living amphibian, or an amphibian-derived system, used to investigate biological processes under controlled laboratory or field conditions. These models are valuable because many species produce externally developing embryos, undergo hormone-regulated metamorphosis, and in some cases regenerate damaged tissues, allowing researchers to connect cellular mechanisms with visible changes in form and function. Amphibian models support studies of embryonic development, gene function, endocrinology, regeneration, environmental toxicology, and disease biology. Their combination of experimental accessibility and vertebrate physiology makes them useful for testing hypotheses that can inform comparative biology, conservation, and biomedical research.

Simultaneous Whole-cell Recordings from Photoreceptors and Second-order Neurons in an Amphibian Retinal Slice Preparation

We describe the preparation of thin retinal slices from aquatic tiger salamanders (Ambystoma tigrinum) and explain how we use these slices to study synaptic processing in the retina by obtaining dual whole-cell voltage clamp recordings from photoreceptors and second-order horizontal and bipolar cells.

Reduced Itraconazole Concentration and Durations Are Successful in Treating Batrachochytrium dendrobatidis Infection in Amphibians

Batrachochytrium dendrobatidis infection in amphibians can be eliminated using a reduced concentration (0.0025%) and shorter duration (five minute baths for six days) of itraconazole than is generally used. Less mortality and fewer negative side effects were observed using this modified treatment protocol.

Isolation and Identification of Bacterial Strains from Skin of Terrestrial Amphibians

This is a method for sampling and isolating culturable bacteria from the cutaneous microbiota of European plethodontid salamanders (Speleomantes genus). Here, we present a protocol for sampling salamander skin with swabs and processing them using culturable approaches. We describe sampling, isolation, and establishment of axenic culture with bacterial strain characterization.

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.
