Enzyme Linked Immunomagnetic Assay

Enzyme Linked Immunomagnetic Assay is an immunoassay that combines antibody-based recognition with magnetic separation and enzyme-generated detection, enabling selective measurement of target molecules in complex biological samples. In this method, antibodies attached to magnetic beads capture the target, a magnet collects the bead-bound complexes, and a labeled detection antibody or enzyme reaction produces a measurable color or signal. The approach can reduce sample interference by washing away unbound substances while concentrating the analyte for analysis. In biology, it supports sensitive detection of proteins, pathogens, cells, and other biomarkers, with applications in research, diagnostics, environmental monitoring, and biotechnology.

In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane. There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen. Enzyme-substrate reaction allows the antigen to be visualized or quantified.

We describe the enzyme-linked lectin assay (ELLA) for measuring influenza neuraminidase (NA)-inhibition antibody titers in sera. The assay uses peanut agglutinin to quantify galactose residues that become accessible when NA removes sialic acid from fetuin-coated, 96-well plates.
