Wing Clip Biopsy

Wing clip biopsy is a biological sampling technique that removes a small section of wing tissue for genetic or molecular analysis while preserving the animal for continued observation or study. The collected tissue is typically lysed to release DNA, which can then be purified and analyzed using methods such as polymerase chain reaction, sequencing, or genotyping. In biology, this approach supports species identification, sex determination, strain verification, and tracking of inherited traits in organisms with accessible wings, including laboratory insects. By linking a physical marker to a DNA-based result, wing clip biopsy helps researchers monitor individuals and populations with limited disruption.

In order to examine gene expression in the pupal wing tissue of Bicyclus anynana, we present an optimized protocol for in situ hybridizations using riboprobes. We also provide guidelines for the further optimization of this protocol for use in pupal wings of other Lepidopteran species.
