293tn Producer Cells

293TN producer cells are human embryonic kidney-derived cells optimized for generating recombinant viral particles, making them a practical tool for delivering genetic material in immunology and infection research. During transient transfection, researchers introduce plasmids encoding the desired transfer genome alongside viral packaging and envelope components; the cells express these elements, assemble viral particles, and release them into the culture medium. The resulting particles can introduce genes into target cells for studying immune signaling, host-pathogen interactions, gene function, and cellular responses. Because production occurs in a controlled laboratory system, 293TN cells support reproducible vector preparation for experiments involving gene delivery and functional analysis.

Here, we use retroviral transduction and concatemeric transfection to create a cell line that can express the components of a lentiviral vector (LV) in the absence of tetracycline. This LV encodes GFP and is pseudotyped with a glycoprotein, SVGmu, which is specific for a receptor on dendritic cells.
