T Cell Activation Assay

T cell activation assays are laboratory methods used to measure how T lymphocytes respond to antigenic or stimulatory signals, making them valuable for immunology and environmental health research. Activation typically requires T cell receptor recognition of peptide–MHC complexes together with co-stimulatory signals, after which cells produce cytokines, proliferate, or alter surface-marker expression. Researchers quantify these responses using methods such as flow cytometry, cytokine immunoassays, or proliferation measurements. In environmental studies, the assays can help evaluate how pollutants, chemicals, allergens, or other exposures influence immune function and can support investigations of immune dysregulation, toxicity, and host responses to environmental challenges.

This article describes the protocol for XTT assay which is a colorimetric assay to assess cell viability and mitochondrial activity.

Luciferase Assay for Measuring γ-Secretase Activity in Human Cells

2025

This video demonstrates the procedure of measuring γ-secretase activity in genetically modified human cells using a tetracycline-inducible system. The process involves inducing APP gene expression with tetracycline, generating amyloid-β peptides, and activating luciferase production. The bioluminescence resulting from the luciferase-substrate reaction is then measured, reflecting γ-secretase activity.

This protocol describes a sensitive, cell-based cytotoxicity assay. By enumerating the decrease in frequency of live target CD4+ T cells in the presence of an increasing number of effector CD8+ T cells, this assay allows for the direct assessment of cytolytic activity of antigen-specific CD8+ T cells.

Monitoring B Cell Activation with Antigenic Liposomes using a Calcium-Flux Assay

2025

This video showcases the assessment of B cell activation through the utilization of antigenic liposomes. B cells treated with calcium-sensitive Indo-1 dye are distinguished using flow cytometry, and their violet-to-blue fluorescence ratio is analyzed. As the cells encounter antigenic liposomes, it triggers B cell activation through calcium influx, progressively enhancing the violet-to-blue fluorescence signal ratio over time.
