Fluorescent Product Formation

Fluorescent product formation is a chemical process in which a reaction generates a product that emits light after absorbing radiation, making molecular transformation easier to detect and measure. The reaction creates or reveals a fluorophore, a molecular structure whose electronic configuration permits excitation to a higher energy state followed by light emission as it returns to a lower state. In chemistry, this principle supports reaction monitoring, analytical assays, and detection of otherwise difficult-toobserve compounds. Measuring fluorescence intensity or emission spectra can provide information about product formation, reaction progress, and chemical concentration.

Intravital Fluorescence Microscopy to Study Microvascular Thrombus Formation

This video demonstrates the intravital fluorescence microscopy technique to study thrombus formation in a mouse model. The mouse is injected with a fluorophore-labeled polysaccharide to monitor blood circulation. Upon illumination with a high-intensity light, the phototoxic induction of an injury in the microvasculature of the ear leads to the formation of a thrombus, which is confirmed by visualizing the obstruction in the blood flow.

We describe an experimental setup to visualize with unprecedented high resolution phagosome formation and closure in three dimensions in living macrophages, using total internal reflection fluorescence microscopy. It allows monitoring of the base of the phagocytic cup, the extending pseudopods, as well as the precise site of phagosome scission.
