Drug Treatment Response Assay

A drug treatment response assay is a laboratory method that measures how cells, tissues, or patient-derived samples react to a therapeutic compound, helping characterize treatment effectiveness and resistance. In a typical assay, samples receive defined drug concentrations for a specified period, after which researchers quantify changes in cell viability, proliferation, apoptosis, or molecular biomarkers to generate a dose-response profile. In medicine, these assays support drug development, toxicity assessment, and investigations of why treatments succeed or fail. They can also inform precision medicine by comparing responses across samples and identifying therapies most likely to benefit particular patients.

This protocol describes an assay for measuring CFTR function and CFTR modulator responses in cultured tissue from subjects with cystic fibrosis (CF). Biopsy-derived intestinal organoids swell in a cAMP-driven fashion, a response that is defective (or strongly reduced) in CF organoids and can be restored by exposure to CFTR modulators.
