Endospore Isolation

Endospore isolation is a microbiological method used to separate dormant, highly resistant bacterial endospores from vegetative cells for identification and study. The process commonly uses heat treatment or another selective condition that destroys less-resistant vegetative cells while preserving endospores; subsequent plating on nutrient media allows surviving spores to germinate and form colonies. In biology, this approach helps detect and characterize spore-forming bacteria in environmental, clinical, and food samples. It also supports research on sporulation, germination, resistance to environmental stress, and the persistence of bacterial populations under unfavorable conditions.

Source: Wunderlin, T., et al. Physical Isolation of Endospores from Environmental Samples by Targeted Lysis of Vegetative Cells. J. Vis. Exp. (2016)This video demonstrates a method to isolate endospores from an environmental sample containing both vegetative cells and various bacteria. The procedure involves filtration, mechanical disruption, and selective chemical treatment to weaken and lyse vegetative cells while preserving endospores. The final step isolates the resilient endospores through...

DiRita1 1 Department of Microbiology and Molecular Genetics, Michigan State University, East Lansing, Michigan, United States of America Bacteria are diverse microorganisms found nearly everywhere on Earth. Many properties help distinguish them from each other, including but not limited to Gram-staining type, shape and arrangement, production of capsule, and formation of spores. To observe these properties, one...

Endospores are specialized, dormant cells primarily formed by Gram-positive bacteria, including Bacillus and Clostridium, enabling survival under extreme environmental conditions. Due to their unique composition and formation process, these structures are highly resistant to physical and chemical insults, such as extreme heat, ultraviolet and ionizing radiation, desiccation, and toxic chemicals. Rare instances of endospore-like structures have also been observed in some Gram-negative bacteria,...
