Exosome Isolation

Exosome isolation is the process of separating exosomes, small extracellular vesicles released by cells, from biological fluids or cell culture media for characterization and downstream analysis. It matters because exosomes carry proteins, lipids, and nucleic acids that can reflect cellular state and mediate intercellular communication. Common workflows exploit differences in vesicle size, density, or surface markers through differential ultracentrifugation, size-exclusion chromatography, polymer-based precipitation, or affinity capture, often followed by washing and concentration. In biology, purified exosomes support studies of cell signaling, disease biomarkers, and potential therapeutic delivery, while isolation quality strongly influences reproducibility and interpretation of results.

These methods are adaptable for exosome isolation from both cell culture media and biological fluids, and can beyond analysis of molecular content also be useful in functional studies.
