Samtools Consensus Generation

Samtools consensus generation is a bioinformatics method for constructing a representative DNA sequence from aligned sequencing reads, helping researchers summarize genetic variation in a sample or reference region. Using BAM or CRAM alignments, samtools examines the bases and quality information covering each genomic position, weighs the read evidence, and selects a consensus base, while configurable thresholds can mark uncertain sites, insertions, deletions, or uncovered positions. In genetics, the resulting sequence supports variant interpretation, genome reconstruction, comparative analysis, and downstream annotation. Because the consensus depends on read depth, alignment quality, and filtering choices, it also provides a compact view of data confidence.

A common protein extraction protocol using urea/thiourea/SDS buffer for human and mice brain tissue allows indentification of proteins by 2D-DIGE and their subsequent characterization by mini 2DE immunoblotting. This method enables one to obtain more reproducible and reliable results from human biopsies and experimental models.
