In Vitro Phagocytosis

In vitro phagocytosis is the laboratory study of how immune cells, such as macrophages and neutrophils, recognize, engulf, and process particles or microorganisms under controlled conditions. During this process, receptors on the phagocyte bind targets, triggering membrane extension and formation of a phagosome, which can mature and fuse with lysosomes for degradation. Researchers measure uptake using microscopy, fluorescence-based assays, or flow cytometry to assess phagocyte function, pathogen clearance, and responses to immune-modulating treatments. These models support immunology and infection research by allowing controlled comparisons of host defenses, microbial evasion strategies, and potential therapeutic interventions.

This work describes a standardized in vitro protocol to quantify and compare the phagocytosis rates of xenogeneic (human) and allogeneic (rat) red blood cells by isolated rat macrophages.

An In Vitro Assay to Measure Antibody-Dependent Cellular Phagocytosis by Phagocytes

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...

Modeling Complement-Mediated Phagocytosis Using Opsonized Red Blood Cells In Vitro

Source: Nagre, N., et al. Alveolar Macrophage Phagocytosis and Bacteria Clearance in Mice. J. Vis. Exp. (2019).This video demonstrates the assessment of complement receptor-mediated phagocytosis using sheep red blood cells opsonized with IgM and complement, followed by their uptake by mouse macrophages. The extent of phagocytosis is quantified by measuring a blue chromogenic product formed through hemoglobin-catalyzed oxidation of diaminofluorene.
