De Novo Lipid Synthesis

De novo lipid synthesis is the biochemical production of fatty acids and other lipids from small carbon precursors, rather than their direct uptake from the environment. In the central pathway, acetyl-CoA is converted to malonyl-CoA, and fatty acid synthase repeatedly condenses and reduces these units using NADPH to form a saturated fatty acid; subsequent elongation, desaturation, and esterification generate diverse lipid molecules. This process supplies membranes, energy stores, and signaling compounds, while its regulation provides important insight into metabolism. In chemistry and biomedical research, studying de novo lipid synthesis supports investigations of metabolic disease, cancer biology, nutrition, and lipid-based therapeutic strategies.

Assessment of de novo Protein Synthesis Rates in Caenorhabditis elegans

Here, we introduce and describe a nonradioactive and noninvasive method to assess de novo protein synthesis in vivo, utilizing the nematode Caenorhabditis elegans and fluorescence recovery after photobleaching (FRAP). This method can be combined with genetic and/or pharmacological screens to identify novel modulators of protein synthesis.
