Orf1 Orf2 Junction

The ORF1-ORF2 junction is the genomic boundary between regions encoding nonstructural replication proteins and structural proteins in several positive-sense RNA viruses, including noroviruses. During replication, viral RNA synthesis produces a subgenomic RNA that begins near this junction, allowing ORF2 to be translated separately from the genomic RNA and enabling coordinated production of replication and capsid proteins. In medicine and virology, this region is an important target for reverse-transcription PCR, sequencing, and strain classification. Analysis of junction sequences supports norovirus genotyping, detection of recombination, outbreak investigation, and surveillance of viral evolution.
