Egg Chamber Isolation

Egg chamber isolation is a biological technique that separates developing egg chambers from an ovary for direct study of oogenesis, cell differentiation, and tissue organization. In Drosophila, researchers dissect ovaries in an appropriate physiological medium and gently tease apart the ovariole structures, releasing egg chambers while preserving their germline and surrounding follicle cells. The isolated chambers can then be examined by fluorescence microscopy, immunostaining, live imaging, or molecular assays to analyze developmental stages, cell-cell signaling, cytoskeletal changes, and gene expression. This method provides a tractable system for investigating reproductive biology and the cellular mechanisms that control tissue development.

Upright Imaging of Drosophila Egg Chambers

The upright imaging method described in this protocol allows for the detailed visualization of the poles of a developing Drosophila melanogaster egg. This end-on view provides a new perspective into the arrangements and morphologies of multiple cell types in the follicular epithelium.
