Trap-rc Protocol

The TRAP-rc protocol is a molecular biology method for profiling messenger RNAs being translated in selected cell types, providing a cell-specific view of gene expression. It works by expressing an affinity-tagged ribosomal protein in target cells, isolating tagged ribosomes from tissue lysates with antibody-coated beads, and recovering the ribosome-associated mRNAs for analysis. This approach can reveal cell-type-specific translational programs without requiring physical separation of the cells. In biology, TRAP-rc supports studies of development, neural circuits, tissue organization, and disease by linking gene expression to active protein synthesis and identifying transcripts that may be missed by whole-tissue RNA analysis.
