Pbmc Proliferation Assay

A PBMC proliferation assay is a laboratory method that measures how peripheral blood mononuclear cells, including lymphocytes and monocytes, respond to stimulation by dividing and increasing in number. In a typical assay, isolated PBMCs are exposed to a mitogen, antigen, or other test condition, and proliferation is quantified through DNA synthesis, cell-tracking dye dilution, or metabolic readouts. Researchers use these assays to evaluate immune-cell activation, antigen-specific responses, immunosuppressive effects, and potential drug toxicity. The results help characterize cellular immunity in studies of infection, vaccination, autoimmune disease, cancer, and therapeutic development.

An In Vitro Assay for Evaluating the Immunomodulatory Impact of Monocytes on Leukocyte Proliferation

2025

This video demonstrates an in vitro assay to explore the immunomodulatory influence of mesenchymal stem cells (MSCs) on effector T cell proliferation. The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

A Technique to Generate a Human PBMC-Engrafted Humanized Xenograft Mouse Model

2025

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

Choroid Sprouting Assay to Study Ocular Microvascular Proliferation Ex Vivo

2025

This video demonstrates the development of an ex vivo model of choroidal sprouting to study ocular microvascular angiogenesis. The assay involves culturing mouse eye explants containing the choroid tissue attached to the sclera and retinal pigment epithelium under specific conditions that promote choroidal microvascular sprouting.

Bromodeoxyuridine Pulse Labelling Assay: A Technique to Measure Cell Proliferation

2023

The video describes the stepwise pulse labeling assay of mammalian cells with bromodeoxyuridine (BrdU) for measuring cell proliferation. The BrdU uptake permits the temporal tracking of cells that were in the synthesis phase at a specific point in time, without requiring the cells to be synchronized.

A Vaccine-Pulsed Dendritic Cell-Based In Vitro Assay to Induce Lymphocyte Proliferation

2025

This video demonstrates a monocyte-derived dendritic cell, MoDC-based assay to measure the immunogenicity of commercially available vaccines. MoDC assays investigate dendritic cell responses to antigens, pathogens, or treatments and can provide insights into immune responses, vaccine development, and immunotherapy research.
