Membrane Vesicle Release

Membrane vesicle release is the cellular process by which lipid-bilayer vesicles carrying proteins, lipids, and nucleic acids leave a cell, enabling communication and influencing tissue responses. Vesicles form through inward budding of endosomal membranes followed by fusion with the plasma membrane, or through outward budding and scission of the cell surface; release is regulated by trafficking machinery and cellular stress. In immunology and infection, these vesicles can transfer antigens, inflammatory signals, and host or pathogen molecules between cells, shaping immune activation, suppression, and pathogen spread. Their molecular cargo also supports biomarker development and may inform vaccines or vesicle-based therapeutic strategies.

Generating a Multivalent-Displaying Outer Membrane Vesicle Vaccine

2025

This video demonstrates a method to generate multivalent-displaying outer membrane vesicle (OMV) vaccines. Mammalian cells are transfected to express and secrete viral antigens fused to a SpyTag and a polyhistidine tag. These antigens are then purified using a nickel-nitrilotriacetic acid (Ni-NTA) affinity column. The purified antigen is mixed with outer membrane vesicles expressing surface-bound SpyCatcher (SC) protein. Multiple antigens covalently attach to SpyCatchers via their SpyTags (ST),...

Source: Jung, A. L., et al. Legionella pneumophila Outer Membrane Vesicles: Isolation and Analysis of Their Pro-inflammatory Potential on Macrophages. J. Vis. Exp. (2017)This video demonstrates the isolation of outer membrane vesicles (OMVs) from Legionella pneumophila using sequential centrifugation, sterile filtration, and ultracentrifugation. The purified OMVs, enriched in membrane proteins and lipopolysaccharides, are suitable for host-pathogen interaction studies.

(2021)This video demonstrates the isolation and concentration of extracellular vesicles (EVs) derived from the outer membrane of genetically engineered bacteria. The process involves culturing the bacteria to release EVs, followed by sequential centrifugation and filtration steps to remove cells and debris. Finally, ultrafiltration is used to...
