Inter-operator Analysis

Inter-operator analysis is the systematic evaluation of how consistently different operators, such as clinicians, technicians, or image assessors, perform a measurement, procedure, or interpretation. It compares results produced by multiple operators under similar conditions, using measures such as inter-rater agreement, correlation, intraclass correlation, or observer variability to distinguish operator effects from true differences among patients or samples. In medicine, this analysis helps validate diagnostic assessments, imaging interpretations, laboratory procedures, and clinical devices by identifying sources of inconsistency. Reliable inter-operator performance strengthens reproducibility, supports standardized training and protocols, and improves confidence in research findings and patient-care decisions.

The protocol described here aims to measure the hydrodynamic diameter of spherical nanoparticles, more specifically gold nanoparticles, in aqueous media by means of Nanoparticle Tracking Analysis (NTA). The latter involves tracking the movement of particles due to Brownian motion and implementing the Stokes-Einstein equation to obtain the hydrodynamic diameter.
